On the use of mixing reactors in flow injection analysis with thermal lens calorimetric detection

1993

Abstract In flow injection analysis with thermal lens calorimetric detection, mixing of reagent streams of different composition before the detection cell produces a large background noise. The noise is due to the heterogeneity of the mixture which produces refractive changes in the probe beam direction. Procedures to measure the homogeneity of the mixture of solutions are derived, and used to evaluate and improve the mixing efficiencies of packed bead reactors. Using well-designed reactors, the mixing noise can be reduced below the noise levels due to other sources.

Slurries Introduction in Flow Injection Atomic Absorption Spectroscopic Analysis of Sewage Sludges

1990

Abstract The direct introduction of slurries in flow Injection-flame atomic absorption spectroscopy has been studied using a single-channel manifold. The influence of flow Injection parameters on the sensitivity and accuracy of this procedure has been established. A method has been developed for the determination of copper, manganese, and lead In sewage sludge where batch digestion of the samples In a microwave oven is carried out before dilution. Both sensitivity and limit of detection of the flow Injection analysis procedure are adequate for the analysis of actual samples (the limit of detection was 0.06, 0.05, and 0.3 ppm for copper, manganese, and lead, respectively). The accuracy of th…

A Novel Approach to Propagation Pattern Analysis in Intracardiac Atrial Fibrillation Signals

2010

The purpose of this study is to investigate propagation patterns in intracardiac signals recorded during atrial fibrillation (AF) using an approach based on partial directed coherence (PDC), which evaluates directional coupling between multiple signals in the frequency domain. The PDC is evaluated at the dominant frequency of AF signals and tested for significance using a surrogate data procedure specifically designed to assess causality. For significantly coupled sites, the approach allows also to estimate the delay in propagation. The methods potential is illustrated with two simulation scenarios based on a detailed ionic model of the human atrial myocyte as well as with real data recordi…

DNA delivery to 'ex vivo' human liver segments.

2011

Hydrodynamic injection is an efficient procedure for liver gene therapy in rodents but with limited efficacy in large animals, using an 'in vivo' adapted regional hydrodynamic gene delivery system. We study the ability of this procedure to mediate gene delivery in human liver segments obtained by surgical resection. Watertight liver segments were retrogradely injected from hepatic vein with a saline solution containing a plasmid bearing the enhanced green fluorescent protein (eGFP) gene, under different conditions of flow rate (1, 10 and 20 ml s(-1)) and final perfused volume. Samples were cultured for 1 to 2 days and used for microscopy and molecular analysis of gene expression. The fluore…

Transgene detection by digital droplet PCR

2014

Somatic gene therapy is a promising tool for the treatment of severe diseases. Because of its abuse potential for performance enhancement in sports, the World Anti-Doping Agency (WADA) included the term 'gene doping' in the official list of banned substances and methods in 2004. Several nested PCR or qPCR-based strategies have been proposed that aim at detecting long-term presence of transgene in blood, but these strategies are hampered by technical limitations. We developed a digital droplet PCR (ddPCR) protocol for Insulin-Like Growth Factor 1 (IGF1) detection and demonstrated its applicability monitoring 6 mice injected into skeletal muscle with AAV9-IGF1 elements and 2 controls over a 3…

Assisted reproductive technology results: Why are live-birth percentages so low?

2014

The present bioessay aims to analyze the impact of parental age, cause of infertility, embryo chromosomal anomalies, assisted reproduction technology (ART) treatments, and environmental and occupational exposures to xenobiotics on ART results, particularly on live-birth percentages per transfer. Special attention is paid to analyzing the effects of these factors on the mitochondrial, genetic, and epigenetic traits of gametes and embryos to ascertain the molecular/cellular mechanisms responsible for the relatively low percentages of live births reported year after year in ART cycles. The bias of age distribution of women attending fertility clinics toward the late thirties and beyond and the…

Lysine synthesis control in Corynebacterium glutamicum RC 115 in mixed substrate (glucose-acetate) medium.

2003

The effect of acetate as a glucose co-substrate on growth, lysine synthesis and experimental lysine yield from carbon substrates by Corynebacterium glutamicum RC 115 was investigated. It was found that low amounts of acetate, injected with a glucose-acetate pulse into the steady-state continuous culture in bioreactor, caused a slight decrease in the specific rates of glucose uptake and bacterial growth, but a significant increase in the cell specific rate of lysine synthesis and an increase in lysine yield. In contrast, acetate injected in high amounts was followed by a drastic decrease in the values of these parameters. A strong increase in experimental lysine yield under the latter condit…

The adenine nucleotide content of rat liver during infusions of carbohydrates and polyols

1972

Injection of large doses of fructose, sorbitol, or a mixture of glucose, fructose and xylitol in rats causes a drop of liver ATP, total adenine nucleotides and Pi and a rise of AMP, which is in agreement with data from the literature. These changes are considered as a transient disturbance of homeostasis by compounds which are rapidly phosporylated in the liver. This is confirmed by the fact that during continuous infusion of these and other compounds at doses of 1,5 g · kg−1 · h−1 there was no such change. It is concluded that infusions of fructose or of the other carbohydrates tested with rates not exceeding those recommended for parenteral nutrition (0,5 g · kg−1 · h−1) are not likely to…

Intracytoplasmic Testicular Sperm Injection: An Effective Treatment for Otherwise Intractable Obstructive Azoospermia

1995

AbstractPurpose: We evaluated the efficacy of intracytoplasmic sperm injection with testicular spermatozoa.Materials and Methods: Intracytoplasmic sperm injection was performed with spermatozoa obtained from testicular biopsy specimens in 15 patients with obstructive azoospermia, in whom standard microsurgical procedures were not feasible or had previously failed. Results: Fertilization was achieved in 14 to 15 cycles. Mean fertilization rate per cycle was 63.6 percent. Four clinical pregnancies occurred, for a pregnancy rate of 26.7 percent per started cycle and 28.6 percent per transfer. Conclusions: Intracytoplasmic testicular sperm injection is followed by high fertilization rates, and of…
