Basement Membrane Mimics of Biofunctionalized Nanofibers for a Bipolar-Cultured Human Primary Alveolar-Capillary Barrier Model

2017

In vitro reconstruction of an alveolar barrier for modeling normal lung functions and pathological events serve as reproducible, high-throughput pharmaceutical platforms for drug discovery, diagnosis, and regenerative medicine. Despite much effort, the reconstruction of organ-level alveolar barrier functions has failed due to the lack of structural similarity to the natural basement membrane, functionalization with specific ligands for alveolar cell function, the use of primary cells and biodegradability. Here we report a bipolar cultured alveolar-capillary barrier model of human primary cells supported by a basement membrane mimics of fully synthetic bifunctional nanofibers. One-step elect…

Wet Chemistry and Peptide Immobilization on Polytetrafluoroethylene for Improved Cell-adhesion

2016

Endowing materials surface with cell-adhesive properties is a common strategy in biomaterial research and tissue engineering. This is particularly interesting for already approved polymers that have a long standing use in medicine because these materials are well characterized and legal issues associated with the introduction of newly synthesized polymers may be avoided. Polytetrafluoroethylene (PTFE) is one of the most frequently employed materials for the manufacturing of vascular grafts but the polymer lacks cell adhesion promoting features. Endothelialization, i.e., complete coverage of the grafts inner surface with a confluent layer of endothelial cells is regarded key to optimal perfo…

Spray dried hyaluronic acid microparticles for adhesion controlled aggregation and potential stimulation of stem cells

2017

Spray-dried microparticles of a derivative of hyaluronic acid (HA) have been engineered to obtain a controlled aggregation with Human Mesenchymal Stem Cells (hMSCs) into 3D constructs. We demonstrated the utility of chemical functionalization of a native constituent of the extracellular matrix to improve processing performances and to control on stem cell adhesion and differentiation. Native hyaluronic acid (HA), cell adhesive peptides (RGD), transforming growth factor β3, dexamethasone are biological agents potentially suitable for chondrogenic stimulation of hMSCS. However unmodified HA suffers of drawbacks in terms of stability and versatility of processing. Functionalization strategies…

Enhanced adhesion and in situ photothermal ablation of cancer cells in surface-functionalized electrospun microfiber scaffold with graphene oxide

2017

The physicochemical characteristics of a biomaterial surface highly affect the interaction with living cells. Recently, much attention has been focused on the adhesion properties of functional biomaterials toward cancer cells, since is expected to control metastatic spread of a tumor, which is related to good probability containing the progression of disease burden. Here, we designed an implantable poly(caprolactone)-based electrospun microfiber scaffold, henceforth PCLMF-GO, to simultaneously capture and kill cancer cells by tuning physicochemical features of the hybrid surface through nitrogen plasma activation and hetero-phase graphene oxide (GO) covalent functionalization. The surface i…

Rational Design of Thermoresponsive Microgel Templates with Polydopamine Surface Coating for Microtissue Applications.

2021

Functional microgels provide a versatile basis for synthetic in vitro platforms as alternatives to animal experiments. The tuning of the physical, chemical, and biological properties of synthetic microgels can be achieved by blending suitable polymers and formulating them such to reflect the heterogenous and complex nature of biological tissues. Based on this premise, this paper introduces the development of volume-switchable core-shell microgels as 3D templates to enable cell growth for microtissue applications, using a systematic approach to tune the microgel properties based on a deep conceptual and practical understanding. Microscopic microgel design, such as the tailoring of the microg…

Label-Free Pyrophosphate Recognition with Functionalized Asymmetric Nanopores

2016

[EN] The label¿free detection of pyrophosphate (PPi) anions with a nanofluidic sensing device based on asymmetric nanopores is demonstrated. The pore surface is functionalized with zinc complexes based on two di(2¿picolyl)amine [bis(DPA)] moieties using carbodiimide coupling chemistry. The complexation of zinc (Zn2+) ion is achieved by exposing the modified pore to a solution of zinc chloride to form bis(Zn2+¿DPA) complexes. The chemical functionalization is demonstrated by recording the changes in the observed current¿voltage (I¿V) curves before and after pore modification. The bis(Zn2+¿DPA) complexes on the pore walls serve as recognition sites for pyrophosphate anion. The experimental re…

Organocatalytic enantioselective functionalization of indoles in the carbocyclic ring with cyclic imines

2019

[EN] An organocatalytic enantioselective functionalization in the carbocyclic ring of indoles with benzoxathiazine 2,2-dioxides is described using a quinine-derived bifunctional organocatalyst. This aza-Friedel-Crafts reaction provides 4-indolyl, 5-indolyl and 7-indolyl sulfamidate derivatives in good yields (up to 99%) and with moderate to high enantioselectivities (up to 86% ee).

Synthesis and characterisation of pack cemented aluminide coatings on metals

2004

Abstract The exposition of metallic materials to high temperature environments leads to their corrosion because of oxidation or sulphidation. One way to protect such materials is to produce an Al 2 O 3 layer which needs to be continuous enough to limit diffusion of oxygen or metallic elements, and withstand this corrosion. Since a few years, it has been proved that aluminide compounds are one of the most effective materials to achieve this goal. Indeed, they possess sufficient Al and many beneficial mechanical properties when exposed to high temperature conditions to make possible the formation of a protective Al 2 O 3 scale. This study is aimed at the elaboration of iron, nickel and molybd…

Affinity Sensors for the Diagnosis of COVID-19

2021

The coronavirus disease 2019 (COVID-19) outbreak caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was proclaimed a global pandemic in March 2020. Reducing the dissemination rate, in particular by tracking the infected people and their contacts, is the main instrument against infection spreading. Therefore, the creation and implementation of fast, reliable and responsive methods suitable for the diagnosis of COVID-19 are required. These needs can be fulfilled using affinity sensors, which differ in applied detection methods and markers that are generating analytical signals. Recently, nucleic acid hybridization, antigen-antibody interaction, and change of reactive oxyge…

Alkaline treatment as a means to boost the activity of TiO(2)in selective photocatalytic processes

2020

In this work, the activity enhancement of TiO(2)photocatalysts by alkaline treatment has been investigated. Commercial (BDH, U.K., anatase phase) TiO(2)samples, treated in alkaline solutions of NaOH at different concentrations and temperatures, were tested for the photocatalytic partial oxidation of 3-pyridinemethanol in water under UVA irradiation to form vitamin B-3. Photocatalyst characterization has been carried out by XRD, BET, TEM, TGA, photoluminescence, FTIR, Raman, DRS, EPR and photocurrent measurements. The alkaline-treated samples showed an increased activity of up to 7 times (by considering first order rate constant values) compared to untreated BDH and the superior performance …
