Monitoring of transglutaminase crosslinking reaction by 1H NMR spectroscopy on model substrates

International audience; A new method based on 1H NMR spectroscopy was developed for monitoring transglutaminase crosslinking reaction with model molecules (CBZ-Gln-Gly and N-α-acetyl-lysine). The transglutaminase reaction led to the appearance of new resonances on NMR spectrum as well as significant decrease in others. The new observed resonances, originated from newly formed ɛ-(γ-glutamyl)lysine isopeptide bonds, evidence the enzymatic reaction and allow to quantify the ɛ-(γ-glutamyl)lysine fragment. Moreover, the decrease in resonance intensity, originated from lysine, permit to determine the crosslinking degree. These results obtained by 1H NMR spectroscopy can be used as an alternative …

Tailoring the physicochemical properties of core-crosslinked polymeric micelles for pharmaceutical applications.

To optimally exploit the potential of (tumor-) targeted nanomedicines, platform technologies are needed in which physicochemical and pharmaceutical properties can be tailored according to specific medical needs and applications. We here systematically customized the properties of core-crosslinked polymeric micelles (CCPM). Polymeric micelles (PM) are extensively used to improve the delivery of hydrophobic drugs. Many different PM have been designed and evaluated over the years, and some of them have steadily progressed through clinical trials. Increasing evidence suggests, however, that for prolonged circulation times and for efficient EPR-mediated drug targeting to tumors and to sites of inflammation, PM need to be stabilized, to prevent premature disintegration. Biodegradable hydrogels obtained by photocrosslinking of dextran and polyaspartamide derivatives

The functionalization of dextran with glycidyl methacrylate (GMA) leads to the formation of a derivative that generates hydrogels for irradiation at 365nm. The effects of various polymer concentrations and irradiation times on the yield and the properties of the obtained hydrogels are reported. The networks have been characterized by FT-IR spectra, dimensional analysis and swelling measurements carried out at different pH values. In vitro studies suggest that all samples undergo a partial chemical hydrolysis, whereas the incubation with dextranases causes a total degradation whose rate depends on the degree of crosslinking. Construction and characterization of models of hypercrosslinked polystyrene

A simple algorithm involving classical molecular dynamics (MD) simulations is here suggested to build up models of hypercrosslinked polystyrene showing macroporous structure. The algorithm is composed by three consecutive stages: MD simulation of a single polystyrene coil, crosslink formation, and finally relaxation of the structure. Effect of Riboflavin/UVA Collagen Cross-linking on Central Cornea, Limbus and Intraocular Pressure. Experimental Study in Rabbit Eyes

The Purpose of present study was to investigate the effect of riboflavin/ultraviolet-A-induced collagen cross-linking (CXL) on central cornea, limbus and intraocular pressure (IOP). This was an animal experimental study. The right corneas of 10 rabbits were ultraviolet-A irradiated (3 mW/cm2 for 30 minutes) after de-epithelialization and instillation of 0.1% riboflavin / 20% Dextran drops. In situ crosslinkable hyaluronan hydrogels for tissue engineering

We describe the development of an injectable, cell-containing hydrogel that supports cell proliferation and growth to permit in vivo engineering of new tissues. Two thiolated hyaluronan (HA) derivatives were coupled to four alpha,beta-unsaturated ester and amide derivatives of poly(ethylene glycol) (PEG) 3400. The relative chemical reactivity with cysteine decreased in the order PEG-diacrylate (PEGDA)>>PEG-dimethacrylate>PEG-diacrylamide>PEG-dimethacrylamide. The 3-thiopropanoyl hydrazide derivative (HA-DTPH) was more reactive than the 4-thiobutanoyl hydrazide, HA-DTBH. Abstract: The modification of polymer architectures by reaction with chemically adsorbed hydroxyl radicals has been thoroughly investigated by electrolyzing dilute aqueous solutions of the biocompatible polymer poly(vinylpyrrolidone) (PVP), using an undivided electrolytic cell with a Ti/IrO2–Ta2O5(DSA®) anode. Several electrolyses were performed to assess the influence of the applied potential, the circulated charge and the PVP concentration, which was always kept low to avoid chain overlapping.
