Abstract The possible contribution of NKT cells to resistance to Mycobacterium tuberculosis infection remains unclear. In this paper we characterized the Vα14 NKT cell population following infection with Mycobacterium bovis bacillus Calmette-Guérin (BCG). BCG infection determined an early expansion of Vα14 NKT cells in liver, lungs, and spleen, which peaked on day 8 and was sustained until day 30. However, an NK1.1+ Vα14 NKT population preferentially producing IFN-γ predominated at an early stage (day 8), which was substituted by an NK1.1− population preferentially producing IL-4 at later stages (day 30). Sponges (Porifera) are the phylogenetically oldest still extant metazoan phylum. Recently elements of their immune system have been cloned and analyzed, primarily from the demosponges Suberites domuncula and Geodia cydonium. By differential display, two genes were identified in S. domuncula, whose translation products are involved in graft rejection/fusion: the allograft inflammatory factor (AIF-1) and the Tcf-like transcription factor (TCF). Since the AIF-1 and TCF genes are upregulated in vivo after tissue transplantation, especially in allografts, we investigated whether this reaction can be monitored in vitro. New amide and dioxopiperazine derivatives from leaves of Breynia nivosa

The first chemical investigation of leaves of Breynia nivosa from Nigeria resulted in the isolation of two new amide derivatives breynivosamides A and B (1 and 2) and two new dioxopiperazine derivatives breynivosines A and B (4 and 5) together with seven known compounds (3, 6-11). The structures of the new compounds were elucidated by 1D, 2D NMR and HRESIMS data as well as by comparison with the literature. All isolated compounds were tested for the cytotoxic and antimicrobial activities. Only cristatin A (6) showed cytotoxicity against the L5178Y mouse lymphoma cell line with an IC50 value of 13.9μM while breynivosamide A (1) exhibited moderate antimicrobial activity against Mycobacterium …

mTORC1 activation in B cells confers impairment of marginal zone microarchitecture by exaggerating cathepsin activity

Mammalian target of rapamycin complex 1 (mTORC1) is a key regulator of cell metabolism and lymphocyte proliferation. It is inhibited by the tuberous sclerosis complex (TSC), a heterodimer of TSC1 and TSC2. Deletion of either gene results in robust activation of mTORC1. Mature B cells reside in the spleen at two major anatomical locations, the marginal zone (MZ) and follicles. The MZ constitutes the first line of humoral response against blood‐borne pathogens and undergoes atrophy in chronic inflammation. Histamine and its possible role in cytolysis of white blood cells in guinea pigs sensitized with tubercle bacilli.

DELAYED or tuberculin-type sensitivity is–apart from some single publications–only transferable by white blood cells or peritoneal cells but not with humoral antibodies of sensitized organisms1. In vitro, after addition of antigen, a lysis of white blood cells2 is seen up to 35 per cent2. That effect is preceded by morphological changes3–5. Cytolysis in the presence of plasma will be discussed elsewhere. Labelled lymphoid cells of sensitized guinea pigs will be found after transfer to normal animals at the side of the dermal tuberculin reaction6. Mūsdienās tuberkuloze ir viena no biežākajiem pacientu nāves cēloņiem pasaulē. Tuberkulozi vairākumā gadījumu izraisa M.tuberculosis. Bakalaura darbā tika analizēta literatūra, lai saprastu M.tuberculosis rezistences mehānismus pret prettuberkulozes līdzekļiem. Praktiskajai daļai tika izmantota BACTEC MGIT sistēma M.tuberculosis rezistences noteikšanai, un arī tika izmantoti rezultāti, kas iegūti SIA „RAKUS” Tuberkulozes un plaušu slimību centra Mikobakterioloģijas laboratorijā un apkopoti laboratorijas informācijas sistēmā (LIS). Darbā apkopota literatūra par esošajām prettuberkulozes zāļu vielu un to metabolītu LC–MS–MS noteikšanas metodēm, raksturotas paraugu sagatavošanas un hromatogrāfisko metožu pielietošanas iespējas. Darba gaitā eksperimentāli pārbaudītas dažādas paraugu sagatavošanas un hromatogrāfiskās noteikšanas metodes, izstrādāta un validēta metode četru pirmās izvēles prettuberkulozes zāļu vielu un to galveno metabolītu noteikšanai cilvēka asins plazmā. Izstrādātās metodes veiktspēju pārbaudīja, analizējot klīniskos paraugus (n=12).
