Streptococcus pneumoniae sortāzes A inhibitoru atlase multirezistentu patogēnu izraisīto infekcijas slimību terapijai

Darba mērķis ir atrast jaunas novatoriskas vielas, kas inhibē Grampozitīvās baktērijas Streptococcus pneumoniae transpeptidāzi sortāzi A. Šo inhibitoru atlasei tika izmantota rekombinanta S.pneumoniae sortāzes A katalītisko domēnu kodējoša konstrukcija bez N terminālā reģiona. Šāda rekombinanta transpeptidāze tika ekspresēta E.coli ekspresijas sistēmā un sintezētais proteīns izrādījās šķīstošs. Pēc attīrīšanas sortāze tika izmantota inhibitoru atlasei, izmantojot skrīningu, kas balstās uz fluorescences rezonanses enerģijas pārnesi (FRET). Excited State N−H Tautomer Selectivity in the Singlet Energy Transfer of a Zinc(II)-Porphyrin-Truxene-Corrole Assembly

International audience; An original corrole-containing polyad for S-1 energy transfer, in which one zinc(II)-porphyrin donor is linked to two free-base corrole acceptors by a truxene linker, is reported. This polyad exhibits a rapid zinc(II)-porphyrin*free-base corrole transfer (4.83x10(10)s(-1); 298K), even faster than the tautomerization in the excited state processes taking advantage of the good electronic communication provided by the truxene bridge. Importantly, the energy transfer process shows approximately 3-fold selectivity for one corrole N-H tautomer over the other even at low temperature (77K). Role of RNA Motifs in RNA Interaction with Membrane Lipid Rafts: Implications for Therapeutic Applications of Exosomal RNAs

RNA motifs may promote interactions with exosomes (EXO-motifs) and lipid rafts (RAFT-motifs) that are enriched in exosomal membranes. These interactions can promote selective RNA loading into exosomes. We quantified the affinity between RNA aptamers containing various EXO- and RAFT-motifs and membrane lipid rafts in a liposome model of exosomes by determining the dissociation constants. Analysis of the secondary structure of RNA molecules provided data about the possible location of EXO- and RAFT-motifs within the RNA structure. FRET fluorescento proteīnu balstītu pH sensoru izveide

Iekššūnas vides pH ir būtisks parametrs, kas raksturo šūnā notiekošos procesus, kā arī šūnas vitalitāti. Fluorescento proteīnu pielietošana pH noteikšanai ir daudzsološa metode. Pieejami atšķirīgas pH jutības fluorescentie proteīni, tie neuzrāda augstu toksicitāti un invazivitāti, padarot tos piemērotus dzīvu šūnu pētījumiem. Maģistra darba mērķis bija izveidot fluorescento proteīnu saturošus konstruktus ar atšķirīgu pH jutības slieksni, izmantojot Aquamarine, ECFP, EYFP un Citrine fluorescentos proteīnus, kas darbosies kā pH sensoru sistēma dzīvās šūnās, izmantojot FRET metodi. Šajā darbā tiek pētīti divi Alq3 atvasinājumi un PVK kombinācijā ar četriem DCM atvasinājumiem. Pētītie savienojumi ar rotējošā diska metodi no šķīduma var veidot amorfas plānas kārtiņas. Tika izmērītas šādas paraugu optiskās īpašības: absorbcijas un emisijas spektri, fotoluminescences kinētika un kvantu iznākums. Labākajām sistēmām tika veikti pastiprinātas spontānās emisijas (ASE) mērījumi. Plāno kārtiņu absorbcijas un emisijas mērījumi liecina, ka FRET starp Alq3 un DCM molekulām ir iespējama Alq3 emisijas spektra un DCM absorbcijas spektra pārklāšanās dēļ. A DNA‐Encoded FRET Biosensor for Visualizing the Tension across Paxillin in Living Cells upon Shear Stress

Paxillin is a potential participant in the direct intracellular force transmission which is considered as the foundation of cells sensing and responding to extracellular environment. However, the detection of tension across paxillin has not been achieved due to lacking microsized tools. Herein, a paxillin tension sensor (PaxTs) based on Fluorescence Resonance Energy Transfer (FRET) technique was constructed. A FRET-based random screening assay was used to generate hit compounds as sortase A inhibitors that allowed us to identify ethyl 3-oxo-2-(2-phenylhydrazinylidene)butanoate as an example of a new class of sortase A inhibitors. Other analogues were generated by changing the ethoxycarbonyl function for a carboxy, cyano or amide group, or introducing substituents in the phenyl ring of the ester and acid derivatives. The most active derivative found was 3-oxo-2-(2-(3,4dichlorophenyl)hydrazinylidene)butanoic acid (2b), showing an IC50 value of 50 µM. Darba mērķis ir atrast jaunus inhibitorus, kas kavē Listeria monocytogenes transpeptidāzes sortāzes A veikto hidrolīzes reakciju, inhibējot sortāzes A darbību. Šī mērķa sasniegšanai tika izveidota DNS konstrukcija, kas kodē Listeria monocytogenes sortāzes A katalītisko domēnu. Tā kā vislabāk izpētīta ir Staphylococcus aureus sortāze A, papildus tika izveidota tās katalītisko domēnu kodējoša DNS konstrukcija. Ekspresija tika sekmīgi veikta E. coli ekspresijas sistēmā, abi proteīni bija šķīstoši.
