Evaluation of the Stratus® CS Acute Care™ D-dimer assay (DDMR) using the Stratus® CS STAT Fluorometric Analyzer: A prospective multisite study for ex…
2012
Abstract Background D-dimer testing is an integral part of the diagnostic algorithm in excluding patients with venous thromboembolism. In this study, we prospectively evaluated the Stratus DDMR D-dimer test in patients suspected of pulmonary embolism (PE) and deep vein thrombosis (DVT). Methods Patients suspected of venous thromboembolism were prospectively enrolled at four different clinical sites, with sodium citrate and lithium heparin plasma was tested using the DDMR D-dimer test on the Stratus CS analyzer. Results 1,012 patients were enrolled for analysis, with 85/603 (14.1%) patients with PE and 80/443 (18.1%) with DVT, and four of the patients (0.4%) with PE and DVT. For the samples …
Neuromuscular adaptations and serum hormones in women during short-term intensive strength training
1992
The effects were investigated in ten women of intensive heavy resistance strength training lasting for 3 weeks on electromyographic (EMG) activity, muscle cross-sectional area (CSA) and voluntary force production characteristics of leg extensor muscles. Blood samples for the determinations of serum hormones were taken from five of the subjects. Significant increases occurred in the higher force portions of the isometric force-time curve with an increase of 9.7 (SD 8.4)% (P less than 0.01) in maximal peak force. An increase of 15.8 (SD 20.9)% (P less than 0.05) took place also in the maximal neural activation (integrated EMG) of the trained muscles, while an enlargement of 4.6 (SD 7.4)% (P l…
Characteristics of urinary luteinizing hormone (LH) during the induction of LH surges of different magnitude in blood
1995
Urinary luteinizing hormone (LH) testing has been proposed as a reliable method for the prediction of ovulation but its accuracy has been challenged by some studies. To check how accurately the oscillations of urinary LH reflected the plasma changes, surges of LH of different magnitude and duration were artificially induced in plasma and the hormone was measured simultaneously in urine. Post-menopausal women (n = 16) were stimulated during 1 week with a combination of transdermal oestradiol (400 micrograms) and i.m. progesterone (25 mg on day 4, 50 mg on day 5) to obtain an LH discharge comparable with the pre-ovulatory LH peak. A short and moderate peak of LH was induced by the i.v. inject…
Synergism between exposure to mercury and use of iodine supplements on thyroid hormones in pregnant women
2015
Objective: To evaluate the association between mercury exposure and thyroid-stimulating hormone (TSH), total triiodothyronine (TT3) and free thyroxine (FT4) levels during pregnancy as well as to explore if there is any synergic action between mercury and intake of iodine from different sources. Methods: The study population was 1407 pregnant women participating in the Spanish INMA birth cohort study. Total mercury concentrations were analyzed in cord blood. Thyroid hormones (THs) were measured in serum samples collected at 13.2 +/- 1.5 weeks of gestation. The association between mercury and TH levels was evaluated with multivariate linear regression models. Effect modification caused by iod…
Influence of the ovary on parameters of LH secretion during the recovery from buserelin-induced desensitization
1994
Abstract This study examined the effect of the ovary on LH pulsatility and on the secretory performance of gonadotrophes during the phase of recovery after treatment with buserelin, a GnRH analogue. We included 12 patients, who received buserelin (1.2 mg/day, intranasally for 3 months) as a reductive therapy for uterine leiomyomatosis prior to hysterectomy. Six patients were oophorectomized and the other 6 patients had their ovaries preserved. LH was measured in samples taken basally up to 36 days after suppression of buserelin. LH pulsatility was studied on day 9 along a 24-h cycle, and the response of the hormone to a double-stimulus GnRH test on days 0, 9, 20, and 34. The concentration o…
Mepanipyrim haptens and antibodies with nanomolar affinity
2013
Mepanipyrim is an anilinopyrimidine fungicide used worldwide for crop protection. With the aim of developing useful immunoreagents for mepanipyrim immunoanalysis, two new functionalized derivatives were prepared and antibodies were generated. Affinity and specificity were assessed by direct and indirect competitive ELISA using homologous and heterologous conjugates. Although all antibodies were selective for the target analyte, the immunizing hapten structure was revealed as a determinant for high-affinity antibody production (IC50 = 3 nM). © 2013 The Royal Society of Chemistry.

Sub- and supercritical fluid extraction of trichloropyridinol from soil prior to immunoassay
1997
A comparative study on the extraction of TCP (3,5,6-trichloro-2-pyridinol, a metabolite of chlorpyrifos) from soil with CO2 and H2O is reported. The polarity of the analyte requires the presence of both a cosolvent (methanol) and an ion-pair reagent [(1R)-(-)-10-camphorsulfonic acid ammonium salt] for 95% extraction in 30 min when supercritical CO2 at 40 degrees C and 383 bar is used as extractant. Subcritical water (250 degrees C and 200 bar) enables complete extraction within 15 min without additives. Quantitation of the target analyte is performed by specific immunoassay using a non-commercial monoclonal antibody which provides a linear determination range between 0.005 and 5 micrograms/…
Exploring alternative hapten tethering sites for high-affinity anti-picoxystrobin antibody generation
2011
The relevance of the linker tethering site in haptens was investigated for antibody generation and immunoassay development. Three derivatives of the strobilurin fungicide picoxystrobin were synthesized with the same functionalized spacer arm located at three different positions. Protein conjugates of those haptens were employed as immunogens, and novel polyclonal antibodies were produced and characterized. All haptens afforded highly specific antibodies, but different affinities to the free analyte were observed among the obtained antisera. Next, competitive enzyme-linked immunosorbent assays were studied in several formats, and site heterology was confirmed as an effective strategy for det…
Immuno-SLM—a combined sample handling and analytical technique
2004
Immuno-supported liquid membrane (immuno-SLM) extraction is a new technique that makes use of antibody (Ab)-antigen interactions as the "extraction force" to drive the mass transfer in a selective way. In immuno-SLM, anti-analyte (Ag) Abs are introduced into the acceptor phase of the SLM unit to trap the Ag that passes from the flowing donor through the SLM into the stagnant acceptor. The amount of immuno-extracted analyte (AbAg) is quantified by connecting the immuno-SLM unit on-line with a non-competitive heterogeneous fluorescence flow immunoassay (FFIA) that makes use of a fluorescein-labeled analyte tracer that titrates the residual excess of Ab present in the acceptor. A restricted ac…
