From six-coordinate to eight-coordinate iron(ii) complexes with pyridyltriazolo-pyridine frameworks

A new octacoordinated Fe(II) compound, [FeII(LN4)2](BPh4)2·3CH2Cl2, as an example of the scarce FeN8 systems, has been isolated with a tetradentate triazolopyridine-based ligand from a solution containing the related hexacoordinated [FeII(LN3)2]2+ complex, with LN3 = pyridyltriazolo-pyridyl-bromopyrimidine and LN4 = bis(pyridyltriazolo-pyridine).
Synthesis and structural properties of hexaaza[5]helicene containing two [1,2,3]triazolo[1,5-a]pyridine moieties

Abstract We have synthesized a novel hexaaza[5]helicene in a straightforward way from neocuproine. The crystal structure has been elucidated with direct-space strategy TALP which demonstrates the power of the powder X-ray diffraction technique. In this crystal structure it is possible to see an interplanar angle of 33(1)° between the two triazolopyridine rings. The centrosymmetric crystal structure is a racemic mixture, but the resolution was not possible due to a ring-chain isomerization in a solution that produces a dynamic racemization.
Synthesis and in vitro leishmanicidal activity of novel [1,2,3]triazolo[1,5-a]pyridine salts

Leishmaniasis remains a significant worldwide problem; it is of great interest to develop new drugs to fight this disease. Recently we described some [1,2,3] triazolo[1,5-a] pyridine compounds with significant leishmanicidal activity. The importance of water solubility in drug action made us realise that we could transform non charged triazolopyridines into charged analogues that could increase the degree of water solubility. Abstract Two novel zinc porphyrin dyes containing either benzotriazole or 2 H -[1,2,3]triazolo[4,5- c ]pyridine as auxiliary acceptors, respectively, were synthesized for dye-sensitized solar cells. These two auxiliary acceptors were firstly applied into porphyrin based dyes. The photophysical and electrochemical properties of the dyes were investigated. These two dyes show the whole visible light absorption when they are adsorbed on the TiO 2 film, which means that they are promising sensitizers for dye-sensitized solar cells. The absorption intensity of the triazolopyridine containing dye is higher than that of the benzotriazole containing dye, resulting in a better light harvesting. Thus…
Triazolopyridyl ketones as a novel class of antileishmanial agents. DNA binding and BSA interaction

A new series of triazolopyridyl pyridyl ketones has been synthetized by regioselective lithiation of the corresponding [1,2,3]triazolo[1,5-a]pyridine at 7 position followed by reaction with different electrophiles. The in vitro antileishmanial activity of these compounds was evaluated against Leishmaniainfantum, Leishmaniabraziliensis, Leishmaniaguyanensis and Leishmaniaamazonensis. Compounds 6 and 7 were found to be the most active leishmanicidal agents. Both of them showed activities at micromolar concentration against cultured promastigotes of Leishmania spp. (IC₅₀=99.8-26.8 μM), without cytotoxicity on J774 macrophage cells. Evaluation and synthesis of 7-arylhydroxymethyltriazolopyridines as potential cardiovascular agents

7-Arylhydroxymethyltriazolopyridines 3a-c and 4a-d were synthesized by regioselective lithiation of [1,2,3]triazolo[1,5-a]pyridines 1 and 2 and subsequent trapping of the 7-lithioderivatives formed using aryl aldehydes as electrophiles. The structural relationship between compounds 3a-c and 4a-d and arylethanolamines suggested their consideration as potential cardiovascular agents. A preliminary evaluation as vascular smooth muscle relaxants was carried out. These compounds did not act as α1-adrenoceptor antagonists and were unable to block calcium entry through voltage-dependent calcium channels. The synthesis from triazolopyridines 1, of novel triazolopyridylcarbonylpyridylcarbonyltriazolopyridines (TPyCOPyCOTPy) (tpcpctp) 7, and polypyridylcarbonylpyridines (pPyCOPy) (ppcp) 14, building blocks in supramolecular chemistry, is described. The heteroaryl-substituted triazolopyridines 3-phenyl-7-(pyrazin-2-yl)-[1,2,3]triazolo[1,5-a]pyridine (2) and 3-[6-(pyridazin-3-yl)-pyridin-2-yl]-[1,2,3]triazolo[1,5-a]pyridine (4) have been synthesized and characterized (by HRMS, IR,1H and13C NMR, XRPD, melting point). [1,2,3]Triazolo[1,5-a]pyridines. A theoretical (DFT) study of the ring-chain isomerization

The ring opening isomerization of [1,2,3]triazolo[1,5-a]pyridines to the corresponding 2-pyridyl derivatives has been studied by means of DFT calculations at the B3LYP/6-31+G(d,p) computational level. The effect of the substitution as well as those of protonation, deprotonation, and lithiation on different positions has been studied. The electronic characteristics of the optimized structures have been analyzed by means of the Atoms In Molecules (AIM), Electron Localization Function (ELF), Molecular Electrostatic Potential (MEP), and Natural Bond Orbital (NBO) methodologies. © 2008 Elsevier Ltd.
