This paper shows the ease of application and usefulness of mid-IR measurements for the investigation of orthogonal cell states on the example of the analysis of Pichia pastoris cells. Killer toxins in new isolates of the yeasts Hanseniaspora uvarum and Pichia kluyveri

From various habitats (plant material, fruits, soil), yeasts belonging to the species of Pichia kluyveri and Hanseniaspora uvarum were isolated that showed killer activity. According to the activity spectrum against other yeasts these strains belonged to 11 different groups that were distinguishable from the killer strains K1-K10. The isoelectric points of the killer proteins were in the range of pH 3.5–3.9, the activity optimum was observed at pH 4.2–4.6. Above pH 5 and above a temperature of 25–35°C the killer proteins were inactivated.

Validation of a predictive model for the growth of chalk yeasts on bread.

The present study focused on the effects of temperature, T, and water activity, aw, on the growth of Hyphopichia burtonii, Pichia anomala, and Saccharomycopsis fibuligera on Sabouraud Agar Medium. Cardinal values were estimated by means of cardinal models with inflection. All the yeasts were xerophilic, and they exhibited growth at 0.85 aw. The combined effects of T, aw, and pH on the growth of these species were described by the gamma-concept and validated on bread in the range of 15-25 °C, 0.91-0.97 aw, and pH 4.6-6.8. Exploring the yeast biodiversity of green table olive industrial fermentations for technological applications

In recent years, there has been an increasing interest in identifying and characterizing the yeast populations associated with diverse types of table olive elaborations because of the many desirable technological properties of these microorganisms. In this work, a total of 199 yeast isolates were directly obtained from industrial green table olive fermentations and genetically identified by means of a RFLP analysis of the 5.8S-ITS region and sequencing of the D1/D2 domains of the 26S rDNA gene. Rational selection of non-Saccharomyces wine yeasts for mixed starters based on ester formation and enological traits

Thirty-eight yeast strains belonging to the genera Candida, Hanseniaspora, Pichia, Torulaspora and Zygosaccharomyces were screened for ester formation on synthetic microbiological medium. The genera Hanseniaspora and Pichia stood out as the best acetate ester producers. Based on the ester profile Hanseniaspora guilliermondii 11027 and 11102, Hanseniaspora osmophila 1471 and Pichia membranifaciens 10113 and 10550 were selected for further characterization of enological traits. Summary (1) The aerobic assimilation of malic acid is not a character of certain yeast genera or species as was shown by testing more than 300 different strains. Single strains of the following-species were found to grow on malic acid as the only carbon source: Candida pulcherrima, C. utilis, C. mycoderma, Torulopsis famata, Pichia membranaefaciens, P. wickerhamii, Hansenula capsulata, Trigonopsis variabilis , and Zygosaccharomyces chevalieri . Melanokortīna otrā tipa receptora un tā palīgproteīna ekspresija rauga Pichia Pastoris ekspresijas sistēmā

Darba ietvaros tika izveidots pPIC9K- MC2R-T4L ekspresijas konstrukts, kas tika ekspresēts P.Pastoris sistēmā. Western blot analīze uzrādīja nepieciešamās molekulmasas zonu. MC2R-T4L izdevās attīrīt uz His- un Ha-tag afinitātes kolonnām. Lai noteiktu funkcionāla MC2R struktūru, to nepiecieciešams ekspresēt kompleksā ar MRAP, tādēļ tika izveidoti vairāki MC2-MRAP konstrukti, ko tālāk izmantot proteīna ekspresijai P. Pastoris šūnās. To aktivitāte tika pārbaudīta zīdītāju šūnu līnijās nosakot cAMP daudzuma izmaiņas pēc stimulācijas ar ACTH (1-24).
