Coupling of the guanosine glycosidic bond conformation and the ribonucleotide cleavage reaction: implications for barnase catalysis.

2007

To examine the possible relationship of guanine-dependent GpA conformations with ribonucleotide cleavage, two potential of mean force (PMF) calculations were performed in aqueous solution. In the first calculation, the guanosine glycosidic (Gχ) angle was used as the reaction coordinate, and computations were performed on two GpA ionic species: protonated (neutral) or deprotonated (negatively charged) guanosine ribose O2 ′. Similar energetic profiles featuring two minima corresponding to the anti and syn Gχ regions were obtained for both ionic forms. For both forms the anti conformation was more stable than the syn, and barriers of ∼4 kcal/mol were obtained for the anti → syn transition. Str…

Stereoselective synthesis of 7,11-guaien-8,12-olides from santonin. Synthesis of podoandin and (+)-zedolactone A.

2000

Photochemical rearrangement of hydroxy ester 2, easily obtained from santonin (1), afforded butenolide 4, a good starting material for the synthesis of 7,11-guaien-8,12-olides. Compound 4 has been transformed into compound 10, which has been used for the synthesis of podoandin (5) and (+)-zedolactone A (ent-6). Regioselective elimination of the acetyl group on C10 afforded directly podoandin (5). For the synthesis of ent-6, a hydroxyl group has been regio- and stereoselectively introduced at the 4alpha-position through the 3alpha,4alpha-epoxide 15. The basic hydrolysis of the 10-acetyl group in compound 18 took place with concomitant intramolecular conjugated addition of the alkoxide to the…

Racemic and Optically Pure Heptahelicene-2-carboxylic Acid: Its Synthesis and Self-Assembly into Nanowire-Like Aggregates

2010

Heptahelicene-2-carboxylic acid was effectively synthesised from suitably functionalised naphthalene building blocks. Methoxy-substituted 1,1'-ethyne-1,2-diylbis(2-but-3-yn-1-ylnaphthalene) was cyclised in the presence of CpCo(CO)(2)/PPh(3) to 2-methoxy-7,8,11,12-tetrahydroheptahelicene, which was converted into heptahelicen-2-yl trifluoromethanesulfonate. This reactive intermediate underwent Pd(OAc)(2)/dppp-catalysed methoxycarbonylation reaction to provide, after hydrolysis, heptahelicene-2-carboxylic acid. The racemate was resolved into enantiomers by semipreparative HPLC on a chiral column. The helicity of (+)-(P)-heptahelicene-2-carboxylic acid was assigned by correlating its CD spectr…

Plant-growth-regulating phosphono peptides

1990

Phosphono peptides exhibited plant‐growth‐regulating activity when tested on Lepidium sativum and Cucumis sativus. The studies have provided evidence that the mechanism of action of these compounds may involve the uptake of the phosphono peptides into the plant, intracellular hydrolysis of the peptide bond and release of the P‐terminal aminoalkylphosphonic acid. This acid or its metabolite is then probably responsible for the observed physiological effect.

CeO2−x nanorods with intrinsic urease-like activity

2018

The large-scale production and ecotoxicity of urea make its removal from wastewater a health and environmental challenge. Whereas the industrial removal of urea relies on hydrolysis at elevated temperatures and high pressure, nature solves the urea disposal problem with the enzyme urease under ambient conditions. We show that CeO2−x nanorods (NRs) act as the first and efficient green urease mimic that catalyzes the hydrolysis of urea under ambient conditions with an activity (kcat = 9.58 × 101 s−1) about one order of magnitude lower than that of the native jack bean urease. The surface properties of CeO2−x NRs were probed by varying the Ce4+/Ce3+ ratio through La doping. Although La substit…

Screening pretreatment methods to enhance thermophilic anaerobic digestion of pulp and paper mill wastewater treatment secondary sludge

2013

Abstract The effect of hydrothermal (150 °C for 10 min and 70 °C for 40 min), enzymatic (Accelerase 1500, 0.07 g/g volatile solids (VS)), ultrasound (45 kHz for 30 min) and chemical pretreatments (HNO3 at pH 3 and NaOH at pH 12) alone or in combination on the chemical composition and methane yield of the pulp and paper mill secondary sludge was studied in batch assays at 55 °C. In total, 12 different pretreatment combinations were compared. Chemical analyses showed that all pretreatments except for HNO3 and ultrasound pretreatments improved the organic matter solubilization. Among the studied pretreatments, hydrothermal (150 °C, 10 min) pretreatment alone or in combination with enzymatic an…

Monoterpenes in grape juice and wines.

2000

The importance of monoterpenes on varietal flavour of wines has been reviewed. These compounds were mainly found linked to sugar moieties in the grape juice and wines, showing no olfactive characteristics. In this way, mechanisms to liberate terpenes were studied, making a comparative study between acidic and enzymic hydrolysis of terpene glycosides. Finally, analytical techniques developed to study these compounds, in both free or glycosidically forms, and also to fractionate glycosidic precursors, have been discussed.

Chemoselective Removal of Protecting Groups from O-Glycosyl Amino Acid and Peptide (Methoxyethoxy)ethyl Esters Using Lipases and Papain

1996

The selective C-terminal deprotection of O-glycopeptide (methoxyethoxy)ethyl esters is achieved under mild conditions (pH 6.6, 37 degrees C) by enzymatic hydrolysis using papain or lipase M from Mucor javanicus to give building blocks useful for chain-extending glycopeptide synthesis. On the other hand, the selective removal of acetyl protecting groups from the saccharide portion of glycopeptides is accomplished by alternative enzymatic hydrolysis with lipase WG from wheat germ to furnish model substrates for enzymatic glycosyl transfer reactions in order to extend the carbohydrate side chain of these conjugates.

Chemie an starren Grenzflächen, 10. Kinetisches Verhalten einiger an „Isothiocyanato-Aerosil” immobilisierter Esterasen – Analogversuche mit Insulin

1988

Amino-Aerosil wird mit Thiophosgen nach Gl. (1) in Isothiocyanato-Aerosil 2 (NCS-Aerosil) ubergefuhrt.

Ursane-Type Triterpene Saponins fromZygophyllum geslini

2007

Four new ursane-based triterpene glycosides, compounds 1–4, as well as the known glycosides zygophylosides E, G, and H, and 3-O-(β-D-quinovopyranosyl)quinovic acid 28-(O-β-D-glucopyranosyl) ester, were isolated from the BuOH-soluble fraction of the MeOH/H2O 7 : 3 extracts of Zygophyllum geslini (roots or aerial parts). Their structures were established mainly by 1D- and 2D-NMR techniques, in combination with HR-MS analysis and acid hydrolysis.
