Induction of micronuclei in V79 Chinese hamster cells by tetrachlorohydroquinone, a metabolite of pentachlorophenol

Tetrachlorohydroquinone, a metabolite of the fungicide pentachlorophenol, induced significant dose-related increases in micronuclei in V79 Chinese hamster cells without exogenous metabolic activation. The lowest observed effective dose was 10 microM, where the relative survival was about 62%. At the highest dose tested, 20 microM, the relative survival was about 8% and the frequency of cells with micronuclei was about 6 times the solvent control frequency. The induction of micronuclei by tetrachlorohydroquinone was significantly inhibited by the hydroxyl radical scavenger dimethyl sulfoxide at 5% (v/v).

The bioengineered tissue plasminogen activator tenecteplase is an important treatment modality of acute myocardial infarction recommended by international guidelines. Following introduction of originator tenecteplase (brand names Metalyse(®) and TNKase(®)), a "biosimilar" tenecteplase became available for commercial use in India under the brand name Elaxim(®) in the absence of Indian biosimilar guidelines which came into force from September 15th, 2012. Regio- and stereoselective metabolism mediated by cytochrome P450 (CYP) and metabolite-dependent cytotoxicity of benzo[c]phenanthrene (B[c]Ph) and its trans-3,4-dihydrodiol, the metabolic precursor of the carcinogenic fjord-region B[c]Ph-3,4-dihydrodiol 1,2-epoxides (B[c]PhDE), were investigated with V79 Chinese hamster cells genetically engineered for three rat and six human CYP isoforms. The vanilloid receptor 1 (VR1) is a heat-activated cation channel which also responds to capsaicin and other chemical stimuli. Protein kinase C has a stimulatory effect on VR1 activity, either alone or after activation with capsaicin. The influence of the cAMP-signaling pathway on the effects of capsaicin is controversial. Correlations in palmitoylation and multiple phosphorylation of rat bradykinin B2 receptor in Chinese hamster ovary cells.

Rat bradykinin B2 receptor from unstimulated Chinese hamster ovary cells transfected with the corresponding cDNA has been isolated, and subsequent mass spectrometric analysis of multiple phosphorylated species and of the palmitoylation attachment site is described. Glutathione content of V79 cells in two- or three-dimensional culture

The cellular glutathione (GSH) content of two- and three-dimensional cell cultures of V79 hamster lung cells has been studied. As previously described, cells in monolayer cultures show a decrease in GSH when they reach the confluent state. Three-dimensional cell cultures (multicell spheroids) allow a smoother transition from the initial proliferating to the nonproliferating status, and they show a central area of necrosis when a certain diameter is reached. Correlation of the Extent of Fjord-Region Oxidation with DNA Binding and Mutagenicity of the Enantiomeric 11,12-Dihydrodiols of Dibenzo[a,l]pyrene

Abstract In vitro studies on the hepatic biotransformation of the enantiomeric trans-11,12-dihydrodiols of dibenzo[a,l]pyrene (DB[a,l]P) using microsomal fractions of animals pretreated with Aroclor 1254 revealed that the formation of fjord-region dihydrodiol epoxides strongly depends on the absolute configuration of the substrate. Both the (-)-11R,12R- and the (+)-11S,12S-enantiomer are converted diastereoselectively to the (-)- and (+)-anti-dihydrodiol epoxide, respectively, by either rat or mouse liver microsomes. Addition of BrdUrd in combination with prednisolone to HSVi-transformed hamster embryo cells induces an alkaline phosphatase (AP). FdUrd enhances, dThd reduces the inducing capacity of BrdUrd and prednisolone. Induction is prevented by addition of cycloheximide or of cytosine arabinoside. BrdUrd reduces transport and phosphorylation of exogenously applied labeled Urd, especially the amount of UTP, UDP and Urd-diphosphate sugars. The Lineweaver-Burk-plot of uridine-up­take after addition of BrdUrd reveales the characteristics of the mixed type inhibition (competi­tive-noncompetitive).
