A novel cholesterol-based detergent

Design, synthesis and characterization of CHAPSTEROL, a novel cholesterol-based detergent developed for functional solubilization of cholesterol-dependent membrane proteins are described. To validate CHAPSTEROL, we employed the oxytocin receptor, a G protein-coupled receptor requiring cholesterol for its high-affinity binding state. Using the photoactivatable cholesterol analogue [3H]6,6-azocholestan-3beta-ol[3alphaH], we demonstrate that solubilization by CHAPSTEROL leads to an enrichment of cholesterol-binding proteins whereas the widely used bile acid derivative CHAPSO leads to a significant depletion of cholesterol-binding proteins. Lipid accumulation and metabolism in polychaete spermatogenesis: Role of the large discoidal lipoprotein

In most oviparous animals, lipoprotein-mediated lipid transport plays an important role in the nutrient supply for the oocyte. In male gametes, lipids are used as energy substrates in spermatozoa but nothing is yet known about their origin and metabolism throughout spermatogenesis. Rafts, or functional domains, are transient nano- or mesoscopic structures in the plasma membrane and are thought to be essential for many cellular processes such as signal transduction, adhesion, trafficking and lipid/protein sorting. Observations of these membrane heterogeneities have proven challenging, as they are thought to be both small and short-lived. Cholesterol is a major constituent of the plasma membrane in eukaryotic cells. It regulates the physical state of the phospholipid bilayer and is crucially involved in the formation of membrane microdomains. Cholesterol also affects the activity of several membrane proteins, and is the precursor for steroid hormones and bile acids. International audience; Addition of long-chain polyunsaturated fatty acids (LC-PUFAs) from marine oil into food products implies preliminary refining procedures of the oil which thermal process affects the integrity of LC-PUFAs. Deodorization, the major step involving high temperatures, is a common process used for the refining of edible fats and oils. The present study evaluates the effect of deodorization temperature on the formation of LC-PUFA geometrical isomers. Biologically Active Triterpene Saponins from Callus Tissue of Polygala amarella

A new bioactive saponin (1), together with a known saponin (polygalasaponin XXVIII) has been isolated from the callus tissue culture of Polygala amarella. Based on spectroscopic data, especially direct and long-range heteronuclear 2D NMR analysis and on chemical transformations, the structure of 1 was elucidated as 3-O-beta-D-glucopyranosyl presenegenin-28-O-beta-D-galactopyranosyl-(1 --> 3)-beta-D-xylopyranosyl-(1 --> 4)-alpha-L-rhamnopyranosyl-(1 --> 2)-[beta-D-glucopyranosyl-(1 --> 3)]-beta-D-fucopyranoside. Both saponins showed significant immunological properties based on the enhancement of granulocyte phagocytosis in vitro.

Impaired cellular cholesterol efflux by oxysterol-enriched high density lipoproteins.

One of the proposed antiatherogenicity role of high-density lipoproteins (HDL) is believed to stimulate removal of cholesterol from the peripheral cells back to the liver for excretion. We have investigated the effects of oxidation-related modifications of HDL on their ability to stimulate cholesterol efflux from cultured cells. Human HDL (HDL3, 1.13 < d < 1.21 g/ml) have been modified either by malondialdehyde or by copper-mediated oxidation (Ox-HDL3). Compared with native HDL3, the modified HDL3 resulted in a significantly reduced efflux of labeled cholesterol from preloaded macrophages (P388D1 cell line). Focused microwave-assisted Soxhlet extraction: An expeditive approach for the isolation of lipids from sausage products

A prototype of extractor based on the conventional Soxhlet principles but assisted in the cartridge zone by focused microwaves is proposed for accelerating the extraction of lipids from sausage products. The extraction process has been optimised using a multivariate design involving the main variables influencing the performance of the prototype (namely, irradiation power P, irradiation time T and number of cycles C). Under the optimum working conditions (P=160 W, T=10 s and C=14), the extraction of lipids from different Spanish sausage products is complete in 45 min. Determination of Anticonvulsant Drugs in Pharmaceutical Preparations by Micellar Liquid Chromatography

A micellar liquid chromatographic method for quality control of pharmaceutical preparations (capsules, pills, tablets, injections, drops, and suppositories) containing the anticonvulsant drugs acetazolamide, carbamacepine, chlordiazepoxide, diazepam, ethosuximide, phenytoin, phenobarbital, and zopiclone has been developed. This methodology involves the use of micellar solutions of cetyltrimethylammonium bromide (CTAB) as mobile phases and UV detection. The proposed approach is rapid and reproducible. Sample preparation only requires dissolution with micellar solvent and adequate dilution with the mobile phase before injection into the chromatographic system.
