DNA Block Copolymer Micelles – A Combinatorial Tool for Cancer Nanotechnology
2008
Selective drug targeting of a specific organ or tissue is a challenging task. This holds especially true for chemotherapeutic cancer treatment because most of the available anticancer agents cannot distinguish between cancerous and healthy cells, leading to systemic toxicity and undesirable side effects. One effective approach to address this problem is the application of polymeric nanoparticles equipped with targeting units for tumor-specific delivery. For instance dendrimers, highly branched macromolecules, can be equipped with targeting units as well as with anticancer drugs because of their large number of surface functionalities. Amphiphilic block copolymers, which self-assemble in dil…
Polyester and Ester Functionalized Dendrimers
2000
Demand for smart and functional materials has raised the importance of the research of dendritic (Greek = tree-like) molecules in organic and polymer chemistry due to their novel physical and mechanical properties. The properties of linear polymers as well as small discrete molecules are combined in this new architectural class of macromolecules, that can be divided into two families: dendrimers and hyperbranched macromolecules, that differ in their branching sequences. Dendrimers contain symmetrically arranged branches emanating from a core molecule together with a well-defined number of end groups corresponding to each generation. This results in an almost monodisperse three-dimensional g…
Advances in organic and organic-inorganic hybrid polymeric supports for catalytic applications
2016
In this review, the most recent advances (2014–2016) on the synthesis of new polymer-supported catalysts are reported, focusing the attention on the synthetic strategies developed for their preparation. The polymer-supported catalysts examined will be organic-based polymers and organic-inorganic hybrids and will include, among others, polystyrenes, poly-ionic liquids, chiral ionic polymers, dendrimers, carbon nanotubes, as well as silica and halloysite-based catalysts. Selected examples will show the synthesis and application in the field of organocatalysis and metal-based catalysis both for non-asymmetric and asymmetric transformations.
Solution processable phosphorescent dendrimers based on cyclic phosphazenes for use in organic light emitting diodes (OLEDs)
2008
A novel solution processable phosphorescent dendrimer based on cyclic phosphazene (CP) cores has been prepared and used as emissive layers in simple OLED architectures, providing efficiencies of 24.0 cd A^-1 and 16.7 lm W^-1. Frequency selective excitation of single chromophores within shape-persistent multichromophoric dendrimers
2003
Isolated multichromophoric dendrimers containing four perylenemonoimide chromophores were investigated in a confocal microscope which allowed imaging of the same set of dendrimers over a wide temperature range. Emission spectra at low temperature (2 K) reveal narrow zero-phonon lines although the electron-phonon coupling is not negligible. By using fluorescence excitation spectroscopy, the sharp purely electronic zero-phonon lines of single chromophores within a single multichromophoric unit could be isolated in the frequency domain. Selective excitation into a higher energy chromophoric site is followed by rapid directional energy transfer into the lowest energy site from which emission th…
Mannose-Decorated Multicomponent Supramolecular Polymers Trigger Effective Uptake into Antigen-Presenting Cells
2018
A modular route to prepare functional self-assembling dendritic peptide amphiphiles decorated with mannosides, to effectively target antigen-presenting cells, such as macrophages, is reported. The monomeric building blocks were equipped with tetra(ethylene glycol)s (TEGs) or labeled with a Cy3 fluorescent probe. Experiments on the uptake of the multifunctional supramolecular particles into murine macrophages (Mφs) were monitored by confocal microscopy and fluorescence-activated cell sorting. Mannose-decorated supramolecular polymers trigger a significantly higher cellular uptake and distribution, relative to TEG carrying bare polymers. No cytotoxicity or negative impact on cytokine producti…
Self-assembly of janus dendrimers into uniform dendrimersomes and other complex architectures
2010
Janus Drug Delivery Vehicle Efficient drug delivery vehicles need to be produced in a limited size range and with uniform size distribution. The self-assembly of traditional small-molecule and polymeric amphiphiles has led to the production of micelles, liposomes, polymeric micelles, and polymersomes for use in drug delivery applications. Now, Percec et al. (p. 1009 ) describe the self-assembly of Janus-type (i.e., two-headed) dendrimers to produce monodisperse supramolecular constructs, termed “dendrimersomes,” and other complex architectures. The structures, which showed long-term stability as well as very narrow size distributions, were easily produced by the injection of an ethanolic so…
Self-assembly of programmed building blocks into structurally uniform dendrimers.
2005
Selective and independent dimerization of tri- and tetraurea derivatives was used to build up dendritic assemblies which are uniform in size and structure. Dendrimers with the total molecular masses of about 25 000 g/mol were obtained. The existence of uniform assemblies was proved by 1H and 1H DOSY NMR experiments and also by dynamic light scattering.
Synthesis, characterization and properties of functional star and dendritic block copolymers of ethylene oxide and glycidol with oligoglycidol branch…
2009
Abstract Well-defined, four-arm star block copolymers of ethylene oxide and glycidol were prepared via controlled anionic polymerization using protected glycidol. The length of the poly(ethylene oxide) block was varied from DP = 10 to 50, while the length of the short polyglycidol block remained nearly constant, at DP = 4–6. Star block copolymers with hydroxyl groups at the ends of the arms after conversion to the corresponding alkoxides were used as multifunctional macroinitiators for the sequential polymerization of ethylene oxide and protected glycidol. After deprotection, the branched block copolymers of ethylene oxide and glycidol had narrow molar mass distributions and multiple hydrox…
