Feeding enhances extracellular lactate of local origin in the rostromedial hypothalamus but not in the cerebellum.

Abstract The use of brain microdialysis together with chronic vascular catheterization allowed us to assay extracellular fluid lactate (ECF L ) in both the ventromedial–paraventricular (VMH–PVN) area of the hypothalamus and the cerebellum, in parallel with measures of plasma levels, and in relation to food intake. A 45 min scheduled meal increased VMH–PVN ECF L by 28%. This increase was not observed in the cerebellum. The prandial increase in plasma glucose (43%, from 4.74 to 6.77 mM) and lactate (84%, from 0.83 to 1.53 mM) showed a different temporal pattern and lasted longer than that of the ECF L . Asymmetries in classically conditioned head movements and cingulate cortex slow potentials in cats.

Presuming that conditioned head movements in the cat indicate a preference for a specific direction, asymmetries were also expected to be found in bilaterally recorded cingulate cortex slow potentials to a symmetrical tone stimulus during discriminative conditioning. Conditioned stimuli (CS, 1000 and 2500 Hz tones) were delivered through two miniature loudspeakers fitted to the head in front of both ears. Conditioned orienting (alpha) and delayed behavioral and evoked neural responses during classical conditioning

A differentiation of short-latency (alpha) and long-latency (delayed) classically conditioned behavioral and evoked neural (hippocampal) responses was attempted. Further, facilitation and retardation of these responses were studied in an experimental design in which 10 paired conditioning sessions either preceded (CC-CO group) or followed (CO-CC group) 10 randomly unpaired presentations of conditioned stimuli (CS) and unconditioned stimuli (UCS). Evidence of the origin of specific spontaneous head turns during intertrial intervals.

Direction and the frequency of spontaneous head movements during the ITIs following forward and backward paired trials were compared to an acquisition of a conditioned orienting (alpha) response directed to the side of the tone source. The head movements were analyzed from video recordings using classification of head turns to preferred and to nonpreferred directions. The results showed a significant increase in the alpha responses during the forward paired conditioning to the preferred direction and rapid extinction during the subsequent backward conditioning sessions. Membrane potential dye imaging of ventromedial hypothalamus neurons from adult mice to study glucose sensing

Studies of neuronal activity are often performed using neurons from rodents less than 2 months of age due to the technical difficulties associated with increasing connective tissue and decreased neuronal viability that occur with age. Here, we describe a methodology for the dissociation of healthy hypothalamic neurons from adult-aged mice. The ability to study neurons from adult-aged mice allows the use of disease models that manifest at a later age and might be more developmentally accurate for certain studies.
