In vitro synthesis of factor B of the alternative pathway of complement activation by mouse peritoneal macrophages

Factor B of the alternative pathway of complement activation was shown to be synthesized and secreted by unstimulated mouse peritoneal macrophages. The activity of B in the culture supernatants from macrophage monolayers was detected by consumption of C3 in reaction mixtures containing supernatant and guinea pig factors C3, D and insoluble C3b. Using a monospecific antiserum, factor B in concentrated culture supernatants was shown by immunodiffusion and immunoelectrophoresis to be identical to factor B in mouse plasma and to form a characteristic complex with cobra venom factor in the presence of D. Mit intracellularen Mikroelektroden werden Aktionspotentiale von Meerschweinchenvorhofen in vitro abgeleitet. Nach Vorbehandlung der Tiere mit Thyroxin (5 Tage lang 1 mg dl-Thyroxin je Kilogramm) ist die Repolarisationsphase und damit die Refraktarperiode im Vergleich zu Kontrolltieren bei gleicher Schlagfrequenz verkurzt.

Influence of Macrophage Activation on the Synthesis of Complement Components C2, C3, C4

Macrophages are a major site of complement synthesis. In the guinea pig production of complement components C1, C2, C4, C3, D, B, and P by peritoneal macrophages has been demonstrated (reviewed in Ref. 1). Whereas marked differences exist in biological activity between resident, elicited and activated macrophages (2), we investigated whether this holds true for the synthesis and secretion of complement components C2, C3, and C4.

Anticomplementary Activity of Guinea Pig Serum Euglobulin: Its Relation to C 1 and to TAMe Esterase

The interaction of complement with a suitable antigen-antibody aggregate triggers a chain of chemical reactions involving consecutively the nine factors of complement (C1 to C9). The triggering reaction is identical with the material fixation of Cl to the immune complex ; this leads to the formation of Ag-Ab-C1. By its fixation Cl is converted from its chemically inert form to a highly reactive state (“fixed” or “activated” C1). The reactivity of fixed Cl is directed first against C4. Consequently C4 rapidly disappears from the fluid phase in the presence of fixed Cl. A strikingly similar process occurs spontaneously in a fraction of guinea pig serum [5, 6].

Linkage of C4 and C4 deficiency to BF and GPLA

The C4, Bf, and GPLA phenotypes of homo- and heterozygous C4-deficient guinea pigs were studied. The electrophoretic patterns suggest that the deficiency in circulating C4 results from an impaired structural gene, allelic to the C4F, C4S, and C4S1 alleles at the C4 locus. In family studies, support for linkage of C4 and Bf to theGPLA system was obtained. The defective gene appears to be the fourth allele, which is rare, in the polymorphism of the fourth component of guinea pig complement.

An isolierten linken Meerschweinchenvorhofen wurde der Einflus von Calcium und Natrium auf das Verhalten der Kaliumkontraktur und des Membranpotentials mit folgenden Ergebnissen untersucht: Die Ergebnisse zeigen, das sich die Kontraktur des Meerschweinchenvorhofs in ihrem Verlauf und ihrer Beeinflusbarkeit durch Calcium und Natrium nicht grundsatzlich von der des Froschherzens unterscheidet. Daraus wird geschlossen, das ihr in beiden Fallen der gleiche Mechanismus zugrunde liegt.

Um Aufschlus uber den Wirkort von Herzglykosiden in der Herzmuskelzelle zu erhalten, wurde die subcellulare Verteilung von 3H-Digitoxin und 3H-Strophanthin untersucht und mit dem positiv inotropen Effekt verglichen. Meerschweinchenherzen wurden nach der Langendorff-Methode mit glykosidhaltiger Tyrodelosung (3 · 10−7 bzw. 2,8 · 10−7 g/ml; 37°C) perfundiert; die Kontraktionskraft wurde fortlaufend isotonisch registriert. SUMMARY The addition of 10 mM CaCl2 to glutaraldehyde fixative leads to the formation of small electron-dense deposits in the organ of Corti of the guinea-pig. These precipitates are mainly attached to cell membranes in contact with different extracellular lymphatic fluids. A higher number of precipitates is localized in the acellular parts of tectorial and basilar membrane. Electron energy-loss spectroscopy (EELS) was used to determine the elemental composition of the deposits formed. The spectra showed a prominent signal at the Ca2+ L2,3 ionization edge. Possible Ca 2+ -dependent mechanism of apical outer hair cell modulation within the cochlea of the guinea pig

Calcium ions were precipitated with potassium antimonate after injection of the inorganic calcium channel blocker MnCl2 or the inorganic potassium channel blockers BaCl2 or CsCl into the perilymph of the scala vestibuli of the guinea pig. The spatial distribution of the formed histochemical reaction products within the organ of Corti was studied by energy-filtering transmission-electron microscopy.
