A BMP7 Variant Inhibits Tumor Angiogenesis In Vitro and In Vivo through Direct Modulation of Endothelial Cell Biology

Bone morphogenetic proteins (BMPs), members of the TGF-β superfamily, have numerous biological activities including control of growth, differentiation, and vascular development. Using an in vitro co-culture endothelial cord formation assay, we investigated the role of a BMP7 variant (BMP7v) in VEGF, bFGF, and tumor-driven angiogenesis. BMP7v treatment led to disruption of neo-endothelial cord formation and regression of existing VEGF and bFGF cords in vitro. Using a series of tumor cell models capable of driving angiogenesis in vitro, BMP7v treatment completely blocked cord formation. The authors studied associations of nature- and infrastructure-based features with physical activity (PA) in different urban neighborhood types; 848 community-dwelling people aged 75–90 years reported PA and three perceived nature-based destinations and seven infrastructure-based features as outdoor mobility facilitators. Neighborhood type was defined using a geographic information system based on proximity to central service areas and residential density (city center, subcenter, and dense and dispersed areas outside centers). Injection of large doses of fructose, sorbitol, or a mixture of glucose, fructose and xylitol in rats causes a drop of liver ATP, total adenine nucleotides and Pi and a rise of AMP, which is in agreement with data from the literature. These changes are considered as a transient disturbance of homeostasis by compounds which are rapidly phosporylated in the liver. This is confirmed by the fact that during continuous infusion of these and other compounds at doses of 1,5 g · kg−1 · h−1 there was no such change. This article presents a stochastic dynamic model to study the demographic evolution per sexes and the corresponding well-being of a general human population. The main model variables are population per sexes and well-being. The considered well-being variable is the Gender-Related Development Index (GDI), a United Nations index.
