Somatic embryogenesis and plant regeneration from pistil transverse thin cell layers of lemon (Citrus limon)

Callus induction, somatic embryogenesis and plant regeneration were obtained in Citrus limon (L.) Burm. (cv. Femminello) from cultures of pistil transverse thin cell layer explants [(t)TCLs]. Explants were cultured on two different media, based on Murashige and Skoog salts and vitamins, supplemented with 500 mg l-1 malt extract (MSI), or 500 mg l-1 malt extract and 13.3 ¼M 6-benzylaminopurine (MSII). Somatic embryogenesis and plant regeneration in a woody species: the European Spindle Tree (Euonymus europaeus L.).

Somatic embryogenesis and subsequent plant regeneration of Euonymus europaeus L (European Spindle Tree) were obtained from square pieces of mature zygotic embryos with an intervening callus phase. Callus and somatic embryos were induced using a Murashige and Skoog's semi-solid basal medium supplemented with several combinations of auxins and cytokinins. The greatest number of somatic embryos was obtained with a continuous exposure to 22.8 μM indoleacetic acid and 0.046 μM kinetin. Somatic Embryogenesis and Plant Regeneration from Hypocotyl Cultures of Digitalis obscura L.

Summary Hypocotyl sections from 20-day-old seedlings of Digitalis obscura L. were used to examine the effect of IAA on somatic embryogenesis. Although embryogenesis was obtained with all IAA concentrations (0.1, 0.5 and 1 ppm) and under both illumination conditions tested (16 h photoperiod and darkness), incubation in photoperiod increased yield of embryos and shortened the embryo induction time. Optimum results were obtained with 1 ppm IAA. About 30 % of the embryos obtained developed into complete plantlets when transferred to basal medium without growth regulators.

Priming maritime pine megagametophytes during somatic embryogenesis improved plant adaptation to heat stress

In the context of global climate change, forest tree research should be addressed to provide genotypes with increased resilience to high temperature events. These improved plants can be obtained by heat priming during somatic embryogenesis (SE), which would produce an epigenetic-mediated transgenerational memory.
