1,2,4-Oxadiazole rearrangements involving an NNC side-chain sequence.

The thermal rearrangement of N-1,2,4-oxadiazol-3-yl-hydrazones into 1,2,4-triazole derivatives is reported. This represents the first example of a three-atom side-chain rearrangement involving an NNC sequence linked at the C(3) of the oxadiazole. The reactions carried out under solvent-free conditions produced good to high yields of the final products.
The reaction of amino-imidazoles, -pyrazoles and -triazoles with α,β-unsaturated nitriles

The reactions of α,β-unsaturated nitriles (1, 9, 12) as bielectrophiles with aminoazoles (2, 4, 6) as binu-cleophiles were investigated. Acrylonitrile (1) reacts almost exclusively in a chemoselective Michael-type addition yielding the substituted azoles 3, 5 and 7, respectively. Cinnamonitriles 9a,b behave in a similar way, but the free CN group adds a second molecule 4 yielding 10a,b and its cyclocondensation product 11a,b as minor component. The attempted formation of azolopyrimidines is best achieved by the reaction of the benzylidenemalononitriles 12a - f with 2 or 4. The process is chemo- and regioselective. Abstract A simple and high yielding preparation of pyrazoles and pyrroles is described. Thermolysis of tetrazolo[1,5- b ]pyridazines, tetrazolo[1,5- a ]pyrimidines and tetrazolo[1,5- a ]pyridines allowed easy ring contraction thus providing a facile preparation of cyanopyrazole and cyanopyrrole heterocycles. Since the cyano group is a versatile precursor of other functionalities, the reaction appears of particular interest for the construction of a variety of pyrazoles and pyrroles. The title compound, C8H11N3O3, crystallizes with Z' = 4. One pyrazole N atom is substituted and excluded from intermolecular contacts. The amine N, acetyl O and an ester O atom are involved in the formation of nearly planar molecular layers. The layers are perpendicular to the c axis, with an interlayer distance of 3.333 Å. The hydrogen-bonding patterns are similar for each molecule, i.e. intramolecular N-H...O, as well as intermolecular N-H...O and C-H...N(pyrazole), bonds are present.

The cathodic reduction of dicationic imidazolium bromides, whose spacer is either an aliphatic chain or a xylyl group, leads to the formation of the corresponding N-heterocyclic carbenes (NHCs), which were isolated as the corresponding thiones, after reaction with elemental sulfur. The behaviour of the dications was compared with the corresponding monocations. The behaviour of dicarbenes depends on the nature of the spacer. This study evidenced that dicarbenes deriving from xylyl dications are less stable than the corresponding aliphatic ones (giving lower yields in thiones), due to a debenzylation reaction. Effects of the organic calcium antagonists verapamil and flunarizine on pentylenetetrazol induced paroxysmal depolarizations were tested in organotypic neocortical explants taken from neonatal rats. In these in vitro experiments the papaverin derivative verapamil depressed, and finally abolished, epileptic discharges in all cases. The piperazine derivative flunarizine, however, which is known to suppress epileptic discharges in hippocampal CA3 neurons (Bingmann and Speckmann 1986), showed no significant antiepileptic effects in the explanted neocortical neurons. International audience; The synthesis of functionalized porphyrins and their conjugates from meso-tetraarylporphyrins through the acylation and the oxidation of beta-aminoporphyrins was investigated. 2,3-Dioxochlorins were prepared by the oxidation of a variety of beta-aminoporphyrins and subsequently used in a condensation reaction with functionalized aromatic aldehydes and ammonium acetate to form beta-functionalized porphyrins bearing a fused imidazole ring. Under optimized experimental conditions both reactions tolerate various functional groups and afford the products in an appropriate overall yield.
