Conductance and Ion Selectivity of a Mesoscopic Protein Nanopore Probed with Cysteine Scanning Mutagenesis

2005

Nanometer-scale proteinaceous pores are the basis of ion and macromolecular transport in cells and organelles. Recent studies suggest that ion channels and synthetic nanopores may prove useful in biotechnological applications. To better understand the structure-function relationship of nanopores, we are studying the ion-conducting properties of channels formed by wild-type and genetically engineered versions of Staphylococcus aureus alpha-hemolysin (alphaHL) reconstituted into planar lipid bilayer membranes. Specifically, we measured the ion selectivities and current-voltage relationships of channels formed with 24 different alphaHL point cysteine mutants before and after derivatizing the c…

Secretion and antigenicity of hepatitis B virus small envelope proteins lacking cysteines in the major antigenic region.

1995

Abstract Disulfide bonds are of crucial importance for the structure and antigenic properties of the hepatitis B virus (HBV) envelope. We have evaluated the role of the eight highly conserved cysteines of the major antigenic region for assembly, secretion, and antigenicity of the envelope proteins. Mutants carrying single or multiple substitutions of alanine for cysteine were analyzed using epitope tagging and transient expression in COS-7 cells. The only single cysteines found to be indispensable for efficient secretion were Cys-107 and Cys-138, but double mutation of Cys-137 and Cys-139 also created a block to secretion. Poorly secreted mutants formed aberrant oligomeric structures. The a…

The Fumarate/Succinate Antiporter DcuB of Escherichia coli Is a Bifunctional Protein with Sites for Regulation of DcuS-dependent Gene Expression

2008

DcuB of Escherichia coli catalyzes C4-dicarboxylate/succinate antiport during growth by fumarate respiration. The expression of genes of fumarate respiration, including the genes for DcuB (dcuB) and fumarate reductase (frdABCD) is transcriptionally activated by C4-dicarboxylates via the DcuS-DcuR two-component system, comprising the sensor kinase DcuS, which contains a periplasmic sensing domain for C4-dicarboxylates. Deletion or inactivation of dcuB caused constitutive expression of DcuS-regulated genes in the absence of C4-dicarboxylates. The effect was specific for DcuB and not observed after inactivation of the homologous DcuA or the more distantly related DcuC transporter. Random and s…

Atherogenic properties of enzymatically degraded LDL: selective induction of MCP-1 and cytotoxic effects on human macrophages.

1998

Abstract —The mechanisms underlying the selective accumulation of macrophages in early atherosclerotic lesions are poorly understood but are likely to be related to specific properties of altered low density lipoprotein (LDL) deposited in the subendothelium. Enzymatic, nonoxidative degradation of LDL converts the lipoprotein to a potentially atherogenic moiety, enzymatically altered LDL (E-LDL), which activates complement and is rapidly taken up by human macrophages via a scavenger receptor–dependent pathway. Immunohistological evidence indicates that E-LDL is present in an extracellular location in the early lesion. We report that E-LDL causes massive release of monocyte chemotactic prote…

A Novel Energy Harvester for Powering Small UAVs: Performance Analysis, Model Validation and Flight Results

2019

The proposed work aims at exploring and developing new strategies to extend mission parameters (measured as travel distance and mission duration (MD)) of a new class of unmanned vehicles, named Micro Air Vehicles (MAVs). In this paper, a new analytical model, identifying all factors, which determine the MAV power consumption, is presented. Starting from the new model, the design of a nanoarray energy harvester, based on plasmonics nano-antenna technology is proposed. The preliminary study was based on a 22,066,058 22,066,058 &times

2012

Purpose To determinate biomechanical and functional changes in human keratoconic corneas after triangular section ring implantation in two designs. Methods Retrospective study of biomechanical properties of 46 patients (60 eyes) with keratoconus undergoing surgery Keraring® intracorneal ring segments divided into two groups according to 5 or 6 mm of diameter. Surgical technique to made the channel was with IntraLase femtosecond laser. UCVA and BSCVA were measured and biomechanical factors provided by the device ORA before surgery and postoperatively one month, 3 months and 6 months. Results After surgery, we obtained a significant increase (p 0.05). At baseline- 6 months, the improvement in…
