Cytoprotection assessment against mycotoxins on HepG2 cells by extracts from Allium sativum L

Abstract Cytoprotection effects of Allium sativum L garlic extract from a local garlic ecotype from Ferrara (Italy) on hepatocarcinoma cells, HepG2 cells, is presented in this study. This garlic type is known as Voghiera garlic and has been characterized as PDO (Protected designation of Origin) product. In this work, we describe how stereochemically complex polycyclic compounds can be generated by applying a synthetic sequence comprising an intramolecular Ugi reaction followed by a Pictet-Spengler cyclization on steroid-derived scaffolds. The resulting compounds, which combine a fragment derived from a natural product and a scaffold not found in nature. are both structurally distinct and globally similar to natural products at the same time, and interrogate an alternative region of the chemical space. Dabas vielu lipīdu uzkrāšanās un acetilholīnesterāzes inhibīcija cilvēku aknu šūnu līnijā HepG2 in vitro

Taukaino aknu slimība ir viena no izplatītākajām aknu slimībām attīstītajās valstīs, un tiek iedalīta alkohola un ne-alkohola taukaino aknu slimībā. Taukaino aknu slimībai ir potenciāls progresēt līdz steatohepatītam un cirozei, bet joprojām nav atrastas mērķētas terapijas slimības progresijas palēnināšanai. Maģistra darba mērķis ir pētīt Vaccinium ģints ogu izspaidu ekstraktu iedarbību uz acetilholīna koncentrāciju, acetiholīnesterāzes aktivitāti un lipīdu akumulāciju HepG2 šūnu modelī. Pētījuma rezultāti liecina, ka ogu izspiedu ekstrakti samazina lipīdu akumulāciju šūnās un gan ekstrakti, gan zināmais aknu šūnu funkciju uzlabotājs silimarīns inhibē acetilholīnesterāzes (AChE) aktivitāti.…

Selenofēnkumarīnu un selenofēnhinolonu salīdzinošā ietekme uz HepG2 šūnu fermentatīvo antioksidatīvo sistēmu

Latvijas Organiskās sintēzes institūtā izstrādātas metodes jaunu selenofēn[3,2-c] un [2,3-c]kumarīnu un selenofēn[3,2-c] un [2,3-c]hinolonu sintēzei, kuriem pierādīts antiproliferatīvais efekts in vitro. Darbā tika noteikts savienojumu inhibējošais efekts uz trim hepatomas HepG2 šūnu galvenajiem antioksidatīvajiem fermentiem (GPx, TrxR, SOD) ar mērķi noskaidrot savienojumu antiproliferatīvā efekta darbības mehānismus. Pētījumā konstatēts, ka visi savienojumi ir efektīvi GPx inhibitori. TrxR aktivitāti inhibē selenofēnhinoloni un [3,2-c] aizvietotie selenofēnkumarīni, savukārt SOD aktivitāti ietekmē tikai divi no savienojumiem. In an extensive screening, the antiviral, antitrypanosomal and anticancer properties of extracts from 82 plants used in traditional Chinese medicine and European phytomedicine were determined. Several promising plants that were highly effective against hepatitis B virus (HBV), bovine viral diarrhoea virus (BVDV)—a flavivirus used here as a surrogate in vitro model of hepatitis C virus, trypanosomes (Trypanosoma brucei brucei) and several cancer cell lines were identified.
