Mammalian intestinal alkaline phosphatase acts as highly active exopolyphosphatase.

Recent results revealed that inorganic polyphosphates (polyP), being energy-rich linear polymers of orthophosphate residues known from bacteria and yeast, also exist in higher eukaryotes. However, the enzymatic basis of their metabolism especially in mammalian cells is still uncertain. Here we demonstrate for the first time that alkaline phosphatase from calf intestine (CIAP) is able to cleave polyP molecules up to a chain length of about 800. The enzyme acts as an exopolyphosphatase degrading polyP in a processive manner. The pH optimum is in the alkaline range. ABSTRACT It is generally accepted that Bacillus thuringiensis Cry toxins insert into the apical membrane of the larval midgut after binding to specific receptors, and there is evidence that the distribution of binding molecules along the midgut is not uniform. By use of the voltage-sensitive dye DiSC 3 (5) and 125 I-labeled Cry1Ac, we have measured the effect of Cry1Ac in terms of permeabilization capacity and of binding parameters on brush border membrane vesicles (BBMV) prepared from the anterior and the posterior regions of the larval midgut from two insect species, Manduca sexta and Helicoverpa armigera . The dipping of berries in a dilute solution of sodium hydroxide during a short time was evaluated as pretreatment undertaken prior to convective dehydration of wine grapes. The impact of the sodium hydroxide content and dipping time on weight loss (WL) at different dehydration times was thoroughly assessed using central composite design (CCD) and response surface methodology (RSM). Furthermore, the effects of these two variables were also investigated on the skin mechanical properties of dehydrated grapes. Our purpose was to analyse the influence of O2 pressure on the adhesion, proliferation, and osteogenic differentiation of human dental pulp stem cells (DPSC) on 3-tricalcium phosphate (3-TCP) scaffold. DPSC isolated from extracted third molars were seeded on 3-TCP and cultured under 3% or 21% O2 pressure. Cell adhesion, proliferation, and differentiation over the biomaterial were evaluated at 7, 13, 18, and 23 days of culture. Cell adhesion was determined by light microscopy, proliferation by DNA quantification, and osteogenic differentiation by alkaline phosphatase activity analysis. The kinetics of the alkaline hydrolysis and the configuration, conformation and electronic structure were studied using IR, NMR spectroscopy, X-ray analysis and AM1 theoretical calculations for a series of (Z)-4-(substituted benzylidene)-2-phenyl-4H-oxazol-5-ones (1) and compared with analogous results reported for (E)-4-benzylidene-2-phenyl-4H-furan-5-ones (5) and related compounds.

Performance of a series of novel N-substituted acrylamides in capillary electrophoresis of DNA fragments

DNA separations by capillary electrophoresis in viscous solutions of novel polymers, made with Ω-hydroxyl, N-substituted acrylamides (notably N-acryloyl amino propanol, AAP and N-acryloyl amino butanol, AAB) are evaluated. Whereas in standard poly(acrylamide), at 6% concentration, the theoretical plate number (N) does not exceed 500 000, in 6% poly(AAP) N reaches 922 000 and in 6% poly(AAB) N values as high as 1 200 000 are obtained. Also, copolymers of AAP and AAB give N values in excess of 1 million plates. A clean analytical method for the spectrophotometric determination of formetanate incorporating an on-line microwave assisted hydrolysis step

Abstract A fast and completely automated procedure is proposed for the spectrophotometric determination of formetanate in waters by means of its reaction with p-aminophenol (PAP). The method involves the on-line alkaline hydrolysis of formetanate to m-aminophenol (MAP) and its reaction, in the presence of KIO 4 as oxidant agent, with the quinoneimine form of PAP, to form a blue indophenol dye which absorbs at 576 nm. Classification of vegetable oils according to their botanical origin using n-alkane profiles established by GC-MS.

n-Alkane profiles established by gas chromatography-mass spectrometry (GC-MS) were used to classify vegetable oils according to their botanical origin. The n-alkanes present in corn, grapeseed, hazelnut, olive, peanut and sunflower oils were isolated by means of alkaline hydrolysis followed by silica gel column chromatography of the unsaponifiable fractions. The n-alkane fraction was constituted mainly of n-alkanes in the range C8-C35, although only those most abundant (15 n-alkanes, from 21 to 35 carbon No.) were used as original variables to construct linear discriminant analysis (LDA) models. Isopropanolic Cimicifuga racemosa is favorable on bone markers but neutral on an osteoblastic cell line

Postmenopausal women treated with an isopropanolic extract of Cimicifuga racemosa underwent a decrease in the urinary concentration of N-telopeptides, a marker of bone resorption, and an increase in alkaline phosphatase, a marker of bone formation, at the third month of therapy. Serum from treated women did not modify the activity of alkaline phosphatase or the expression of three genes, runt-related transcription factor-2 (Runx-2), alkaline phosphatase, and osteocalcin, when added to the MC3T3-E1 osteoblastic cell line.
