‘Therapeutic’ concentrations of digitoxigenin (10−7−3× 10−7 g/ml), producing positive inotropic effects, did not alter resting and action potentials of electrically driven guinea-pig auricles. However, ‘toxic’ concentrations of digitoxigenin (5 × 10−7−10−8 g/ml), producing arrhythmias and contracture of the myocardium, had a marked influence upon resting and action potentials and conduction velocity.

1. The action of iodoacetic acid (IA) was studied on the isolated rat diaphragm at 20–22° C. Membrane and action potentials were led off with intracellular microelectrodes. 2. IA causes a contracture which is complete in 4 hours. As long as t he contracture develops, the membrane potential remains constant. A contracture identical with that seen in a normal preparation can be produced by IA in the presence of isotonic K2SO4. 3. The effect of dibutyryl cyclic AMP (DB-c-AMP; 3 X 10(-4)-3 X 10(-3) M) on electrically induced twitch and high potassium (142.4 mM KCl)-induced contracture tension was studied in papillary muscles from normal and reserpinized cats ([Ca]0 1.8 mM; 25 degrees C; pH 7.4). In both groups of preparations, the increase in twitch tension evoked by DB-c-AMP was accompanied by an abbreviation of the time to peak force and of relaxation time.
