Zearalenone (ZEA), α-zearalenol (α-ZEL) and β-zearalenol (β-ZEL) (ZEA's metabolites) are co/present in cereals, fruits or their products. All three with other compounds, constitute a cocktail-mixture that consumers (and also animals) are exposed and never entirely evaluated, nor in vitro nor in vivo. Effect of ZEA has been correlated to endocrine disruptor alterations as well as its metabolites (α-ZEL and β-ZEL); however, toxic effects associated to metabolites generated once ingested are unknown and difficult to study. The mechanism of the reaction between a prototypical β-ketoester, ethyl 2-oxocyclopentanonecarboxylate, and N-methylindole has been studied experimentally by monitoring the temporal course of the reaction by (1)H NMR, kinetic isotope effect studies, and control experiments. [EN] Chiral complexes of BINOL-based ligands with zirconium tert-butoxide catalyze the Friedel-Crafts alkylation reaction of indoles with beta-trifluoromethyl-alpha,beta-unsaturated ketones to give functionalized indoles with an asymmetric tertiary carbon center attached to a trifluoromethyl group. The reaction can be applied to a large number of substituted alpha-trifluoromethyl enones and substituted indoles. The expected products were obtained with good yields and ees of up to 99%.

Es werden die Additionen an Tetraphenylcyclopentadienon (1+23), N-Phenylmaleinimid (1+45) und Tropon (1+68 bzw. 10) beschrieben. Auf der Basis von MNDO-Rechnungen werden Peri-, Regio- und Stereoselektivitat anhand von primaren und sekundaren Orbitalwechselwirkungen diskutiert. Preparation of 1,5-Cyclooctadiyne and 1,3,5,7-Cyclooctatetraene from 1,5-Cyclooctadiene Bromination of 1,5-cyclooctadiene (1) and stepwise dehydro-bromination first with KOtBu and then with KOtBu/18-crown-6 yields 1,5-cyclooctadiyne (5). A prolonged interaction of the base causes a complete transformation to cyclooc-tatetraene (6). Diyne 5 and even more the intermediate enyne 4 are highly reactive dienophiles. The cycloadducts 10–14 formed with 1,3-cyclohexadiene, carbon disulfide, and tetraphenylcyclopentadienone were isolated and characterized. Treatment of 5 with titanium tetrachloride induces a vigorous polymerization.

Oxidative stress is involved in the pathogenesis of various neurodegenerative disorders, conventional antioxidant strategies have yet been of limited success. We have employed transgenic Caenorhabditis elegans expressing DsRed2 in dopaminergic neurons and CFP pan-neuronally, to characterize in larval and adult animals the effects of rotenone and 1-methyl-4-phenyl-pyridinium (MPP(+)) on the dopaminergic system. Rottlerin induces a transformed phenotype in human keratinocytes.

PKCdelta plays a fundamental role in cell cycle control. Consistent with its proposed tumour suppressor function, ras transfection of the human keratinocyte cell line HaCaT results in a loss of PKCdelta expression mediated by TGFalpha (Exp. Cell Res., 219, 299, 1995). To get more insight into the role of PKCdelta in keratinocytes, we investigated the effects of Rottlerin, a specific inhibitor of protein kinase Cdelta, in HaCaT cells.
