A simple protocol for extraction and detection of pyrazines produced by fungi in solid state fermentation

A protocol for the extraction and detection of pyrazines produced by fungi in Solid State Fermentation that gave good recovery of these compounds was devised together with specific techniques for detection by Gas Chromatography (GC). The protocol involved solid-liquid and liquid-liquid extractions, concentration of the extracts and GC analysis. Extraction yields were measured from a model system in which synthetic pyrazines were added. Extracts were analysed with a gas chromatograph equipped with a thermo-ionic detector to allow specific pyrazine detection.

New method to determine the true transmissibilities and permeabilities of oxygen in hydrogel membranes

In the present paper, an electrochemical method to obtain the true transmissibilities and permeabilities of hydrogels to oxygen is proposed. The method involves the measurement of the electric current arising from the flow of oxygen through a potentiostat cell integrated by a top water layer, the hydrogel membrane, and a thin water layer between the membrane and the cathode, where the oxygen is reduced. Varying the thickness of the top water layer, the true permeation characteristics of the hydrogels are obtained. A study of size exclusion and enrichment of proteins employing strong cation-exchange diol silica restricted access material (SCX-RAM) under saturation conditions is presented. Experiments were carried out with bacitracin, protamine, ribonuclease, lysozyme and bovine serum albumin as individual proteinaceous analytes as well as comprehensive binary mixtures and with human urine samples. Protein size dependent capacity features of the SCX-RAM column was observed. Bacitracin demonstrated the highest capacity followed by protamine while adsorption capacities of both ribonuclease and lysozyme were found smaller by a factor of 10. Preferential solvation of lysozyme and bovine serum albumin in copper salt solutions. A quantitative chromatographic study

Preferential solvation λ parameters for systems containing water-copper salt-protein (lysozyme or bovine serum albumin) have been determined by gel permeation chromatography. When water is preferentially adsorbed by the protein, good agreement is found between λ values determined by this method and by equilibrium dialysis-differential refractometry. The influence of the concentration and type of anion component of the copper salt, protein concentration and temperature has been investigated. The methodology used also allows direct visualization of the metal ion bound to the protein and to determine binding parameters. Adsorption of proteins on porous and non-porous poly(ethyleneimine) and tentacle-type anion exchangers

Abstract Adsorption isotherms of proteins [bovine serum albumin (BSA), soybean trypsin inhibitor and alcohol dehydrogenase] on anion exchangers were measured by on-line and off-line methods. The poly(ethyleneimine) (PEI) type and the tentacle-type materials exhibited principally different modes of adsorption. On thin layers of PEI, bonded to non-porous silica, BSA adsorption data corresponded to a monolayer of molecules, with 80% adsorbed side-on, with a high affinity constant for binding, and 20% adsorbed more weakly. Abstract A method has been developed for the fast spectrophotometric determination of propoxur, carbaryl, and ethiofencarb in water samples using injection analysis in the stopped-flow mode. The method is based on the reaction between p -aminophenol and the phenolic compounds obtained from the pesticides, after a previous hydrolysis with 0.05 M NaOH at room temperature for 15 min. Objective: The aim was to analyze effects of a percutaneous neuromodulation (PNM) treatment on the radial nerve, regarding pain, functionality, electrophysiologic excitability, and morphology, in patients with chronic lateral epicondylalgia (LE). Methods: Twenty-four patients with chronic unilateral elbow pain were recruited for this preliminary study and were divided into two groups: control (n = 12) and PNM group (n = 12). The production and the chronobiology of emergence of the cercariae of Euparyphium albuferensis from Gyraulus chinensis experimentally infected with a single miracidium were established during 28 consecutive days from the first day of cercarial shedding. Moreover, the effect of a sudden change in light-dark cycling was investigated. Although the daily cercarial shedding rates show great variability, a progressive increase in cercarial production was observed in the first weeks of the cercarial shedding periods, probably in relation to the demography of intramollusean larval stages.
