Intragenic G-quadruplex structure formed in the human CD133 and its biological and translational relevance.

Cancer stem cells (CSCs) have been identified in several solid malignancies and are now emerging as a plausible target for drug discovery. Beside the questionable existence of CSCs specific markers, the expression of CD133 was reported to be responsible for conferring CSC aggressiveness. Here, we identified two G-rich sequences localized within the introns 3 and 7 of the CD133 gene able to form G-quadruplex (G4) structures, bound and stabilized by small molecules. We further showed that treatment of patient-derived colon CSCs with G4-interacting agents triggers alternative splicing that dramatically impairs the expression of CD133. Targeting of the Peritumoral Adipose Tissue Microenvironment as an Innovative Antitumor Therapeutic Strategy

The tumor microenvironment (TME) plays a key role in promoting and sustaining cancer growth. Adipose tissue (AT), due to its anatomical distribution, is a prevalent component of TME, and contributes to cancer development and progression. Cancer-associated adipocytes (CAAs), reprogrammed by cancer stem cells (CSCs), drive cancer progression by releasing metabolites and inflammatory adipokines. In this review, we highlight the mechanisms underlying the bidirectional crosstalk among CAAs, CSCs, and stromal cells. Moreover, we focus on the recent advances in the therapeutic targeting of adipocyte-released factors as an innovative strategy to counteract cancer progression.

Capillaroscopic and histologic examination of the human labial mucosa: A new non invasive methodology compared with the histological exam

The aim of our study is to compare the capillaroscopic examination in vivo with the histological examination in vitro of labial microcirculation in healthy subjects. Methods. Twenty-nine subjects (19 females, 10 males; mean±SD age 55.61±2.55; range 45-65 years) were examined in our laboratory. The characteristics of labial microcirculation were examined using computerised videomicroscopic techniques and histological exam. Results. The results obtained with both methods showed some analogies: in fact, the differences in capillary diameter were in the range of ±0.3 µ; regarding capillary density as well, a variation of ±2 capillaries per mm2 was found. Development of a microfluidic design for an automatic lab-on-chip operation

Simple and easy to use are the keys for developing lab-on-chip technology. Here, a new microfluidic circuit has been designed for an automatic lab-on-chip operation (ALOCO) device. This chip used capillary forces for controlled and precise manipulation of liquids, which were loaded in sequence from different flowing directions towards the analysis area. Using the ALOCO design, a non-expert user is able to operate the chip by pipetting liquids into suitable inlet reservoirs.
