Geschlechts-Chromatin und Handleistenmuster in einer Landesnervenklinik

Die 804 mannlichen Patienten einer allgemeinen Landesnervenklinik wurden systematisch nach Kerngeschlecht und Handleistenmustern untersucht. Dabei fanden sich 0,62% chromatin-positive, nach der Chromosomenanalyse verschiedene Typen des Klinefelter-Syndroms. Wenn auch eine Reihe von Papillarleistenmustern fur Aberrationen der Geschlechtschromosomen charakteristisch sind, so ist die Auspragung doch zu variabel, um diese Aberrationen untereinander zu differenzieren. Die Auswahl nach Grose und kriminellen Vorgeschichte forderte in diesem Material keinen YY-Fall zu Tage.

The authors present a novel method for obtaining chromatin profiles from low cell numbers without prior nuclei isolation. The method is successfully implemented in generating epigenetic profile from 100 cells with high signal-to-noise ratio.

Association between C′3 phenotypes and various diseases

The distribution of C′3 phenotypes in leprosy, rheumatism, diabetes, hyperlipedemia and hepatitis were studied. There was a significant excess of FF-phenotypes in patients with rheumatic factor while in hepatitis the SS-phenotype was significantly lower and a relatively high frequency of FF was stated. The results are discussed.

Gypsy homologous sequences in Drosophila subobscura (gypsyDS).

Characterization of sequences homologous to the Drosophila melanogaster gypsy transposable element was carried out in Drosophila subobscura (gypsyDS). They were found to be widely distributed among natural populations of this species. From Southern blot and in situ analyses, these sequences appear to be mobile in this species. GypsyDS sequences are located in both euchromatic and heterochromatic regions. A complete gypsyDS sequence was isolated from a D. subobscura genomic library, and a 1.3-kb fragment which aligns with the ORF2 of the D. melanogaster gypsy element was sequenced. Alkaline hydrolysis/methylation-acetylation: a new technique for ultrastructural DNA cytochemistry.

SUMMARY A new technique for the visualization of DNA-containing structures in electron microscopy is described. Samples of glutaraldehyde-fixed bone marrow from rats were subjected to alkaline hydrolysis to remove RNA and the phosphate of phospho-proteins, followed by a combined blockage of protein carboxyl and amino groups through methylation-acetylation. After uranyl acetate staining of epoxy-embedded ultrathin sections, chromatin from all cell types showed a highly selective and intense electron opacity. Staining methods for DNA were also positive in semithin sections. This simple procedure could be very useful in ultrastructural cytochemistry of DNA and chromatin.

Redox signaling and histone acetylation in acute pancreatitis

Histone acetylation via CBP/p300 coordinates the expression of proinflammatory cytokines in the activation phase of inflammation, particularly through mitogen-activated protein kinases (MAPKs), nuclear factor-κB (NF-κB), and signal transducers and activators of transcription (STAT) pathways. In contrast, histone deacetylases (HDACs) and protein phosphatases are mainly involved in the attenuation phase of inflammation. The role of reactive oxygen species (ROS) in the inflammatory cascade is much more important than expected. Site specificity of yeast histone acetyltransferase B complex in vivo

Saccharomyces cerevisiae Hat1, together with Hat2 and Hif1, forms the histone acetyltransferase B (HAT-B) complex. Previous studies performed with synthetic N-terminal histone H4 peptides found that whereas the HAT-B complex acetylates only Lys12, recombinant Hat1 is able to modify Lys12 and Lys5. Here we demonstrate that both Lys12 and Lys5 of soluble, non-chromatin-bound histone H4 are in vivo targets of acetylation for the yeast HAT-B enzyme. Moreover, coimmunoprecipitation assays revealed that Lys12/Lys5-acetylated histone H4 is bound to the HAT-B complex in the soluble cell fraction. A chromatin-associated histone deacetylase from pea (Pisum sativum)

Abstract A histone deacetylase activity has been found in preparation of chromatin from pea (Pisum sativum) embryonic axes. This activity readily deacetylates free histones and is somewhat specific for H2A and H2B; this property and its chromatographic behaviour allowed us to identify the enzyme with the previously described histone deacetylase HD2 (Sendra et al., Plant Mol. Biol., 11 (1988) 857). HD2 is only loosely associated to chromatin but the enzymatic activity is enhanced when chromatin adopts a folded conformation. Polyamines and divalent cations activate the enzyme, probably due to their effect on chromatin folding.

Protein phosphatases and chromatin modifying complexes in the inflammatory cascade in acute pancreatitis

Acute pancreatitis is an inflammation of the pancreas that may lead to systemic inflammatory response syndrome and death due to multiple organ failure. Acinar cells, together with leukocytes, trigger the inflammatory cascade in response to local damage of the pancreas. Amplification of the inflammatory cascade requires up-regulation of pro-inflammatory cytokines and this process is mediated not only by nuclear factor κB but also by chromatin modifying complexes and chromatin remodeling.
