Synthesis and Antiproliferative Activity of 2,5-bis(3′-Indolyl)pyrroles, Analogues of the Marine Alkaloid Nortopsentin

2,5-bis(3′-Indolyl)pyrroles, analogues of the marine alkaloid nortopsentin, were conveniently prepared through a three step procedure in good overall yields. Derivatives 1a and 1b exhibited concentration-dependent antitumor activity towards a panel of 42 human tumor cell lines with mean IC50 values of 1.54 μM and 0.67 μM, respectively. Investigating human tumor xenografts in an ex-vivo clonogenic assay revealed selective antitumor activity, whereas sensitive tumor models were scattered among various tumor histotypes.

Drug resistant focal epilepsy can be treated by resecting the epileptic focus requiring a precise focus localisation using stereoelectroencephalography (SEEG) probes. As commercial SEEG probes offer only a limited spatial resolution, probes of higher channel count and design freedom enabling the incorporation of macro and microelectrodes would help increasing spatial resolution and thus open new perspectives for investigating mechanisms underlying focal epilepsy and its treatment. Anastomotic leakage is a frequent complication of intestinal surgery and a major source of surgical morbidity. The timing of anastomotic failures suggests that leaks are the result of inadequate mechanical support during the vulnerable phase of wound healing. Effect of cytochrome P450 inhibitors (diethyl dithiocarbamate, ketoconazole and grapefruit juice) on the pharmacokinetics of all-trans-retinoic acid.

Diethyl dithiocarbamate (DEDTC) has been reported to be a more powerful inhibitor of all-trans-retinoic acid (ATRA) in vitro metabolism than the well-established cytochrome P450 (CYP) inhibitor ketoconazole (KC). In recent years grapefruit juice (GJ) has been shown to be able to increase the oral bioavailability of several drugs by inhibiting intestinal CYP. This study investigated the in vivo effect of these CYP inhibitors on the pharmacokinetics of ATRA. Maģistra darbā tika izvērtēti jaunizveidotie biomateriāli, kas ietver glikoproteīnu, efektivitāte brūču dzīšanas procesos Wistar žurku tēviņiem un veikti akūtas toksicitātes noteikšanas pētījumi ICR peļu tēviņiem. Joprojām ir aktuāla dzīšanu stimulējošu, jaunu funkcionālu biomateriālu izveide, lai nodrošinātu ātrāku un efektīvāku brūču dzīšanas procesu klīnikā. Maģistra darbā tika izvērtēti jaunizveidotie biomateriāli, kas ietver glikoproteīnu, efektivitāte brūču dzīšanas procesos Wistar žurku tēviņiem un veikti akūtas toksicitātes noteikšanas pētījumi C57BL/6 līnijas peļu tēviņiem. Iegūtie rezultāti uz brūču dzīšanas procesu apstiprina jaunizveidoto biomateriālu (1 un 2) efektivitāti un progresīvu dinamiku brūču dzīšanas procesos, jau sākot ar pētījuma ceturto dienu, salīdzinot ar kontroles grupas rezultātiem.
