Development a mitigation strategy of enniatins in pasta under home-cooking conditions

2016

The effect of pasta cooking on the enniatins (ENs) contents was evaluated in white and whole-grain pasta. The cooking of pasta was performed for 10 min in boiling water. ENs were extracted from raw and cooked pasta by Solid-Liquid extraction with acetonitrile, and from cooking water by Dispersive Liquid–Liquid Micro-Extraction (DLLME) with a mixture of acetonitrile and carbone tetrachloride. Mycotoxin analysis was performed by Liquid Chromatography coupled to Tandem Mass Spectrometry. The effect of pasta cooking has a marked influence on the reduction of ENs levels. Reductions of 98–100% for ENA, 94–95% for ENA1, 14–49% for ENB and 53–65% for ENB1 were achieved after the process. ENs were d…

Current methodologies for phytosterol analysis in foods

2021

Abstract Plant sterols (PS) (phytosterols and phytostanols) are bioactive compounds of all vegetable foods where can be found as free sterol alcohols and as conjugates. These latter forms have been less studied, although they may have potential beneficial effects, whereas some PS have several approved health claims, especially in lowering blood cholesterol levels. In this context, it is necessary to establish analytical methods for food authentication, assessing PS health benefits and unfolding future applications. Several extraction methodologies have been employed for the extraction of PS from food matrixes (usually solvent extraction and saponification) although solid-phase extraction, s…

Method development for the determination of 1,1-dimethylhydrazine by the high-performance liquid chromatography-mass spectrometry technique.

2018

Unsymmetrical dimethyl hydrazine is highly toxic, carcinogenic compound, widely used for organic synthesis and drug development. Therefore, due to its high reactivity, direct analysis is problematic. Current study proposes to use derivatization reaction to increase selectivity and sensitivity of high-performance liquid chromatography–mass spectrometry method. Different derivatization agents were tested and optimal reaction media was found. Derivatization was performed by using small amounts of reagents to lower the cost of analysis. The full validation of the method was performed and it can be used in a routine control in pharmaceutical analysis. Method sensitivity is 0.15 ppm, and lineari…

Determination of pyrethroid insecticide residues in vegetable oils by using combined solid-phases extraction and tandem mass spectrometry detection

2005

Abstract A methodology has been developed for the determination of pyrethroid insecticide residues in vegetable oils, based in a solid-phase extraction (SPE) with combined solid supports phases. Several solid phases such as florisil, alumina, C18 and graphitized carbon black were tested in order to minimize fat residues, being a combination of treated alumina and C18 which provided the best results. Oil samples were partitioned with acetonitrile–hexane (1:1) and then eluted through a combined column packed with deactivated basic-alumina and C18, employing the acetonitrile extract as elution solvent. Gas chromatography tandem mass spectrometry was used for determination. Tetramethrin, Bifent…

Evaluation of selected buffers for simultaneous determination of ionic and acidic pesticides including glyphosate using anion exchange chromatography…

2019

Ion chromatography coupled with mass spectrometry is an established technique for determination of ionic analytes, however, sophisticated buffer removal equipment is required to eliminate inorganic compounds from the eluate before introduction into the ion source of mass spectrometer. A standard high-performance liquid chromatography coupled with tandem mass spectrometry setup using an ion exchange column (Metrosep® A Supp 5) is proposed as an alternative approach. For that reason, some buffers including non-volatile carboxylic acid based solutions have been evaluated for simultaneous trace determination of ionic and acidic pesticides including glyphosate in the same extract without a need …

Casein phosphopeptides released by simulated gastrointestinal digestion of infant formulas and their potential role in mineral binding

2006

Abstract Adapted and follow-up milk-based infant formulas were subjected to gastrointestinal digestion simulating physiological conditions. The naturally occurring casein phosphopeptides (CPPs) generated were fractionated by anion exchange high-performance liquid chromatography and sequenced by tandem mass spectrometry. In both infant formula digests, a total of 19 CCPs from bovine casein were identified, of which 7 corresponded to α s1 -casein and 12 to α s2 -casein. Most CPPs had the cluster sequence SpSpSpEE, representing the binding sites for minerals. The distribution of calcium, iron and zinc content in CPP fractions eluted from the anion exchange column was also studied. The results …

Statistical Approach for Biomarker Discovery Using Label-Free LC-MS Data: An Overview

2016

The identification of new diagnostic, prognostic, or theranostics biomarkers is one of the main aims of clinical research. Technologies like mass spectrometry (MS) focus on the discovery of proteins as biomarkers and are commonly being used for this purpose. Mass spectrometry consists in the separation by gas of charged molecules, based on their mass-over-charge. Liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) first involves a separation by liquid chromatography (LC) followed by mass spectrometry in the MS and MS/MS modes.

Recent trends in liquid chromatography-tandem mass spectrometry to determine pesticides and their metabolites in food

2007

Abstract The applications of liquid chromatography-mass spectrometry (LC-MS) have exploded in innumerable analytical fields, including pesticide-residue determination. There is no doubt that LC-MS is currently competing with gas chromatography (GC)-MS for the status of ‘reference’ analytical technique to determine pesticide residues and that its ever-increasing application is bound to the evolution of modern instruments and their growing performance qualities. We highlight the large number and variety of pesticides that can be readily determined using such instruments, the respective merits of the different mass analyzers, and the improvements brought about by tandem MS (MS 2 ). We also dis…

Capabilities of different liquid chromatography tandem mass spectrometry systems in determining pesticide residues in food

2007

Abstract Three different liquid chromatography–mass spectrometry (LC–MS) instruments equipped with triple quadrupole (QqQ), quadrupole ion trap (QIT) and quadrupole-time-of-flight (QqTOF), suitable to carry out tandem mass spectrometry, were examined to determine pesticide residues in food. Twelve pesticides (acrinathrin, bupirimate, buprofezin, cyproconazole, λ-cyhalothrin, fluvalinate, hexaflumuron, kresoxim-methyl, propanil, pyrifenox, pyriproxyfen and tebufenpyrad) and six matrices (oranges, strawberries, cherries, peaches, apricots and pears) were taken as model. The comparison was focused on two aspects: the quantitative, covering sensitivity, precision and accuracy as well as the qua…
