We present an X-ray fluorescence spectroscopy (XRF) study combined with a multivariate approach that allow to detect compositional differences and similarities among the glass supports of a large set of reverse glass paintings belonging to the collection of the Mistretta museum. Reverse painting on glass is an old decorative technique used since the Roman time consisting in applying a cold paint layer on the reverse side of a glass support. The collection shows a large spreading of provenience and dating of the items. The feasibility of partial least squares (PLS) regression modeling of X-ray fluorescence (XRF) spectra of estuarine sediments has been evaluated as a tool for rapid trace element content monitoring. Multivariate PLS calibration models were developed to predict the concentration of Al, As, Cd, Co, Cr, Cu, Fe, Mg, Mn, Ni, Pb, Sn, V and Zn in sediments collected from different locations across the estuary of the Nerbioi-Ibaizabal River (Metropolitan Bilbao, Bay of Biscay, Basque Country). A simple, fast, inexpensive, and reliable method useful for the simultaneous, routine determination of thiamin and riboflavin in mushrooms is examined. It uses the extraction procedure, with slight modifications, proposed by the AOAC for the extraction of thiamin and riboflavin, followed by a liquid chromatographic separation on a reversed-phase Spherisorb ODS column with methanol/water as mobile phase gradient. Staphylococcal alpha-toxin, the prototype of an oligomerizing, pore-forming cytotoxin, is sensitive to biochemical modifications and cannot be labeled with biotin or fluorescein under preservation of its biological activity. In this study, we have used site-directed mutagenesis to introduce cysteine residues at positions 69, 130, and 186. Each mutant was fully and rapidly reactive with several sulfhydryl-specific reagents, indicating superficial location. This study characterizes the phosphorylated pathway of Ser biosynthesis (PPSB) in Arabidopsis thaliana by targeting phosphoserine phosphatase (PSP1), the last enzyme of the pathway. Lack of PSP1 activity delayed embryo development, leading to aborted embryos that could be classified as early curled cotyledons. The embryo-lethal phenotype of psp1 mutants could be complemented with PSP1 cDNA under the control of Pro35S (Pro35S:PSP1). Abstract Compartmentalization of chemical reactions inside cells are a fundamental requirement for life. Encapsulins are self‐assembling protein‐based nanocompartments from the prokaryotic repertoire that present a highly attractive platform for intracellular compartmentalization of chemical reactions by design. Using single‐molecule Förster resonance energy transfer and 3D‐MINFLUX analysis, we analyze fluorescently labeled encapsulins on a single‐molecule basis. The human inducible heat shock protein 70 (hHsp70), which is involved in several major pathologies, including neurodegenerative disorders and cancer, is a key molecular chaperone and contributes to the proper protein folding and maintenance of a large number of protein structures. Despite its role in disease, the current structural knowledge of hHsp70 is almost exclusively based on its Escherichia coli homolog, DnaK, even though these two proteins only share ~50 % amino acid identity. Au–MnO heteroparticles were immobilized on the surface of TiO2 nanowires and tagged subsequently with a fluorescent ligand. The immobilization of the Au@MnO heteroparticles was achieved by functionalizing the TiO2 nanowire support with a polymer containing catechol anchor groups for binding to the metal oxide surface and amine groups for conjugation to the Au domains of the Au@MnO heteroparticles.
