Limits of multi-linear gradient optimisation in reversed-phase liquid chromatography

Abstract The concept of limiting peak purity was applied to quantify the degree of completion of the separation capability of a chromatographic system using multi-linear gradients. The objective was to check whether the complexity of a gradient program deserves be increased to enhance resolution by inserting more linear segments, or on the contrary, no significant improvements can be expected under more complex gradients. A method to classify extra virgin olive oils (EVOOs) according to their genetic variety using sterol profiles obtained by high-performance liquid chromatography (HPLC) with mass spectrometry (MS) detection has been developed. Sterol extracts were chromatographed on a dC18 Atlantis column (100x3 mm, 3 microm) with a gradient of acetonitrile/water (0.01% acetic acid) at a flow rate of 1.0 mL min(-1) and positive-ion mode MS detection. Resolution of inherently chiral resorcarene derivatives by enantioselective HPLC

Reversed-phase high-performance liquid chromatography of proteins and polypeptides on polystyrene-coated silica supports

Abstract A new type of hydrolytically stable reversed-phase packing material prepared by multi-point covalent binding of polystyrene chains onto the surface of porous silica was examined in the high-performance liquid chromatography of proteins and polypeptides. Whereas wide-pore material was shown to give a rapid and efficient resolution of proteins, packings with smaller pores provided better selectivities for peptides.

Determination of benperidol and its reduced metabolite in human plasma by high-performance liquid chromatography and electrochemical detection.

An isocratic high-performance liquid chromatographic method with electrochemical detection for the quantification of benperidol and its suggested reduced metabolite TVX Q 5402 in human plasma is described. The method included a two-step solid-phase extraction on reversed-phase and cation-exchange material, followed by separation on a cyanopropyl silica gel column (5 microns; 250 mm x 4.6 mm I.D.). The eluent was 0.15 M acetate buffer (pH 4.7) containing 25% acetonitrile (w/w). Spiperone served as internal standard. Native mesoporous silica beads were subjected to a sequence of post-treatment procedure including hydrochloric acid treatment, calcination and subsequent rehydroxylation. The post-treated silica beads were converted into RP-18 silica by silanization with monochloro- and dimethoxy-n-octadecylsilanes, respectively. The influence of post-treatments and silanization conditions on the physico-chemical characteristics and on the chromatographic behaviour of the RP-silicas was studied. Also the changes of the pore structural parameters and the silanol group densities during the post-treatment and silanization were assessed.

Sweeteners determination in table top formulations using FT-Raman spectrometry and chemometric analysis

Abstract A partial least squares (PLS) Fourier transform Raman spectrometry procedure based on the measurement of solid samples contained inside standard glass vials, has been developed for direct and reagent-free determination of sodium saccharin and sodium cyclamate in table top sweeteners. A classical 22 design for standards was used for calibration, but this system provides accuracy errors higher than 13% w/w for the analysis of samples containing glucose monohydrate. Quantitation of hydrophobicity in micellar liquid chromatography

Abstract Micellar liquid chromatography (MLC) is shown to be a promising technique for measuring the hydrophobicity of solutes. The presence of micelles has a profound effect on the chromatographic characteristics of reversed-phase columns. The linear relationships between the logarithm, log k , of the retention factor and such diverse properties as the number of carbon atoms in homologous series, octanol–water partition coefficients and solvatochromic parameters, which are observed in conventional reversed-phase liquid chromatography (RPLC), are not usually valid in MLC.
