Biochemical and structural features of a novel cyclodextrinase from cow rumen metagenome.

A novel enzyme, RA.04, belonging to the alpha-amylase family was obtained after expression of metagenomic DNA from rumen fluid (Ferrer et al.: Environ. Microbiol. 2005, 7, 1996-2010). The purified RA.04 has a tetrameric structure (280 kDa) and exhibited maximum activity (5000 U/mg protein) at 70 degrees C and was active within an unusually broad pH range from 5.5 to 9.0. It maintained 80% activity at pH 5.0 and 9.5 and 75 degrees C. The enzyme hydrolyzed alpha-D-(1,4) bonds 13-fold faster than alpha-D-(1,6) bonds to yield maltose and glucose as the main products, and it exhibited transglycosylation activity. Biosorption of lead(II), zinc(II) and nickel(II) from industrial wastewater by Stenotrophomonas maltophilia and Bacillus subtilis

Abstract The biosorption of Pb(II), Zn(II) and Ni(II) from industrial wastewater using Stenotrophomonas maltophilia and Bacillus subtilis was investigated under various experimental conditions regarding pH, metal concentration and contact time. The optimum pH values for the biosorption of the three metals were in the range 5.0-6.0, while the optimal contact time for the two bacterial species was 30 min. Experimental data was analyzed using Langmuir and Freundlich isotherms; the former had a better fit for the biosorption of Pb(II), Zn(II) and Ni(II). The adsorption properties of water molecules on oligo and polysaccharides are attributed to the OH groups of the glucose rings in the molecules. The water molecules are adsorbed onto OH groups by hydrogen bond formation. Near infrared spectroscopic and gravimetric techniques were used in analysing the adsorption characteristics of malto-oligosaccharides and some polysaccharides. Near infrared spectra of the dry oligo and polysaccharides were acquired during the adsorption of water molecules at a relative humidity of 50-55%. I risultati delle prove 2005-2006 nelle 10 località del Centro-sud e Isole hanno evidenziato buone medie produttive con la conferma a leader della varietà Prosa. I buoni risultati in generale evidenziano ulteriormente l’adattabilità di questa specie alle condizioni mediterranee.
Influence of the storage conditions on some physicochemical and mycological parameters of honey

The authors studied the changes in yeast and mould counts, pH, free and lactone acidity, colour and sugar composition undergone by honeys with water contents of 169, 188 and 220 g kg-1 on storage at refrigeration temperature (4–7°C), room temperature and 28°C for two years. Of the solid culture media assayed, honey agar and osmophilic medium fostered yeast growth while malt-extract agar and yeast-extract-malt-extract agar allowed for better development of filamentous fungi. The yeasts isolated belonged to the genera Saccharomyces, Schizosaccharomyces and Zygosaccharomyces, while filamentous moulds were of the genera Aspergillus, Penicillium, Fusarium and Alternaria.
