The generalized H-point standard-additions method to determine analytes present in two different chemical forms in unknown matrix samples. Part II. C…

2000

The generalized H-point standard-additions method (GHPSAM) is used in order to obtain the total Cr(VI) and chromate concentration in water samples whose matrices are completely unknown. Moreover, a new methodology, which is a modification of the GHPSAM, is proposed for the simultaneous determination of the two major chemical forms of Cr(VI) present in the sample. The method is based on the location of spectral intervals where the behaviour of the interferent absorbance can be considered as linear. From these intervals, the analyte concentration free from bias error can be estimated. Spiked samples of dig and harbour water measured in the UV–visible spectral region have been tested to check …

Development of immunoaffinity columns for pyraclostrobin extraction from fruit juices and analysis by liquid chromatography with UV detection

2010

Abstract Pyraclostrobin belongs to a new generation of fungicides widely used to preserve high valuable crops. In the present study, three monoclonal antibodies with different affinities to this modern strobilurin have been evaluated for their usefulness in the production of immunoaffinity columns suitable for the solid-phase extraction, concentration, and clean-up of residues from food commodities. Different immunosorbents were produced and characterized in terms of antibody immobilization efficiency, immunosorbent binding capacity, optimum elution conditions, and reusability. Covalent coupling of the antibodies to Sepharose–CNBr gel took place with high yield (over 90%), whereas the immun…

High-pressure flow-injection assembly. Indirect determination of glycine by atomic absorption spectrometry.

1991

A procedure for the determination of glycine is described. The method is based on the reaction of the analyte with finely powdered, solid copper(II) carbonate in a continuous-flow assembly. The optimum experimental conditions of pH, temperature, sample volume, flow-rate, column length and internal diameter, and the linear range of calibration, were studied. Interference from foreign substances that accompany this amino acid in pharmaceutical formulations was studied, and the method was applied to the determination of glycine.

Selective determination of trimethylamine in air by liquid chromatography using solid phase extraction cartridges for sampling.

2004

Abstract The selective determination of trimethylamine (TMA) in air by liquid chromatography is reported. Sampling is effected by flushing air through C18-packed solid-phase extraction (SPE) cartridges at a flow rate of 15 mL/min for 15 min. Next, TMA is desorbed from the cartridges and injected into the chromatographic system. The analyte is then selectively retained on a precolumn ( 20 mm ×2.1 mm i.d., packed with 30 μm, Hypersil C18 phase), and derivatized on-line by injecting 9-fluorenylmethyl chloroformate (FMOC). Finally, the TMA-FMOC derivative is transferred to the analytical column ( 125 mm ×4 mm i.d., LiChrospher 100 RP18, 5 μm), and monitored at 262 nm. The method was applied to …

Mepanipyrim haptens and antibodies with nanomolar affinity

2013

Mepanipyrim is an anilinopyrimidine fungicide used worldwide for crop protection. With the aim of developing useful immunoreagents for mepanipyrim immunoanalysis, two new functionalized derivatives were prepared and antibodies were generated. Affinity and specificity were assessed by direct and indirect competitive ELISA using homologous and heterologous conjugates. Although all antibodies were selective for the target analyte, the immunizing hapten structure was revealed as a determinant for high-affinity antibody production (IC50 = 3 nM). © 2013 The Royal Society of Chemistry.

Fully Automated Electric-Field-Driven Liquid Phase Microextraction System with Renewable Organic Membrane As a Front End to High Performance Liquid C…

2019

This article reports for the first time a programmable-flow-based mesofluidic platform that accommodates electric-field-driven liquid phase microextraction (μ-EME) in a fully automated mode. The miniaturized system is composed of a computer-controlled microsyringe pump and a multiposition rotary valve for handling aqueous and organic solutions at a low microliter volume and acts as a front-end to online liquid chromatographic separation. The organic membrane is automatically renewed and disposed of in every analytical cycle, thus minimizing analyte carry-over effects while avoiding analyst intervention. The proof-of-concept applicability of the automated mesofluidic device is demonstrated b…

Application of solid-phase microextraction combined with derivatization to the determination of amphetamines by liquid chromatography

2004

This work evaluates the utility of solid-phase microextraction (SPME) in the analysis of amphetamines by liquid chromatography (LC) after chemical derivatization of the analytes. Two approaches have been tested and compared, SPME followed by on-fiber derivatization of the extracted amphetamines, and solution derivatization followed by SPME of the derivatives formed. Both methods have been applied to measure amphetamine (AP), methamphetamine (MA), and 3,4-methylenedioxymethamphetamine (MDMA), using the fluorogenic reagent 9-fluorenylmethyl chloroformate (FMOC) and carbowax-templated resin (CW-TR)-coated fibers. Data on the application of the proposed methods for the analysis of different kin…

Tuning the selectivity of molecularly imprinted polymer extraction of arylcyclohexylamines: From class-selective to specific

2020

Abstract A molecularly imprinted polymer (MIP) has been prepared in presence of 3-hydroxy phencyclidine (3-OH PCP) as template by bulk polymerization using N,N-dimethylformamide, as porogenic solvent, for the selective solid-phase extraction (SPE) of arylcyclohexylamines from oral fluids. Experimental variables of the extraction procedure have been studied in order to increase both, extraction recovery of 3-OH PCP, used as model analyte, and imprinting factor. By modifying the composition of the washing solvent, the selectivity of the MIP extraction procedure can be tuned, moving from an arylcyclohexylamine selective method to a 3-OH PCP specific method. The applicability of the synthesized…

Determination of trichothecenes and zearalenones in grain cereal, flour and bread by liquid chromatography tandem mass spectrometry

2012

Although analytical methods have been already reported for legislated mycotoxins as trichothecenes and zearalenone (ZON) separately, we describe the optimization of a simple and rapid multimycotoxin method for the determination of a total of 12 mycotoxins simultaneously, nine trichothecenes (NIV, DON, FUS-X, DAS, 15-AcDON, 3-AcDON, NEO, HT-2, T-2 T2), and zearalenone and its metabolites (ZON, a-ZOL, b-ZOL), of different origin (wheat, oat, barley and spelt) and in three different products where these substance can be present (grain, flour and bread) reach the food chain and cause toxic effect either in humans or animals. The extraction procedure was based on a mixture of acetonitrile/water …
