Abstract Background While growing in natural environments yeasts can be affected by osmotic stress provoked by high glucose concentrations. The response to this adverse condition requires the HOG pathway and involves transcriptional and posttranscriptional mechanisms initiated by the phosphorylation of this protein, its translocation to the nucleus and activation of transcription factors. One of the genes induced to respond to this injury is YHR087W. It encodes for a protein structurally similar to the N-terminal region of human SBDS whose expression is also induced under other forms of stress and whose deletion determines growth defects at high glucose concentrations. <p style="text-align: justify;">In this study, we have investigated the influence of ethanol on yeast film formation and cell surface hydrophobicity (CSH). A yeast strain (P3) previously isolated from film yeast was grown in a medium containing increasing ethanol concentration ranging from 0 to 14 p. cent (v/v). It results from this study that up to 10 p. cent ethanol, the greater was the ethanol concentration, the greater was the growth of film. Using two different techniques (phase partition method, magnobead assay), we have shown that ethanol altered the CSH of the yeast. Aims: The purification and biochemical properties of the 1,4-β-xylosidase of an oenological yeast were investigated. Methods and Results: An ethanol-tolerant 1,4-β-xylosidase was purified from cultures of a strain of Pichia membranifaciens grown on xylan at 28°C. The enzyme was purified by sequential chromatography on DEAE cellulose and Sephadex G-100. The relative molecular mass of the enzyme was determined to be 50 kDa by SDS-PAGE. The activity of 1,4-β-xylosidase was optimum at pH 6·0 and at 35°C. The activity had a Km of 0·48 ± 0·06 mmol l−1 and a Vmax of 7·4 ± 0·1 μmol min−1 mg−1 protein for p-nitrophenyl-β-d-xylopyranoside. International audience; Circular dichroism (CD) spectroscopy of secondary structure conformation of the purified green bell pepper hydroperoxide lyase (HPL), cloned in the yeast Yarrowia lipolytica, was investigated. The CD spectra of HPL in iso-octane, obtained at 60 °C, in the presence of the reducing agent dithiothreitol showed dramatic increase in α-helix content. During fermentation, the geraniol concentration showed a dramatic drop and a small amount was transformed into linalool and -terpineol by an acid catalysed chemical reaction. Yeast are responsible for changing most of the geraniol to citronellol through enzymatic reactions. Abstract Glycoconjugated aroma precursors from neutral grape varieties were fractionated on 5 g C18 RP cartridge. Each of the seven fractions collected was divided in three parts: first part was hydrolysed with Pectinol in order to determine total aromatic heterosides, while the second part was treated with an active strain of the yeast Saccharomyces cerevisiae UCLM 325 and the third part with acetonic powder of the same yeast strain. Genome-wide analysis of the wine yeast strain Saccharomyces cerevisiae PYCC4072 identified 36 genes highly expressed under conditions of low or absent nitrogen in comparison with a nitrogen-replete condition. Reverse transcription-PCR analysis for four of these transcripts with this strain and its validation with another wine yeast strain underlines the usefulness of these signature genes for predicting nitrogen deficiency and therefore the diagnosis of wine stuck/sluggish fermentations.
