Brain Delivery of Multifunctional Dendrimer Protein Bioconjugates

Abstract Neurological disorders are undoubtedly among the most alarming diseases humans might face. In treatment of neurological disorders, the blood‐brain barrier (BBB) is a challenging obstacle preventing drug penetration into the brain. Advances in dendrimer chemistry for central nervous system (CNS) treatments are presented here. A poly(amido)amine (PAMAM) dendrimer bioconjugate with a streptavidin adapter for the attachment of dendrons or any biotinylated drug is constructed. In vitro studies on porcine or murine models and in vivo mouse studies are performed and reveal the permeation of dendronized streptavidin (DSA) into the CNS. Tunable Superstructures of Dendronized Graphene Nanoribbons in Liquid Phase

In this Communication, we report the first synthesis of structurally well-defined graphene nanoribbons (GNRs) functionalized with dendritic polymers. The resultant GNRs possess grafting ratios of 0.59-0.68 for the dendrons of different generations. Remarkably, the precise 3D branched conformation of the grafted dendrons affords the GNRs unprecedented 1D supramolecular self-assembly behavior in tetrahydrofuran (THF), yielding nanowires, helices and nanofibers depending on the dimension of the dendrons. The GNR superstructures in THF exhibit near-infrared absorption with maxima between 650 and 700 nm, yielding an optical bandgap of 1.2-1.3 eV. Ultrafast photoconductivity analyses unveil that …

Synthesis, characterization, and thermal behavior of steroidal dendrons

A series of novel dendritic steroidal polyesters of first and second generation has been synthesized in convergent fashion by the use of 2,2-bis(hydroxymethyl)propionic acid as a repeating unit. The first- and second-generation hydroxy-functionalized dendrons with a variety of surface modifications were produced through the use of four bile acids: lithocholic acid (LCA), ursodeoxycholic acid (UDCA), deoxycholic acid (DCA), and cholic acid (CA). Optical Switching and Antenna Effect of Dendrimers with an Anthracene Core

Dendrimers 6G(i) (i=1-4) consisting of an anthracene core and Fréchet dendrons which are attached via a CH(2)OCH(2) chain in the 9-position undergo quantitative and completely reversible intramolecular [4pi+4pi] cycloaddition. The process can be monitored by absorption and fluorescence measurements. Dendritic octa-CMPO derivatives of calix[4]arenes

Abstract Calix[4]arenes substituted at the narrow or wide rim by eight carbamoylmethyl-phosphine oxide (CMPO) functions in a dendritic manner were synthesised and studied in extraction of Eu 3+ and Am 3+ from aqueous nitric acid into o -nitrophenylhexyl ether. 1 H NMR relaxivity titrations for a wide rim octa-CMPO reveal the clear formation of a solvent-free 1:2 ligand/metal complex, while the wide rim tetra-CMPO formed oligomeric complexes under similar conditions.

Multivalency Beats Complexity: A Study on the Cell Uptake of Carbohydrate Functionalized Nanocarriers to Dendritic Cells.

Herein, we report the synthesis of carbohydrate and glycodendron structures for dendritic cell targeting, which were subsequently bound to hydroxyethyl starch (HES) nanocapsules prepared by the inverse miniemulsion technique. The uptake of the carbohydrate-functionalized HES nanocapsules into immature human dendritic cells (hDCs) revealed a strong dependence on the used carbohydrate. A multivalent mannose-terminated dendron was found to be far superior in uptake compared to the structurally more complex oligosaccharides used.

Preparation and Photochemistry of Dendrimers with Isolated Stilbene Chromophores

In addition to the model compounds 4a and 4b, the dendrimers 11 and 14 with trans-stilbene chromophores in the core and on the periphery of the dendrons were prepared and their photochemistry was studied in solution and in neat films. Due to the flexibility of the arms, intramolecular and intermolecular CC bonds are formed on irradiation. Thus, the generation of nanoparticles, which represent small oligomers, is much more likely than for the cross-linking of rigid, cross-conjugated stilbenoid dendrimers. Dendrimers with peripheral stilbene chromophores

Abstract Two types of dendritic nanoparticles were prepared, which contain ( E )-stilbene chromophores in the terminal positions of the dendrons. The compounds showed a highly efficient photoreactivity in the course of which statistical CC bond formations led to a crosslinking of the particles. Finally, all stilbene chromophores reacted and the typical ( E )-stilbene absorption and fluorescence disappeared completely.
