Inhibition of imidazolidinone intermediate formation in the aldol reactions catalyzed by zinc–prolinamide complexes

2011

Abstract The use of zinc salts as cocatalysts in aldol condensations catalyzed by single prolinamide (and in the extension by other more complex prolinamides) can prevent the formation of the parasitic intermediate imidazolidinone, with faster and also more stereoselective reactions than those catalyzed by the free amine. This new finding, together with this ion’s already known properties, make zinc salts highly suitable additives for aldol reactions catalyzed for prolinamide derivatives.

Use of Saccharomyces cerevisiae as a whole cell system for aldol condensation in organic medium: Study of the factors affecting the biotransformation

2011

Abstract Saccharomyces cerevisiae has been employed as a whole cell catalyst for a number of asymmetric transformations. This work explores the ability of this microorganism to carry out the asymmetric aldol condensation between 4-nitrobenzaldehyde and acetone. For this purpose, lyophilized cells of the FY86 laboratory strain from stationary phase cultures were employed. This reaction shows stereoselectivity, and its progress is affected by the water concentration in the medium, temperature and the growth stage of the yeast culture. Cell lysis experiments indicate that activity responsible for this biotransformation is located in the soluble fraction.

Identification of antigenic proteins from Echinostoma caproni (Trematoda) recognized by mouse immunoglobulins M, A and G using an immunoproteomic app…

2008

Antigenic proteins of Echinostoma caproni (Trematoda) against mouse IgM, IgA, IgG, IgG1 and IgG2a were investigated by immunoproteomics. Excretory/secretory products (ESP) of E. caproni separated by two-dimensional (2D) gel electrophoresis were transferred to nitrocellulose membranes and probed with the different mouse immunoglobulin classes. A total of four proteins (enolase, 70 kDa heat-shock protein (HSP-70), actin and aldolase) were accurately identified. Enolase was recognized in eight different spots of which seven of them were detected in the expected molecular weight and were recognized by IgA, IgG or IgG and IgG1. Another spot identified as enolase at 72 kDa was only recognized by …

Expression of Phosphofructokinase Is Not Sufficient to Enable Embden-Meyerhof-Parnas Glycolysis in Zymomonas mobilis ZM4

2019

Zymomonas mobilis is a bacterium that produces ethanol from glucose at up to 97% of theoretical efficiency on a carbon basis. One factor contributing to the high efficiency of ethanol production is that Z. mobilis has a low biomass yield. The low biomass yield may be caused partly by the low ATP yield of the Entner-Doudoroff (ED) glycolytic pathway used by Z. mobilis, which produces only one ATP per glucose consumed. To test the hypothesis that ATP yield limits biomass yield in Z. mobilis, we attempted to introduce the Embden-Meyerhof-Parnas (EMP) glycolytic pathway (with double the ATP yield) by expressing phosphofructokinase (Pfk I) from Escherichia coli. Expression of Pfk I caused growth…

Stereoselective synthesis of the published structure of feigrisolide A. Structural revision of feigrisolides A and B.

2006

The total synthesis of the proposed structure of feigrisolide A is reported. Ethyl (S)-3-hydroxybutyrate was the chiral starting material. A Brown asymmetric allylation and an Evans aldol reaction were key steps of the synthesis. The NMR data of the synthetic product are different from those of the natural product. The published structure of feigrisolide A is therefore erroneous. A subsequent comparison of spectral data strongly suggests that feigrisolides A and B are identical with (-)-nonactic acid and (+)-homononactic acid, respectively.

Stereoselective Synthesis of the Cytotoxic Macrolide FD-891

2006

[reaction: see text] A total synthesis of the naturally occurring, cytotoxic macrolide FD-891 is described. Three fragments were first stereoselectively constructed using mainly asymmetric aldol and allylation reactions. The complete framework was then assembled using two Julia-Kocienski olefinations to connect the three fragments and a Yamaguchi reaction to close the macrolactone ring.

Natural eutectogels: sustainable catalytic systems for C–C bond formation reactions

2021

Natural eutecto gels were prepared combining properties of amino acids with the ones of deep eutectic solvents. Soft materials obtained were fully characterised determining the gel-sol transition temperatures and analysing mechanical and morphological features through rheological measurements and polarised optical microscopy. All eutecto gels were tested as catalytic reaction media for the enantioselective aldol reaction and, the best performing one was also used to carry out the Michael addition. In both cases, eutecto gels proved to have a great potential as sustainable reaction media, allowing to perform processes under mild conditions, obtaining excellent yields and, in some cases, also…

Highly enantioselective aza-Henry reaction with isatin N-Boc ketimines.

2014

A highly enantioselective aza-Henry reaction with isatin N-Boc ketimines using a Cu(II)–BOX complex as a catalyst is described. The reaction, which does not require protection of the N1 atom, provides the corresponding nitroamines bearing a quaternary stereocentre with high yields and enantiomeric excesses (up to 99.9%).

Concise asymmetric synthesis of Linezolid through catalyzed Henry reaction

2013

A new asymmetric synthesis of the antibiotic Linezolid was performed through a copper-catalyzed Henry reaction as the key step. The use of camphor-derived aminopyridine ligands helped to improve the yields of the chiral precursor and to obtain Linezolid in good overall yield and enantiomeric excess.

A catalytic highly enantioselective direct synthesis of 2-bromo-2-nitroalkan-1-ols through a Henry reaction.

2008

Highly enantiomerically enriched 2-bromo-2-nitroalkan-1-ols are prepared by direct condensation of aliphatic and aromatic aldehydes with bromonitromethane in the presence of a catalytic amount of copper(II) acetate and a C1-symmetric camphorderived amino pyridine ligand.
