Acid Titrations of poly(dG-dC).poly(dG-dC) in Aqueous Solution and in a w/o Microemulsion

The model polynucleotide poly(dG-dC).poly(dG-dC) (polyGC) was titrated with a strong acid (HCl) in aqueous unbuffered solutions and in the quaternary w/o microemulsion CTAB/n-pentanol/n-hexane/water. The titrations, performed at several concentrations of NaCl in the range 0.005 to 0.600 M, were followed by recording the modifications of the electronic absorption and of the CD spectra (210or = lambdaor =350 nm) upon addition of the acid. In solution, the polynucleotide undergoes two acid-induced transitions, neither of which corresponds to denaturation of the duplex to single coil. Easy Route to Functionalize Iron Oxide Nanoparticles via Long-Term Stable Thiol Groups

International audience; The functionalization of superparamagnetic iron oxide nanoparticles (SPIOs) by meso-2,3-dimercaptosuccinic acid (DMSA) was investigated. Under ambient conditions, the thiol groups from DMSA are not stable and do not allow a direct functionalization without storage in stringent conditions or a chemical regeneration of free thiols. In this study, we have developed a protocol based on poly(ethylene glycol) (PEG) grafting of SPIO prior to DMSA anchoring. We have observed that PEG helps to increase the stability of thiol groups under ambient conditions. Alkaline titrations of poly(dG-dC).poly(dG-dC): microemulsion versus solution behaviour

PolyGC was titrated with a strong base in the presence of increasing concentrations of NaCl (from 0.00 to 0.60M) either in water solution or with the polynucleotide solubilized in the aqueous core of reverse micelles, i.e., the cationic quaternary water-in-oil microemulsion CTAB/n-hexane/n-pentanol/water. The results for matched samples in the two media were compared. CD and UV spectroscopies and, for the solution experiments, pH measurements were used to follow the course of deprotonation. In vitro pulsatile and continuous transdermal delivery of buserelin by iontophoresis

Abstract Transdermal delivery of buserelin, a nonapeptide, from hydroxyethylcellulose hydrogel through isolated human stratum comeum was studied. In vitro studies were carried out in a drug release apparatus (DAB 10/USP XII) using self designed rotary disk cells equipped with platinum electrodes. Different forms of current were examined. Interaction of Quinine with Model Lipid Membranes of Different Compositions

The binding of a drug such as Quinine with neutral and negatively charged small unilamellar lipid vesicles at pH 7 and 37 degrees C was investigated. Changes in the fluorescence properties of the drug after association with the liposomes were used to obtain binding isotherms over a range of phospholipid compositions at different ionic strengths. Under such conditions, the association was strongly enhanced by the negative net charge of the bilayer but diminished by the increasing presence of electrolytes in the aqueous media. Abstract The association of polyanions with liposomes based on dimyristoyl phosphatidyl choline and stearylamine at a ratio of 50:50 was investigated in terms of the binding model and, to a lesser extent, of the partition model. Aqueous size-exclusion chromatography has been proposed as a major technique for monitoring the interactions between liposomes and polyelectrolytes and the results were compared with those obtained by fluorescence spectroscopy. The effect of both the phospholipid composition of the bilayer and the ionic strength of the medium on the association between poly(2-vinyl pyridine) and mixed liposomes based on dimyristoyl phosphatidic acid and dimyristoyl phosphatidil choline was investigated using fluorescence spectroscopy. Intensity changes upon addition of vesicles have allowed us to estimate the extent of the association and data have been processed as association isotherms. Freeze-dried eudragit-hyaluronan multicompartment liposomes to improve the intestinal bioavailability of curcumin.

This work aimed at finding an innovative vesicle-type formulation able to improve the bioavailability of curcumin upon oral administration. To this purpose, phospholipid, Eudragit® S100 and hyaluronan sodium salt were combined to obtain eudragit-hyaluronan immobilized vesicles using an easy and environmentally-friendly method. Microfluidic Fabrication of Physically Assembled Nanogels and Micrometric Fibers by Using a Hyaluronic Acid Derivative

The employ of a hyaluronic acid (HA) derivative, bearing octadecyl (C18) and ethylenediamine (EDA) groups, for microfluidic fabrication of nanogels and microfibers is reported in this study. Two HA-EDA-C18 derivatives (125 and 320 kDa) having ionic strength sensitive properties are synthesized and characterized. The control of the rheological properties of HA-EDA-C18 aqueous dispersions by formation of inclusion complexes with hydroxypropyl-β-cyclodextrins (HPCD) is described. Reversibility of C18/HPCD complexation and physical crosslinking is detected in media with different ionic strength through oscillation frequency tests.
