The comparative behavior of Lactococcus lactis in free and immobilized culture processes

This study presents the changes in the physiological state of Lactococcus lactis in different actively growing systems. The intracellular pH (pHin), NADH/NAD ratio and lactate dehydrogenase (LDH) activity were measured in free cell batch culture with or without external pH (pHout) control, and in continuous free or immobilized cell reactors. In free cell batch culture it has been shown that the pHin is dependent both on pHout and age of the culture, while in free cell continuous culture the pHin and the pHout are close together. Inorganic polyphosphates (polyP) are linear polymers of orthophosphate (Pi) residues linked by high-energy phosphoanhydride bonds. These polymers are widely distributed in nature, from archaebacteria, eubacteria, fungi, algae, and protozoa to higher plants and animals (for reviews, see Kulaev 1979; Wood and Clark 1988; Kornberg 1994, 1995). PolyP molecules are stable in neutral aqueous solutions, but are hydrolyzed by heat-treatment, and under acidic or alkaline conditions. Abstract Marine sponges as sessile filter feeders are inevitably under a constant influence of changes in their environment. Mediation of extracellular signals and regulation of cellular response to environmental stress is a key function of cellular protein kinases. Expression, proteolytical cleavage and phosphorylation of stress-responsive KRS_SD protein kinase, in control and tributyl-tin (TBT) treated sponges were investigated. In control sponge, two KRS_SD proteins were expressed: KRS_SD1 (54 kDa) corresponding to KRS_SD calculated molecular weight, and KRS_SD2 (50 kDa). Females live longer than males. This could be in part due to the higher levels of estrogens in females, which protect them against aging. Physiological concentrations of estrogens have antioxidant effects as they induce the expression of manganese superoxide dismutase and glutathione peroxidase by stimulating estrogen receptors and the mitogen-activated protein kinase and nuclear factor kappa B pathways. However, estrogens can have undesirable effects such as they are feminizing to males, so other alternatives need to be searched. The distribution of light energy between the two photosystems as well as the light-induced turnover of PSII proteins are regulated by the reversible phosphorylation of LHCII and the PSII-core proteins. Electrophilic substitution and cyclization of 2,2′-bis(N-methylindolyl): A simple access to potential protein kinase C inhibitor

A strategy is described for the synthesis of functionalized and cyclized 2,2′-bisindolyl derivatives related to several basic systems of natural products. The starting 2,2′-bis(N-methylindolyl) (8) reacts with a variety of electrophiles and electrophilic dienophiles to furnish the novel, functionalized and cyclized bisindolyl derivatives 9–16. In addition, some reactivity and structural aspects are discussed; an X-ray crystallographic analysis of the 2,2′-bisindolyl 8 provided valuable information for the conformational analyses.

Cellular metabolism is a tightly controlled process in which the cell adapts fluxes through metabolic pathways in response to changes in nutrient supply. Among the transcription factors that regulate gene expression and thereby cause changes in cellular metabolism is the basic leucine-zipper (bZIP) transcription factor CCAAT/enhancer-binding protein alpha (C/EBPα). Protein lysine acetylation is a key post-translational modification (PTM) that integrates cellular metabolic cues with other physiological processes. Phorbol Esters and Muscarinic Receptor Agonists Activate Phospholipase D in Heart and Brain

Phospholipase D (PLD) hydrolyzes phosphatidylcholine and thereby seems to play a key role in a novel pathway of signal transduction. PLD activity in rat hippocampal slices and atria of rat, guinea pig and chicken hearts was determined by measuring the catalytic products choline (Ch), phosphatidic acid (PA) and, in the presence of a primary alcohol, phosphatidylpropanol or phosphatidylethanol. It was found that the PLD activity was high, even under resting conditions, in both tissues, especially in the hippocampus, and that the enzyme activity could be enhanced by activation of protein kinase C and by muscarinic receptor stimulation.

Dauerinfusionen mit Glycerin an Ratten fuhren bei Dosierungen oberhalb der durchschnittlichen maximalen Umsatzkapazitat (0,74 g.kg−1.h−1) zu Veranderungen an den Nieren in Form einer Gewichtszunahme bei gleichzeitiger Proteinverarmung und eines Aktivitatsverlustes an Glycerokinase bei deren gleichzeitigem Auftreten im Harn. Bei Dosierungen von 0,6 g.kg−1.h−1 und mehr kommt es zu einem ATP-Abfall in den Nieren. Im Gegensatz dazu sind Veranderungen in der Leber uneinheitlicb und in ahnlicher Form nur bei Dosierungsraten oberhalb der maximalen Umsatzkapazitat zu finden.
