Isolation of the DNA minisatellite probe MZ 1.3 and its application to DNA ‘fingerprinting’ analysis

1990

Abstract A minisatellite probe, MZ 1.3, detecting hypervariable fragment patterns was isolated from a human genomic library. A repetitive sequence of 27 bp length was identified which is contained in the probe approx. 40 times. The MZ 1.3 repeat shows variable homology of 53–73% to the repetitive sequence of the protein III gene of the bacteriophage M13 genome. Polymorphic restriction fragment patterns were found with MZ 1.3 using the enzymes Hinf I, BstN I, Hae III, Mbo I, PstI PvuII , and Rsa I. An average of 18 polymorphic fragments was observed using Hinf I as enzyme. The band sharing frequency after Hinf I digestion among unrelated individuals was determined to be 23.8 ± 7.2%. An examp…

Emerging GII.4 norovirus variants affect children with diarrhea in Palermo, Italy in 2006

2008

Although the genetic/antigenic heterogeneity of human noroviruses (NoVs) is impressive, a few genogroup II strains of genotype 4 (GII.4) are dominant worldwide. GII.4 NoVs evolve rapidly and in the last 15 years six epidemic variants have been identified. In 2005-2006, surveillance of sporadic viral gastroenteritis in children in Palermo, Italy, resulted in the detection of NoV strains in 20.9% of the patients admitted to hospital. By restriction fragment length polymorphism (RFLP) and sequence analysis of region A in the RNA-dependent RNA-polymerase (RdRp) gene, 59 NoV strains were successfully characterized. Eighty-one percent of the strains were characterized as GII.4, 14% as GIIb/Hilver…

Second-order approximations to laser-assisted charged-particle scattering.

1990

Multiphoton free-free transitions within a full second-order treatment of the scattering potential are considered with two specific aims: (i) to test some widely used approximations against exact calculations; (ii) to present differential cross sections for the laser-assisted positron scattering, in the regime of intermediate field intensities (up to ${10}^{13}$ W/${\mathrm{cm}}^{2}$). In the case of a field linearly polarized along the incident particle momentum, the results clearly discriminate among the approximations, setting interesting limitations to each of them as functions of the scattering potential features and of the field parameters. Each approach shows good performance for par…

Multicore optical fibres for astrophotonics

2011

We report progress towards multimode (MM) fibre filters for suppressing the OH emission that hinders ground-based observation of the early Universe. Fibre Bragg gratings (FBGs) can filter these narrow spectral lines in single-mode (SM) fibres [1]. Implementing them in MM fibres well-matched to astronomical instruments requires transitions between the MM fibre and several SM fibres [2]. Such hand-crafted “photonic lanterns” require many identical FBGs to be made and spliced in place. Instead we are pursuing the idea in multicore (MC) fibres, Fig. 1(a). The FBG is written at once in all the SM cores. The fibre is jacketed with low-index glass and tapered to form the core and cladding of a MM …

Progressive active mobilization with dose control and training load in critically ill patients (PROMOB): Protocol for a randomized controlled trial

2020

The dose of progressive active mobilization is still uncertain. The purpose of this study is to identify if the addition of a protocol of progressive active mobilization with dose and training load control to usual care is effective in reducing the length of stay in intensive care unit (ICU) and the improvement of the functioning, incidence of ICU-acquired weakness (ICUAW), mechanical ventilation duration and mortality rate in patients hospitalized in ICU. It is Double-blind randomised clinical trial. The setting for this trial will be medical and surgical ICU of a university hospital. The study participants will be 118 patients aged> 18 years admitted to ICU for less than 72 hours. Partici…

Muscle-Tendon Interaction During Human Dolphin-Kick Swimming

2019

Without high impact forces, it is not clear how humans can utilize tendon elasticity during low-impact activities. The purpose of the present study was to examine the muscle-tendon behavior together with the electromyographic (EMG) activities of the vastus lateralis (VL) muscle during the human dolphin-kicking. In a swimming pool, each subject (n = 11) swam the 25 m dolphin-kicking at two different speeds (NORMAL and FAST). Surface EMGs were recorded from the VL and biceps femoris (BF) muscles. Simultaneous recordings of the knee joint angle by electro-goniometer and of the VL fascicle length by ultrasonography were used to calculate the muscle-tendon unit and tendinous length of VL (LMTU a…

Dépendance de la fatigue à la longueur musculaire : approche mécanique et neurophysiologique

2006

The objective of this work is to evaluate the fatigue dependency on the knee extensor muscles length. Exercises varying the type of contraction (isometric and eccentric) and intensity of contraction (maximal and sub-maximal) are performed in order to disrupt the neuromuscular system by different factors of fatigue: central and peripheral. These exercises are performed at two muscular lengths (S and L, corresponding respectively to 40 and 100° of knee flexion). Neuromuscular parameters (maximal torque, muscular twitch, voluntary activation level, electromyographic activity, etc.) are analyzed in pre- and post-fatigue conditions in the type of contraction required by the fatiguing exercise. I…

Polyphasic identification of yeasts isolated from bark of cork oak during the manufacturing process of cork stoppers.

2003

A two-step protocol was used for the identification of 52 yeasts isolated from bark of cork oak at initial stages of the manufacturing process of cork stoppers. The first step in the identification was the separation of the isolates into groups by their physiological properties and RFLPs of the ITS-5.8S rRNA gene. The second step was the sequencing of the D1/D2 domains of the 26S rRNA gene of selected isolates representing the different groups. The results revealed a predominance of basidiomycetous yeasts (11 species), while only two species represented the ascomycetous yeasts. Among the basidiomycetous yeasts, members representing the species Rhodosporidium kratochvilovae and Rhodotorula n…
