DETERMINATION OF PENTOBARBITAL IN BIOLOGICAL SAMPLES BY MICELLAR LIQUID CHROMATOGRAPHY

A liquid chromatographic procedure for the determination of pentobarbital in urine and plasma samples is described. The proposed system uses a Spherisorb octadecyl-silane ODS-2 C18 analytical column, a guard column of similar characteristics, and a 0.02 M CTAB-15% 1-propanol at pH 7.5 mobile phase. The UV detector was set at 250 nm. Pentobarbital was isolated from urine and plasma samples by using a single solid phase extraction procedure with LMS cartridges. Mephobarbital was used as internal standard. Limits of detection were 0.53 μg/mL and 0.60 μg/mL in urine and plasma samples respectively. Quality control of pharmaceuticals containing clenbuterol by thermal lens spectrometry.

An ultrasensitive absorptiometric procedure for the determination of clenbuterol in pharmaceutical preparations was developed. Clenbuterol was diazotized with nitrite and coupled with 1-(naphthyl)ethylenediamine, and the absorbance of the azo dye formed was measured by both spectrophotometry and ultrasensitive thermal lens spectrometry (TLS). The TLS limit of detection was 1.5 ppb, 14-fold lower than with a Hewlett-Packard diode array spectrophotometer. Thus, the TLS procedure can be advantageously applied to quality control of clenbuterol at the individual dose level and in small samples. Repeatability as relative standard deviation was 1.5% (50 ppb, n = 6).

A simple and effective procedure is proposed for the study and simultaneous determination of adrenaline and noradrenaline. The fluorimetric determination of both substances is performed in a flow injection assembly and by oxidation of both drugs with the solved molecular oxygen. The influence of different parameters is empirically studied and the interpretation of the reaction mechanism is also added. The determination of adrenaline is monitored at 450 nm and the outputs at 520 nm correspond to the adrenaline and noradrenaline global amount; for both lectures lambda(exc) 329 nm. Quantification of low levels of amorphous content in sucrose by hyperDSC.

A method was developed for the quantification of low levels of amorphous content in sucrose with hyperDSC. The method was based on the fact that the change of specific heat at the glass transition is linearly proportional to the amorphous content. It was found out that as annealing time increased, the glass transition temperature moved to a higher temperature and the change of specific heat increased. DeltaC(p) for annealed totally amorphous sucrose was 0.761+/-0.012 Jg(-1) degrees C(-1). Synthetic mixtures with various proportions of crystalline and amorphous sucrose were prepared. The cyclic voltammograms of levonorgestrel (LNG) in Britton-Robinson buffers of pH 2-11 at the hanging mercury drop electrode showed a single two-electron irreversible cathodic peak over the whole pH range. This peak may be attributed to the reduction of the 3-keto-delta-4-group in the A-ring of the LNG molecule. Encapsulation of Alcohol Dehydrogenase in Mannitol by Spray Drying

The retention of the enzyme activity of alcohol dehydrogenase (ADH) has been studied in various drying processes such as spray drying. The aim of this study is to encapsulate ADH in mannitol, either with or without additive in order to limit the thermal denaturation of the enzyme during the drying process. The retention of ADH activity was investigated at different drying temperatures. When mannitol was used, the encapsulated ADH was found inactive in all the dried powders. This is presumably due to the quick crystallization of mannitol during spray drying that resulted in the impairment of enzyme protection ability in comparison to its amorphous form.
