Determination of the lowest-energy oxidation site in nucleotides: 2'-deoxythymidine 5'-monophosphate anion.

High level ab initio computations anticipate nucleobases as the most favorable sites for oxidation in nucleotides. At the CASPT2 level, the lowest ionization channel for the 2'-deoxythymidine 5'-monophosphate anion is related to a pi-orbital of the thymine base. The present findings lead to revision of the recent assignments of the photodetachment photoelectron spectra of mononucleotide anions in the gas phase and support the classical view of the nucleobase being the main actor in the oxidation process of both nucleosides and nucleotides.

Synthesis of cyanoacetic acid by carbon dioxide and electrogenerated acetonitrile anion in undivided cells equipped with sacrificial anodes

The electrochemical synthesis of cyanoacetic acid was performed by cathodic reduction of tetraalkylammonium salts (R4NX) in acetonitrile in undivided cells equipped with sacrificial anodes with concomitant or subsequent addition of carbon dioxide. These syntheses avoid the use of dangerous reagents, yield cyanoacetic acid in moderate faradic yields and confirm that the cathodic reduction of tetraalkylammonium salts in the presence of acetonitrile leads to the formation of the anion CNCH2-.
Potential role of branched-chain amino acid catabolism in regulating fat oxidation.

Insulin-resistant or obese individuals have increased serum branched-chain amino acid (BCAA) levels. Recent findings relate increased BCAA catabolism to increased fatty acid oxidation and better metabolic health in physically active individuals. We hypothesize that, via glyceroneogenesis, BCAA catabolism mediates increased constitutive use of fatty acids for β-oxidation in subjects with increased inherent or acquired aerobic capacity both during exercise and at rest.

Racemic and chiral nonracemic α-substituted and α-unsubstituted β-fluoroalkyl β-amino acid derivatives 6 and 9 have been synthesized in two steps starting from fluorinated imidoyl chlorides 1 and ester enolates. This approach is based on the chemical reduction of previously obtained γ-fluorinated β-enamino esters 4 by using ZnI2/NaBH4 in a nonchelated aprotic medium (dry CH2Cl2) as the reducing agent. A metal-chelated six-membered model has been suggested to explain the stereochemical outcome of the reduction reaction. Abstract Some 1,2-disubstituted cyclopentanes were synthesized. These were methyl 9-(2-butyl-cyclopentyl)-8-nonenoate, methyl 7-(2-hexyl-cyclopentyl)-6-heptenoate and methyl 5-(2-octyl-cyclopentyl)-4-pentenoate and their corresponding alkanoates. The synthesis involved a Michael addition of alkylmagnesium bromide to an unsaturated cyclic ester. The resulting cis and trans ethyl 2-alkylcyclopentane carboxylate were further converted to the alcohols then to the aldehydes. High-level electronic structure calculations were carried out for the study of the reaction pathways in the OH-initiated oxidations of methyl vinyl ketone (MVK) and methacrolein (MACR). For the two conformers of MVK (called synperiplanar and antiperiplanar), the addition channels of OH to the terminal and central carbon atom of the double bond dominate the overall rate constant, whereas the abstraction of the methyl hydrogen atoms has no significant kinetic role. In the case of MACR, only the antiperiplanar conformer is important in its reactivity. The impact of Pleurotus ostreatus on organic matter transformation processes.

This study showed the effect of Pleurotus ostreatus on the external organic matter (OM) transformation processes. The kinetics of these changes were determined. The experiment was conducted as 42-day pot experiment done in four combinations (with four replications). The four combinations of different substrates were: (1) organic, (2) composted hay, (3) organic soil + composted hay, (4) organic soil + composted hay + lead. The study results showed that P. ostreatus had the ability to transform external OM as well as metal–organic complexes. Determination of steady-state levels of oxidative DNA base modifications in mammalian cells by means of repair endonucleases

The alkaline elution technique in combination with various repair endonucleases (Fpg protein, endonuclease III, exonuclease III, T4 endonuclease V) was used to quantify steady-state (background) levels of oxidative base modifications in various types of mammalian cells. In human lymphocytes the number of base modifications sensitive to Fpg protein, which include 8-hydroxyguanine, was 0.25 +/- 0.05 per 10(6) base pairs. Even lower levels (0.07 +/- 0.02 per 10(6) bp) were observed in HeLa cells. The numbers of sites sensitive to the other repair endonucleases were below the detection limit (0.05 per 10(6) bp).
