Group II metabotropic glutamate receptor activation by agonist LY379268 treatment increases the expression of brain derived neurotrophic factor in th…

2009

A number of in vitro and in vivo studies using selective agonists have indicated a neuroprotective role for group-II metabotropic glutamate (mGlu2/3) receptors in various models of neuronal injury. Although an interplay among neurotrophic factors and mGlu2/3 receptors signalling system has been suggested as possible mechanism involved on neuroprotection, at present poor information are available concerning the in vivo regulation by mGlu2/3 receptors activation of specific neurotrophic factors. By using in situ hybridization and western blotting methods the aim of present study was to analyse the potential regulatory role of selective mGluR2/3 agonist LY379268 treatment on brain derived neur…

Characterization of a new murine retinal cell line (MU-PH1) with glial, progenitor and photoreceptor characteristics

2013

Unlike fish and amphibians, mammals do not regenerate retinal neurons throughout life. However, neurogenic potential may be conserved in adult mammal retina and it is necessary to identify the factors that regulate retinal progenitor cells (RPC) proliferative capacity to scope their therapeutic potential. Müller cells can be progenitors for retinal neuronal cells and can play an essential role in the restoration of visual function after retinal injury. Some members of the Toll-like receptor (TLR) family, TLR2, TLR3 and TLR4, are related to progenitor cells proliferation. Müller cells are important in retinal regeneration and stable cell lines are useful for the study of retinal stem cell bi…

Colorimetric susceptibility testing for Aspergillus fumigatus: comparison of menadione-augmented 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazo…

1996

Two colorimetric methods that use Alamar Blue or 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) for assaying the in vitro activities of antifungal agents have been described. We report that both tests performed similarly when the antifungal activity of amphotericin B against Candida albicans was determined. However, only the MTT test generated interpretable data when Aspergillus fumigatus was used.

Cyclic triureas—synthesis, crystal structures and properties

2008

The synthesis of 24-membered macrocycles is described, in which rigid xanthene units (X) and/or diphenyl ether units (D) as flexible analogues are linked via urea groups. All four possible combinations (XXX, XXD, XDD, DDD) have been obtained with yields of 40-72% for the cyclisation step. In two cases, the respective cyclic hexamers (XXDXXD, XXXXXX) were also isolated. Two compounds have been characterised by a single crystal X-ray analysis of the free triurea (XXD, XDD) and one example (DDD) by its complex with tetrabutylammonium chloride. It shows the chloride anion in the centre of the macrocycle, held by six NH...Cl- hydrogen bonds. The interaction with various other anions has been stu…

Macrocyclic oligoureas with xanthene and diphenyl ether units

2009

Two cyclic oligoureas with 64- and 80-membered rings in which two sets of three or four rigid xanthene (X) units are connected via flexible diphenyl ether (D) units were synthesized by a stepwise fragment condensation. The compounds were characterized by (1)H NMR and ESI mass spectrometry. The structure of the cyclic octamer (XXXDXXXD) was additionally confirmed by single crystal X-ray analysis. The molecule assumes a strongly folded conformation with distorted C(2)-symmetry, stabilized by intramolecular hydrogen bonds. Surprisingly, intermolecular hydrogen bonds between the macrocycles were not observed. (1)H NMR spectra suggest a C(2) symmetrical conformation of the octamer in solution al…

Chemistry and Photochemistry of 2,6-Bis(2-hydroxybenzilidene)cyclohexanone. An Example of a Compound Following the Anthocyanins Network of Chemical R…

2014

The kinetics and thermodynamics of the 2,6-bis(2-hydroxybenzilidene)cyclohexanone chemical reactions network was studied at different pH values using NMR, UV-vis, continuous irradiation, and flash photolysis. The chemical behavior of the system partially resembles anthocyanins and their analogue compounds. 2,6-Bis(2-hydroxybenzilidene)cyclohexanone exhibits a slow color change from yellow to red styrylflavylium under extreme acidic conditions. The rate constant for this process (5 × 10(-5) s(-1)) is pH independent and controlled by the cis-trans isomerization barrier. However, the interesting feature is the appearance of the colorless compound, 7,8-dihydro-6H-chromeno[3,2-d]xanthene, isolat…

Osmium(III) Complexes with POP Pincer Ligands: Preparation from Commercially Available OsCl3·3H2O and Their X-ray Structures

2010

Complexes OsCl3{dbf(PiPr2)2} [1; dbf(PiPr2)2 = 4,6-bis(diisopropylphosphino)dibenzofuran], OsCl3{xant(PiPr2)2} [2; xant(PiPr2)2 = 9,9-dimethyl-4,5-bis(diisopropylphosphino)xanthene], and OsCl3{xant(PPh2)2} [3; xant(PPh2)2 = 9,9-dimethyl-4,5-bis(diphenylphosphino)xanthene] have been obtained in high yield by the reaction of the corresponding diphosphine with OsCl3·3H2O. The ruthenium(III) counterparts RuCl3{dbf(PiPr2)2} (4), RuCl3{xant(PiPr2)2} (5), and RuCl3{xant(PPh2)2} (6) are similarly obtained from RuCl3·3H2O in moderate yields. The X-ray structures of dbf(PiPr2)2 and complexes 1−3 are also reported.

The Interplay of Inverted Redox Potentials and Aromaticity in the Oxidized States of New π-Electron Donors: 9-(1,3-Dithiol-2-ylidene)fluorene and 9-(…

2006

Derivatives of 9-(1,3-dithiol-2-ylidene)fluorene (9) and 9-(1,3-dithiol-2-ylidene)thioxanthene (10) have been synthesised using Horner-Wadsworth-Emmons reactions of (1,3-dithiol-2-yl)phosphonate reagents with fluorenone and thioxanthen-9-one. X-ray crystallography, solution electrochemistry, optical spectroscopy, spectroelectrochemistry and simultaneous electrochemistry and electron paramagnetic resonance (SEEPR), combined with theoretical calculations performed at the B3P86/6-31G** level, elucidate the interplay of the electronic and structural properties in these molecules. These compounds are strong two-electron donors, and the oxidation potentials depend on the electronic structure of t…

Penicillium auxotrophic mutants can be detected by suing xanthene dyes

1987

Auxotrophic mutants ofPenicillium spp. have been directly isolated after mutagenic treatment from agar plates containing Xanthene dyes. They grow as characteristic small colored colonies. Some strains were tested and they showed a differential response depending on the Xanthene dye used.
