Colonization of adrenal glands and ovaries of mice by HSV-2 variants

HSV-2 strain ER was shown to consist of variants with different pathogenic phenotype: Variant ER+ replicates to high titers in the adrenal glands and the ovaries but much less in the spleen; the testes were not colonized. ER+ migrates to the spinal ganglia and is highly neuroinvasive after i.p. inoculation. Variant ER- replicates 100-1,000 fold less in the adrenal glands and the ovaries, but proceeds to the spinal ganglia without invading the CNS. However, both variants are highly neuropathogenic after direct i.c. injection. Herpes simplex virus (HSV) has been associated with various neurological disorders. In contrast, HSV infection is very rarely found in acute polyneuroradiculitis. In this report, a patient is described with a severe course of Guillain-Barr¿ syndrome (GBS). HSV IgM-specific antibodies and a rise of complement-fixation antibodies were detected. During the acute phase of neurologic syndrome, a nerve biopsy showed myelin damage and IgM deposits on the inner layer of the perineurium. Plasma exchange, in combination with immunosuppression, was successfully applied as a treatment in the relapsing course of GBS. Finally, after recovery, HSV-specific IgM antibodies disappeared.

Infection of murine hepatocyte cultures by herpes simplex virus (HSV) 1 and 2

A study was undertaken of the interaction between liver cells and Herpes Simplex Virus (HSV) in vitro. Hepatocytes were obtained from HSV-resistant (C57/B16) and from HSV-susceptible (BALB/c, A/J, C3H) mouse strains and cultured according to standard methods. Each culture was infected with several strains of HSV-type 1 or of HSV-type 2, respectively. The multiplicity of infection was 5. The cytopathic effect was evaluated by light- and electron-microscopy. The number of infectious particles was determined using rabbit kidney or Vero cell cultures. Three cases of neonatal herpes simplex virus infection presenting as fulminant hepatitis

We report three cases of neonatal herpes simplex virus (HSV) infection presenting as fulminant hepatitis. None of the patients had clear risk factors for HSV infection and they all died. Antiviral treatment for HSV is currently available but must be administered early in the course of the disease before irreversible liver tissue damage is present. Since the diagnosis may be difficult to establish, we wish to draw the attention of clinicians to the presentation of neonatal HSV infection and suggest that in such cases viral cultures, including culture of liver tissue, should be obtained early and antiviral treatment administered while awaiting the culture results.

The number of TNF-alpha and IL-1 beta producing cells was investigated during the acute replication phase of herpes simplex virus (HSV) in trigeminal ganglia after intranasal infection with strains of different virulence. The highly virulent strain WAL replicated strongly and induced many cytokine producing cells early in the ganglia. The low virulent strain HFEM replicated less, only few cytokine producing cells were detected late. Although vaccination has provided as a very efficient preventive tool, antiviral therapy is still needed to control viral infections not avoidable by prophylaxis with vaccines; those caused by viruses for which a vaccine is available, but vaccination is not universally implemented or does not result in complete, long-term protection; and in immunocompromised individuals with reduced immune control of viral replication. Addition of BrdUrd in combination with prednisolone to HSVi-transformed hamster embryo cells induces an alkaline phosphatase (AP). FdUrd enhances, dThd reduces the inducing capacity of BrdUrd and prednisolone. Induction is prevented by addition of cycloheximide or of cytosine arabinoside. BrdUrd reduces transport and phosphorylation of exogenously applied labeled Urd, especially the amount of UTP, UDP and Urd-diphosphate sugars. The Lineweaver-Burk-plot of uridine-up­take after addition of BrdUrd reveales the characteristics of the mixed type inhibition (competi­tive-noncompetitive).

Thymidine-, uridine- and choline-kinase in rabbit kidney cells infected with herpesvirus hominis, type I and II.

Data are presented about the activity of the thymidine-, uridine- and choline-kinase after infection with 21 strains ofherpesvirus hominis of serotype I or II in rabbit kidney cells. Type I strains increase the activity of the thymidine-kinase 15–20 fold over the controls, whereas the type II strains demonstrate a moderate activity, the level of the enzyme is increased 2–5 fold. One giant cell forming strain exhibits unusual properties, the TK activity decreases in correlation to the controls.
