Critical Structural Defects Explain Filamin A Mutations Causing Mitral Valve Dysplasia
2019
Mitral valve diseases affect approximately 3% of the population and are the most common reasons for valvular surgery because no drug-based treatments exist. Inheritable genetic mutations have now been established as the cause of mitral valve insufficiency, and four different missense mutations in the filamin A gene (FLNA) have been found in patients suffering from non-syndromic mitral valve dysplasia (MVD). The FLNA protein is expressed, in particular, in endocardial endothelia during fetal valve morphogenesis and is key in cardiac development. The FLNA-MVD causing mutations are clustered in the N-terminal region of FLNA. How the mutations in FLNA modify its structure and function, have mos…
Retene, pyrene and phenanthrene cause distinct molecular-level changes in the cardiac tissue of rainbow trout (Oncorhynchus mykiss) larvae, Part 2 – …
2020
Polycyclic aromatic hydrocarbons (PAHs) are global contaminants of concern. Despite several decades of research, their mechanisms of toxicity are not very well understood. Early life stages of fish are particularly sensitive with the developing cardiac tissue being a main target of PAHs toxicity. The mechanisms of cardiotoxicity of the three widespread model polycyclic aromatic hydrocarbons (PAHs) retene, pyrene and phenanthrene were explored in rainbow trout (Oncorhynchus mykiss) early life stages. Newly hatched larvae were exposed to sublethal doses of each individual PAH causing no detectable morphometric alterations. Changes in the cardiac proteome and metabolome were assessed after 7 o…
Vernacular budgeting and accounting routines – a longitudinal constructive case study
2022
PurposeThis case study aims to report a longitudinal analysis of the development and use of local “vernacular” accounting practice and a digital rolling-forecast system known as TeamBudget in a public sector organization.Design/methodology/approachThe study employs the constructive research approach which utilizes participative observation in the development of TeamBudget over the 15 years since 2004. The empirical data utilized includes eight interviews and documentary data for the system created.FindingsThe study demonstrates how the actions of employees responsible for developing a locally relevant financial planning system, TeamBudget, facilitated the emergence of new accounting routine…
Kurzweil-Henstock type integration on Banach spaces
2004
In this paper properties of Kurzweil-Henstock and Kurzweil-Henstock-Pettis integrals for vector-valued functions are studied. In particular, the absolute integrability for Kurzweil-Henstock integrable functions is characterized and a Kurzweil-Henstock version of the Vitali Theorem for Pettis integrable functions is given.

Luokittelumenetelmän evaluointimenetelmien vertailu pienten aineistojen tapauksessa simulointikokein
2016
Luokitteluanalyysin tyypillinen ongelma on mallin valinta ja/tai muuttujien eli piirteiden valinta. Tavoitteena on estimoida luotettavasti luokitteluvirheen todennäköisyys opetusaineiston avulla - ideaalissa tilanteessa käyttäen riippumatonta testiaineistoa. Tällöin saadaan estimaatti yleistämisvirheelle, joka kuvaa opetusaineistosta estimoidun mallin kykyä ennustaa riippumatonta testiaineistoa. Usein kuitenkin aineiston niukkuudesta johtuen ei voida käyttää erillistä riippumatonta testiaineistoa, jolloin ei myöskään voida estimoida suoraan yleistämisvirhettä. Tällöin voidaan kuitenkin käyttää otoksen uudelleenkäyttömenetelmiä, joista käytetyimmät lienevät ristiinvalidointi ja bootstrap. Me…
Neutrino transport in coherent quasiparticle approximation
2012
We introduce the coherent quasiparticle approximation (cQPA), a model in thermal quantum field theory which describes various effects of temporally varying thermal medium on particle propagation. We present the cQPA Feynman rules and develop related calculational tools. Using these methods we calculate neutrino self energies in the Standard Model and derive an equation of motion for neutrino propagation in a very general framework. Some immediate implications of this equation are discussed. (More detailed abstract in the PDF file.)
Magnomechanics in suspended magnetic beams
2021
Cavity optomechanical systems have become a popular playground for studies of controllable nonlinear interactions between light and motion. Owing to the large speed of light, realizing cavity optomechanics in the microwave frequency range requires cavities up to several mm in size, hence making it hard to embed several of them on the same chip. An alternative scheme with much smaller footprint is provided by magnomechanics, where the electromagnetic cavity is replaced by a magnet undergoing ferromagnetic resonance, and the optomechanical coupling originates from magnetic shape anisotropy. Here, we consider the magnomechanical interaction occurring in a suspended magnetic beam -- a scheme in…

Evolutionary plasticity of SH3 domain binding by Nef proteins of the HIV-1/SIVcpz lentiviral lineage
2021
The accessory protein Nef of human and simian immunodeficiency viruses (HIV and SIV) is an important pathogenicity factor known to interact with cellular protein kinases and other signaling proteins. A canonical SH3 domain binding motif in Nef is required for most of these interactions. For example, HIV-1 Nef activates the tyrosine kinase Hck by tightly binding to its SH3 domain. An archetypal contact between a negatively charged SH3 residue and a highly conserved arginine in Nef (Arg77) plays a key role here. Combining structural analyses with functional assays, we here show that Nef proteins have also developed a distinct structural strategy—termed the "R-clamp”—that favors the formation …
