Experimental studies on the detachment of multi-walled carbon nanotubes by a mobile liquid interface

2017

International audience; Retention and detachment of colloidal particles from surfaces is often considered only in terms of spontaneous chemical dispersion when the surface is already fully submerged. Nevertheless, interfacial processes, where the particles are caught on a mobile liquid contact line by capillary effects are ubiquitous. Theoretical description of such interfacial processes exist for spherical microcolloids, while for anisotropic shapes the literature is limited. Arc-discharge synthesized multiwalled carbon nanotube (MWNT) material contains besides the very anisotropic tubes also irregular amorphous carbon particles (ACP) that both are strongly hydrophobic. As a water–air–soli…

Importance of denitrifiers lacking the genes encoding the nitrous oxide reductase for N2O emissions from soil

2010

Analyses of the complete genomes of sequenced denitrifying bacteria revealed that approximately 1/3 have a truncated denitrification pathway, lacking the nosZ gene encoding the nitrous oxide reductase. We investigated whether the number of denitrifiers lacking the genetic ability to synthesize the nitrous oxide reductase in soils is important for the proportion of N2O emitted by denitrification. Serial dilutions of the denitrifying strain Agrobacterium tumefaciens C58 lacking the nosZ gene were inoculated into three different soils to modify the proportion of denitrifiers having the nitrous oxide reductase genes. The potential denitrification and N2O emissions increased when the size of ino…

Horizontal gene transfer of atrazine-degrading genes (atz) from Agrobacterium tumefaciens St96-4 pADP1::Tn5 to bacteria of maize-cultivated soil

2005

International audience; The plasmid pADP1::Tn5 derived from pADP1[Atr(+)] carrying a TnS transposon conferring kanamycin and streptomycin resistances was constructed and introduced in Agrobacterium tumefaciens St96-4. This genetically modified strain was inoculated (similar to 108 cfu g(-1)) in potted soils planted with maize and treated or not with atrazine (1.5 mg kg(-1)). Bulk and maize rhizosphere soils were sampled 39 days after planting to look for soil indigenous bacteria that had acquired pADP1::Tn5. Four transconjugants were isolated from four different soil samples. The estimated transfer frequency of pADP1::Tn5 was 10(-4) per donor. Maize rhizosphere and atrazine treatment had no…

Genetic transfer of the mcd gene in soil.

2003

Aims: To investigate the role of horizontal gene transfer of mcd (methylcarbamate-degrading) gene in high genetic diversity of carbofuran-degrading bacteria. Methods and Results: The actuality of genetic transfer from degraders to an Agrobacterium tumefaciens strain was determined in liquid medium. The mcd gene was chosen for transfer experiments. Transconjugants were obtained irrespective of the type of the donor strain (Gram-positive or Gram-negative), size of the inoculum, or nature and concentration of the pesticide in the medium. Soil microcosms, inoculated with or without the donor and/or recipient strains were used. The size of the initial degrading population (treated or untreated s…

A High-Resolution Penalization Method for large Mach number Flows in the presence of Obstacles

2009

International audience; A penalization method is applied to model the interaction of large Mach number compressible flows with obstacles. A supplementary term is added to the compressible Navier-Stokes system, seeking to simulate the effect of the Brinkman-penalization technique used in incompressible flow simulations including obstacles. We present a computational study comparing numerical results obtained with this method to theoretical results and to simulations with Fluent software. Our work indicates that this technique can be very promising in applications to complex flows.

Misdiagnosis of familial Mediterranean fever in patients with Anderson-Fabry disease

2012

Fabry disease (FD) is an underdiagnosed pathology due to its symptomatology that overlaps with various systemic and rheumatic disorders, including familial Mediterranean fever (FMF). We examined the Mediterranean fever (MEFV) and α-galactosidase A (GLA) genes, whose mutations are responsible for FMF and FD, respectively, in 42 unrelated patients diagnosed with FMF, which revealed significant ambiguity regarding some of the symptoms which are also present in FD. The objective of this study was to determine the spectrum of mutations present in these genes, in order to identify cases of mistaken diagnosis of FMF and/or missed diagnosis of FD. Ten out of 42 patients had one mutation in homozygo…

Hybrid WENO schemes for polydisperse sedimentation models

2015

International audience; Polydisperse sedimentation models can be described by a strongly coupled system of conservation laws for the concentration of each species of solids. Typical solutions for the sedimentation model considered for batch settling in a column include stationary kinematic shocks separating layers of sediment of different composition. This phenomenon, known as segregation of species, is a specially demanding task for numerical simulation due to the need of accurate numerical simulations. Very high-order accurate solutions can be constructed by incorporating characteristic information, available due to the hyperbolicity analysis made in Donat and Mulet [A secular equation fo…

Development of a green fluorescent tagged strain of Aspergillus carbonarius to monitor fungal colonization in grapes.

2011

An enhanced green fluorescent protein has been used to tag an OTA-producing strain of Aspergillus carbonarius (W04-40) isolated from naturally infected grape berries. Transformation of the fungus was mediated by Agrobacterium tumefaciens. The most efficient transformation occurred when the co-cultivation was done with 104 conidia due to higher frequency of resistance colonies (894 per 104 conidia) and lower background obtained. To confirm the presence of the hph gene in hygromycin resistant colonies, 20 putative transformants were screened by PCR analysis. The hph gene was identified in all the transformants. Variation on the expression levels of the eGFP was detected among the transformant…

Comparative analysis of two paradigm bacteriophytochromes reveals opposite functionalities in two-component signaling

2021

Bacterial phytochrome photoreceptors usually belong to two-component signaling systems which transmit environmental stimuli to a response regulator through a histidine kinase domain. Phytochromes switch between red light-absorbing and far-red light-absorbing states. Despite exhibiting extensive structural responses during this transition, the model bacteriophytochrome from Deinococcus radiodurans (DrBphP) lacks detectable kinase activity. Here, we resolve this long-standing conundrum by comparatively analyzing the interactions and output activities of DrBphP and a bacteriophytochrome from Agrobacterium fabrum (Agp1). Whereas Agp1 acts as a conventional histidine kinase, we identify DrBphP a…

Microfabrication of hybrid fluid membrane for microengines

2015

International audience; This paper describes the microfabrication and dynamic characterization of thick membranes providing a technological solution for microengines. The studied membranes are called hybrid fluid-membrane (HFM) and consist of two thin membranes that encapsulate an incompressible fluid. This work details the microelectromechanical system (MEMS) scalable fabrication and characterization of HFMs. The membranes are composite structures based on Silicon spiral springs embedded in a polymer (RTV silicone). The anodic bonding of multiple stacks of Si/glass structures, the fluid filling and the sealing have been demonstrated. Various HFMs were successfully fabricated and their dyna…
