Mepanipyrim haptens and antibodies with nanomolar affinity

Mepanipyrim is an anilinopyrimidine fungicide used worldwide for crop protection. With the aim of developing useful immunoreagents for mepanipyrim immunoanalysis, two new functionalized derivatives were prepared and antibodies were generated. Affinity and specificity were assessed by direct and indirect competitive ELISA using homologous and heterologous conjugates. Although all antibodies were selective for the target analyte, the immunizing hapten structure was revealed as a determinant for high-affinity antibody production (IC50 = 3 nM). © 2013 The Royal Society of Chemistry.

Real-time polarimetric biosensing using macroporous alumina membranes

We report the first demonstration of real-time biosensing in free standing macroporous alumina membranes. The membranes with their 200 nm diameter pores are ideal candidates for biosensing applications where fast response times for small sample volumes are needed as they allow analytes to flow through the pores close to the bioreceptors immobilized on the pores walls. A bulk refractive index sensitivity of 5.2x10 -6 refractive index units was obtained from signal responses to different concentrations of NaCl solutions flowing through the pores. Carbon nanotube-protein carriers enhance size-dependent self-adjuvant antibody response to haptens

Carbon nanotubes (CNTs) are nanomaterials with interesting emerging applications. Their properties make CNTs excellent candidates for use as new nanovehicles in drug delivery, immunization and diagnostics. In the current study, we assessed the immune-response-amplifying properties of CNTs to haptens by using azoxystrobin, the first developed strobilurin fungicide, asamodel analyte. Abstract Hemolytic activity in the hemolymph of Spirographis spallanzanii was estabilished against Bufo, sheep, calf, rabbit, rat, human A, B, O erythrocytes. Chemico-physical treatments suggest that the hemolytic factor could be a thermo-labile protein whose activity requires calcium ions and which is active over broad pH and temperature ranges. Some of these properties and the sigmoidal curve obtained by plotting the quantity of hemolymph against the percentage of hemolysis, produce some analogies with the mammalian lytic system.

Com o objetivo de avaliar pela morfometria o efeito do tenoxicam e do seu diluente no endotélio venoso, foram utilizados 48 coelhos (Oryctolagus cuniculus), brancos, da linhagem Nova Zelândia, machos, com idade acima de 10 semanas, com peso variando entre 2.350 e 3.500 gramas, divididos em dois grupos, denominados Experimento e Controle, que foram observados nos tempos de 6, 12 e 24 horas. A Cryptosporidium parvum sporozoite and oocyst lambda gt11 cDNA library was screened with a hyperimmune rabbit serum that was developed against insoluble fragments of ultrasonicated oocysts. A clone named Cp22.4.1 encoding a protein of 231 amino acids with 4 zinc-finger domains characterized by a Cys-X2-Cys-X4-His-X4-Cys motif was isolated and characterized. There was a complete match between the sequencing data of the coding region of Cp22.4.1 and the corresponding gene at chromosomal level. This study initiates a new method of developing an antigen which might be useful in the prevention of HIV-1 infection. Using a mannan preparation from Saccharomyces cerevisiae neutralizing antiserum was raised in rabbits which prevents HIV-1 infection in vitro up to a titre of 1:128. The corresponding antibody preparation neutralized the in vitro infectivity down to a concentration of 5 micrograms/ml. Analytical studies suggest that the antibodies are directed against the mannose residues of the HIV-1 glycoprotein (gp) 120 and its precursor gp 160.

Cell surface hydrophobicity (CSH) of Aspergillus fumigatus grown both in complex medium (yeast extract/peptone/dextrose; YPD) and minimal (Vogel's N) medium was monitored by assessing attachment of polystyrene microspheres to the cell surface. It was found that mature mycelium was hydrophobic. Treatment of intact mycelium with beta-mercaptoethanol (beta ME) abolished binding of the microspheres to hyphal elements, and coating of the microspheres with beta ME extracts from mycelium inhibited their attachment to intact mycelial cells.
