Performance of polyester-based electrospun scaffolds under in vitro hydrolytic conditions: From short-term to long-term applications
2019
The evaluation of the performance of polyesters under in vitro physiologic conditions is essential to design scaffolds with an adequate lifespan for a given application. In this line, the degradation-durability patterns of poly(lactide-co-glycolide) (PLGA), polydioxanone (PDO), polycaprolactone (PCL) and polyhydroxybutyrate (PHB) scaffolds were monitored and compared giving, as a result, a basis for the specific design of scaffolds from short-term to long-term applications. For this purpose, they were immersed in ultra-pure water and phosphate buffer solution (PBS) at 37 °
Water absorption and hydrothermal performance of PHBV/sisal biocomposites
2014
[EN] The performance of biocomposites of poly(hydroxybutyrate-co-valerate) (PHBV) and sisal fibre subjected to hydrothermal tests at different temperatures above the glass transition of PHBV (TH ¿ 26, 36 and 46 C) was evaluated in this study. The influences of both the fibre content and presence of coupling agent were focused. The water absorption capability and water diffusion rate were considered for a statistical factorial analysis. Afterwards, the physico-chemical properties of water-saturated biocomposites were assessed by Fourier-Transform Infrared Analysis, Size Exclusion Chromatography, Differential Scanning Calorimetry and Scanning Electron Microscopy. It was found that the water d…
Thermal and thermo-oxidative stability and kinetics of decomposition of PHBV/sisal composites
2017
The decomposition behaviours of composites made of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) and sisal were assessed in terms of thermal stability and decomposition kinetics, under inert and oxidative conditions, by means of multi-rate linear non-isothermal thermogravimetric experiments. A statistical design of experiments was applied to study the influence of the addition of sisal (0-10-20-30%wt), the presence coupling agent (Yes/No) and the applied conditions of work (inert or oxidative). An improvement of the thermal and thermo-oxidative stability of PHBV with the addition of sisal was observed for all cases. An accurate methodology based on iso-conversional methods was applied…
Effects of indole-3-acetic acid on Sinorhizobium meliloti survival and on symbiotic nitrogen fixation and stem dry weight production
2009
We evaluated the effects of the main auxin phytohormone, indole-3-acetic acid (IAA), on the central metabolism of Sinorhizobium meliloti strain 1021. We either treated the Sinorhizobium meliloti 1021 strain with 0.5 mM IAA (1021+) or use a derivative, RD64, of the same strain harbouring a pathway for IAA biosynthesis converting tryptophan into IAA via indoleacetamide. We assayed the activity of key enzymes in the major energy-yielding pathways (Entner-Doudoroff, Embden-Meyerhof-Parnas, pentose phosphate, glyoxylate bypass and tricarboxylic acid cycle). We found that activity of two main regulative tricarboxylic acid (TCA) cycle enzymes was increased. Citrate synthase (CS) activity, as compa…
Influence of substrate and temperature on the biodegradation of polyester-based materials: Polylactide and poly(3-hydroxybutyrate-co-3-hydroxyhexanoa…
2020
[EN] The extended use of polymers from renewable resources such as aliphatic polyesters or polyhydroxyalkanoates boosted the necessity to understand their behaviour in an end-of-life scenario. Although they can be degraded in reasonable shorter times than traditional polymers, understanding the degradation mechanisms under dissimilar conditions will contribute to further developments in this field. This work aimed to study the effect of temperature and substrate in the degradation of polylactide (PLA) and poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHBH) in a simulated laboratory scale to ascertain their contribution, separately or in combination. For this purpose, nine parallel degradat…
Neuronal receptors display cytoskeleton-independent directed motion on the plasma membrane
2018
Summary Directed transport of transmembrane proteins is generally believed to occur via intracellular transport vesicles. However, using single-particle tracking in rat hippocampal neurons with a pH-sensitive quantum dot probe that specifically reports surface movement of receptors, we have identified a subpopulation of neuronal EphB2 receptors that exhibit directed motion between synapses within the plasma membrane itself. This receptor movement occurs independently of the cytoskeleton but is dependent on cholesterol and is regulated by neuronal activity.
Poly(HydroxyButyrate-co-HydroxyValerate) (PHBHV) Nanocarriers for Silymarin Release as Adjuvant Therapy in Colo-rectal Cancer
2017
The aim of this study was to address one of the major challenges of the actual era of nanomedicine namely, the bioavailability of poorly water soluble drugs such as Silymarin. We developed new, biodegradable, and biocompatible nanosized shuttles for Silymarin targeted delivery in colon-cancer cells. The design of these 100 nm sized carrier nanoparticles was based on natural polymers and their biological properties such as cellular uptake potential, cytotoxicity and 3D penetrability were tested using a colon cancer cell line (HT-29) as the in vitro culture model. Comparative scanning electron microscopy (SEM) and atomic force microscopy (AFM) measurements demonstrated that the Silymarin load…
Draft genome sequence of Thalassobius mediterraneus CECT 5383T, a poly-beta-hydroxybutyrate producer
2016
Thalassobius mediterraneus is the type species of the genus Thalassobius and a member of the Roseobacter clade, an abundant representative of marine bacteria. T. mediterraneus XSM19T (=CECT 5383T) was isolated from the Western Mediterranean coast near Valencia (Spain) in 1989. We present here the draft genome sequence and annotation of this strain (ENA/DDBJ/NCBI accession number CYSF00000000), which is comprised of 3,431,658 bp distributed in 19 contigs and encodes 10 rRNA genes, 51 tRNA genes and 3276 protein coding genes. Relevant findings are commented, including the complete set of genes required for poly-beta-hydroxybutyrate (PHB) synthesis and genes related to degradation of aromatic …
