Characterization of protofibrillar aggregates of bovine serum albumin by tryptophans fluorescence lifetime

2010

We report an experimental study on the thermally induced aggregation of Bovine Serum Albumin at basic pH. In these conditions, we observe the growth of simple protofibrillar structures via the formation of intermolecular beta-sheets promoted by the increased electrostatic repulsion. Here we present a study on the time resolved fluorescence of Tryptophans (Trp) along the aggregation kinetics in the above reported conditions. We use the lifetimes distribution approach as a useful tool for the interpretation of the fluorescence decay in terms of protein conformational substates and interconversion dynamics. Trp fluorescence lifetime depends from protein conformations, also in relation with sol…

Cardiac arrhythmias induced by an electrical stimulation at a cellular level

2008

To provide insights into the impulse propagation between cardiac myocytes, we performed studies of excitation spread with cellular resolution in confluent monolayers of cultured cardiomyocytes (CM). Multisite field potentials have been recorded using microelectrode arrays (MEA) technology in a basal condition and in proarrhythmic conditions induced by a high frequency electrical stimulation. The in vitro observation of spiral waves opens a new way to test the anti-arrhythmic drugs or strategies at cellular level.

Effect of RF Field Inhomogeneity and Sample Restriction on Spectral Resolution of CP/MAS-13C NMR Spectra of Natural Organic Matter

2010

It is well known that the induced B1 magnetic field in an NMR coil is inhomogeneously distributed. However, this issue has so far received little attention in the field of environmental NMR. As this research field often aims at quanti- tative results as well as relaxation phenomena, the repercussions of such inhomogeneity on peak integrals and relaxation times need to be taken into account. The objective of the present study was to test standard recording conditions on different sample positions in an NMR coil in order to determine the effect of the RF field inhomogeneity on the spectrum of a molecularly complex humic material and on some standard molecules of known structure and conformati…

The performance of the ATHENA X-ray Integral Field Unit

2018

The X-ray Integral Field Unit (X-IFU) is a next generation microcalorimeter planned for launch onboard the Athena observatory. Operating a matrix of 3840 superconducting Transition Edge Sensors at 90 mK, it will provide unprecedented spectro-imaging capabilities (2.5 eV resolution, for a field of view of 5') in the soft X-ray band (0.2 up to 12 keV), enabling breakthrough science. The definition of the instrument evolved along the phase A study and we present here an overview of its predicted performances and their modeling, illustrating how the design of the X-IFU meets its top-level scientific requirements. This article notably covers the energy resolution, count-rate capability, quantum …

A mathematical study on effective wavenumber, an operative computation procedure, and its use with radiance-temperature relationships

2003

In this paper we study the existence of effective wavenumber for any sensor by means of simple mathematics. We implement a numeric algorithm, based on the developed mathematical background, into a computer program named 'srf2radi', which is freely available via anonymous File Transfer Protocol (FTP) at ftp://ftp.uv.es/pub/src/. The number of effective wavenumbers for each temperature and their accuracy are also computed and studied for NOAA-16 Advanced Very High Resolution Radiometer Version 3 (AVHRR/3) channel 4. These effective wavenumbers can be used to obtain radiance-temperature relationships with physical meaning for any sensor in user-defined temperature intervals.

Advanced fluorescence microscopy for in vivo imaging of neuronal activity

2019

Brain function emerges from the coordinated activity, over time, of large neuronal populations placed in different brain regions. Understanding the relationships of these specific areas and disentangling the contributions of individual neurons to overall function remain central goals for neuroscience. In this scenario, fluorescence microscopy has been proved as the tool of choice for in vivo recording of brain activity. Optical advances combined with genetically encoded indicators allow a large flexibility in terms of spatiotemporal resolution and field of view while keeping invasiveness in living animals to a minimum. Here we describe the latest advancements in the field of linear and nonl…

Phonon Driven Floquet Matter.

2018

The effect of electron–phonon coupling in materials can be interpreted as a dressing of the electronic structure by the lattice vibration, leading to vibrational replicas and hybridization of electronic states. In solids, a resonantly excited coherent phonon leads to a periodic oscillation of the atomic lattice in a crystal structure bringing the material into a nonequilibrium electronic configuration. Periodically oscillating quantum systems can be understood in terms of Floquet theory, which has a long tradition in the study of semiclassical light-matter interaction. Here, we show that the concepts of Floquet analysis can be applied to coherent lattice vibrations. This coupling leads to p…

Survival of Floquet–Bloch States in the Presence of Scattering

2021

Floquet theory has spawned many exciting possibilities for electronic structure control with light, with enormous potential for future applications. The experimental demonstration in solids, however, remains largely unrealized. In particular, the influence of scattering on the formation of Floquet-Bloch states remains poorly understood. Here we combine time- and angle-resolved photoemission spectroscopy with time-dependent density functional theory and a two-level model with relaxation to investigate the survival of Floquet-Bloch states in the presence of scattering. We find that Floquet-Bloch states will be destroyed if scattering-activated by electronic excitations-prevents the Bloch elec…

Bed Topography Reconstruction in a Large Amplitude Meandering Flume: Application of Close Range Photogrammetry

2014

Natural rivers are characterized by continuous variations in bed topography, especially along curved reaches. High resolution topographic data are necessary to analyze the mutual interactions between the downstream flow and the cross-stream flow, which determine the distribution of the bed-shear stress along the channel. Because of the difficulty in acquiring good and accurate data in rivers, the major part of studies have been conducted in laboratory flumes. This paper reports on a laboratory study in which the automatic digital photogrammetric survey was applied to derive the high-resolution Digital Surface Model (DSM) of the bed topography in a large amplitude meandering flume. In order …
