Simultaneous gas chromatographic determination of aliphatic C2−C20 n-carboxylic acids and their methyl esters using SE-30 and OV-351 quartz capillary…

1983

Simultaneous determination of aliphatic C2−C20 carboxylic acids and their methyl esters was carried out using SE-30 and OV-351 quartz capillary columns with temperature programming. Complete resolution of all 36 components was achieved. On SE-30 the methyl ester and the corresponding free acid are eluted in turn, whereas on OV-351 acetic acid eluted after methyl octanoate, after which the Cn methyl ester and Cn-6 carboxylic acid (n>9) are eluted one after the other. A non-polar SE-30 column is better for long-chain acids giving sharp peaks without tailing, the analysis time of the mixture being less than 16 minutes. The weight response correction factors for compounds under optimum operatin…

Separation of Proteic Primary Amino Acids under Several Reversed‐Phase Liquid Chromatographic Conditions

2006

Abstract The reversed‐phase liquid chromatographic (RPLC) analysis of proteic primary amino acids with acetonitrile‐water, using pre‐column derivatisation with o‐phthalaldehyde (OPA) and N‐acetylcysteine (NAC), was compared with RPLC modes using trifluoroacetic acid or pentadecafluorooctanoic acid and evaporative light‐scattering detection, or sodium dodecyl sulphate micelles with pre‐ and post‐column derivatisation. The importance of column lifetime, risk of potential damages in the instrumentation, retention and resolution, was considered. Among the assayed approaches, the best is still aqueous‐organic RPLC with pre‐column derivatisation. It not only yields the most reliable results, but …

Prediction of the genetic variety of Spanish extra virgin olive oils using fatty acid and phenolic compound profiles established by direct infusion m…

2007

Abstract The genetic varieties of Spanish extra virgin olive oils (Arbequina, Hojiblanca and Picual) were predicted by direct infusion of the samples in the electrospray ionization source of a mass spectrometer, followed by linear discriminant analysis of the spectral data. The samples were 1:50 diluted (v/v) with an 85:15 propanol/methanol (v/v) mixture containing 40 mM KOH and infused. The abundances of the [M–H] − peaks of the free fatty acids (7 peaks) and 28 phenolic compounds (20 peaks) were measured. Ratios of pairs of peak abundances were used as predictors in the construction of the linear discriminant analysis models. An excellent resolution between the three genetic varieties was…

Determination of cationic surfactants by capillary zone electrophoresis and micellar electrokinetic chromatography with deoxycholate micelles in the …

2000

Mixtures of the cationic surfactants benzalkonium chloride (BKC) and cetylpyridinium chloride (CPC) were quickly resolved and reproducibly and reliably determined by using background electrolytes (BGEs) containing 80 mM borate, pH 8.5, bile salts and large concentrations of an organic solvent. When the bile salt is present, the separation mechanism changes from capillary zone electrophoresis (CZE) to a mixed micellar electrokinetic chromatography (MEKC)-CZE, with predominant MEKC interactions, which lead to an excellent resolution of all the solutes, including the C12-C18 homologues of BKC and CPC. A BGE containing 50 mM sodium deoxycholate and 30% ethanol for an extreme resolution, or 20% …

Human neuroglobin: crystals and preliminary X-ray diffraction analysis

2002

Neuroglobin, a recently discovered member of the haemoglobin superfamily, is primarily expressed in the brain of humans and other vertebrates, where it has been proposed to enhance O(2) supply in response to hypoxia or ischaemia, protecting the neuron from hypoxic injury. Neuroglobin is the first example of a vertebrate haemoglobin in which a hexacoordinate haem geometry has been detected. A triple mutant (replacing three Cys residues) of human neuroglobin (151 amino acids) has been expressed in Escherichia coli, purified and crystallized in two crystal forms, the best of which diffracts to 1.95 A resolution using synchrotron radiation. The crystals belong to space group P2(1), with unit-ce…

The sanidic polyamide [NM-Ph(CO 2 R) 3 (CO 2 Me)-NMCO-Ph(OR) 2 CO] n (R=C 12 H 25 ) is studied. It is shown that this polymer has a biaxial structure with quasi 2-D long range translational correlations. The correlations in one direction are limited by the length of the molecule. The oriented regions are separated by regions in which the molecules are less oriented

Small-Angle Scattering Techniques for Analyzing Conformational Transitions in Hemocyanins

2004

Publisher Summary The precise delivery of oxygen from respiratory surfaces to the tissues is mediated by cooperative and allosterically regulated carrier proteins, such as hemoglobin or hemocyanin. To establish cooperativity, these proteins must be able to adopt different conformations. These conformations are characterized by different ligand affinities, which have their basis in different structures as is the case for the deoxy and oxy states of human hemoglobin. To understand the cooperative interaction of these molecules at the molecular level, the structures of these conformations must be resolved and the transitions between them must be monitored. Because of the nature of sample prepa…

Peptide–Membrane Interactions Monitored by Fluorescence Lifetime Imaging: A Study Case of Transportan 10

2021

The interest on detailed analysis of peptide-membrane interactions is of great interest in both fundamental and applied sciences as these may relate to both functional and pathogenic events. Such interactions are highly dynamic and spatially heterogeneous, making the investigation of the associated phenomena highly complex. The specific properties of membranes and peptide structural details, together with environmental conditions, may determine different events at the membrane interface, which will drive the fate of the peptide-membrane system. Here, we use an experimental approach based on the combination of spectroscopy and fluorescence microscopy methods to characterize the interactions …

Multifunctional polymer-derivatized γ-Fe2O3 nanocrystals as a methodology for the biomagnetic separation of recombinant His-tagged proteins

2008

Abstract Multifunctional polymer-derivatized superparamagnetic iron oxide (γ-Fe2O3) nanoparticles were prepared for biomagnetic separation of histidine-tagged recombinant proteins building up a faster and efficient method for protein separation by making use of their intrinsic magnetic properties. Using polymer bound γ-Fe2O3 nanocrystals, a 6× histidine-tagged recombinant protein (silicatein) with a molecular weight of 24 kDa has been isolated and purified. The supermagnetic iron oxide nanocrystals were characterized by transmission electron microscopy (TEM), high-resolution TEM (HRTEM), SQUID and Mossbauer and the polymer functionalization of the γ-Fe2O3 nanocrystals was monitored by UV–vi…

Size Exclusion Chromatography of Proteins on Improved Bonded Silica Columns

1985

Publisher Summary This chapter discusses the size exclusion chromatography (SEC) of proteins on improved bonded silica columns. Over the past few years, the use of high performance liquid chromatography (HPLC) has been extended to protein chemistry as a powerful separation and isolation technique. A spectrum of variants providing high resolution potential—ion exchange, reversed phase, hydrophobic interaction, affinity, and size exclusion—is available. Of these, SEC is the simplest with respect to method development and equipment design: proteins are separated according to their size in a short and predictable time span under isocratic conditions. The SEC columns are packed with micro-partic…
