Filter paper disk techniques for assay of nucleotidase

A DE filter disk technique for assaying the activity of nucleotidase is described. This method is based on the observation that nucleotides bind to the filters at 5 mM Tris-HCl (pH 7.8) while nucleosides do not. As parameter for the nucleotidase activity the decrease of bound nucleotides is determined. In parallel experiments the amount of the product (nucleoside) formed can be measured by DEAE Sephadex column chromatography. The filter disk technique can be applied for the determination of vmax and Km of a nucleotidase by using different ribonucleosidase monophosphate substrates.

Background: Primary hyperparathyroidism (PHPT) origins from a solitary adenoma in 70-95% of cases. Moreover, the advances in methods for localizing an abnormal parathyroid gland made minimally invasive techniques more prominent. This study presents a micro-cost analysis of two parathyroidectomy techniques. Patients and methods: 72 consecutive patients who underwent minimally invasive parathyroidectomy, video-assisted (MIVAP, group A, 52 patients) or "open" under local anaesthesia (OMIP, group B, 20 patients) for PHPT were reviewed. The simplest extension of the SM to account for the observed neutrino masses and mixings is the addition of at least two singlet fermions (or right-handed neutrinos). If their masses lie at or below the GeV scale, such new fermions would be produced in meson decays. Similarly, provided they are sufficiently heavy, their decay channels may involve mesons in the final state. Although the couplings between mesons and heavy neutrinos have been computed previously, significant discrepancies can be found in the literature. In vivo and ex vivo fluorescence video microscopy used to be a well-established method in life science with a variety of applications, such as in inflammation or cancer research. In this book chapter, we describe a model of in vivo fluorescence microscopy of the rat's lung with the exclusive advantage of qualitative and quantitative in vivo analysis of cell adhesion within the complex microenvironment of the ventilated and perfused lung. This investigation forms part of a study on the suitability of human omentum mesothelial cells (HOMES) as an alternative to endothelial cells (EC) for seeding vascular grafts. Isolated HOMES were grown in primary culture and characterized by their morphology (light microscopy and scanning electron microscopy (SEM)), as well as by fluorescence-activated cell sorting (FACS) and immunocytochemistry. The latter two methods showed cells which were positive for smooth muscle-type actin and cytokeratin, but negative for factor VIII-related antigen. Diplomdarba mērķis bija izpētīt, kā video materiālu ar angļu subtitriem pielietošana var palielināt lasīšanas ātrumu un izpratni 11. klasē. Pētījuma metode bija gadījuma pētījums ar 12 skolēnu izlasi. Datu vākšanas gaitā tika veikti skolēnu lasīšanas ātruma un izpratnes mērījumi, ievāktas atgriezeniskās saites anketas un veiktas daļēji strukturētas intervijas ar divām skolotājām.
