Mapping antigenic epitopes of potato virus Y with antibodies affinity-purified by using overlapping synthetic peptides

Synthetic, overlapping peptides representing the entire amino acid sequence of potato virus Y (PVY) coat protein were used to affinity-purify antibodies from polyclonal antisera to PVY. In testing the binding of the purified antibodies to PVY particles, antigenic epitopes were identified. The N-terminal and C-terminal regions of the PVY coat protein were found to contain most of the antigenic epitopes. The results will facilitate the development of detection methods for PVY based on synthetic peptides.

Fight against Citrus tristeza virus (CTV) implies the use of CTV-resistant rootstocks, such as citranges [C. Sinensis (L.) Osb. × P. Trifoliata (L.) Raf.]. The 'C35' represents a promising tolerant to CTV genotype. The employment of innovative in vitro techniques, such as encapsulation, could open new perspectives for this genotype propagation, particularly in the nursery activity. Es wird uber den Einflus von Cytosin-Arabinosid, Hydroxy-Urea und von Mitomycin C auf die Entstehung membranstandiger oder extrahierbarer Antigene, auf die Riesenzellbildung sowie auf die Herpesvirus-bedingte Hemmung der Zell-RNS-Synthese berichtet. Durch die Verabreichung der drei Substanzen wird die Reduplikation der DNS des eingedrungenen Virus vollig blockiert. Trotzdem lassen sich die Phanomene der Riesenzellbildung, der Virus-Antigen-Synthese und der Synthesehemmung der Zell-RNS in vollem Umfange nachweisen.

The icosahedral double-stranded DNA bacteriophages containing an internal membrane belong to two families, the Tectiviridae and the Corticoviridae. Members of the Tectiviridae have a linear dsDNA genome (e.g., PRD1) and those of the Corticoviridae have a circular genome (the only example being PM2). The DNA is covered by a membrane which is in turn covered by a protein capsid. The infection cycle begins when spikes on the capsid recognize the host cell receptor and the virus starts the delivery of the genome into the cell cytoplasm. Virazole (Ribavirin) a Cytostatic Agent

Virazole (1-β-D-ribofuranosyl-1,2,4-triazole-3carboxamide,ribavirin) is a synthetic triazol nucleoside with a broad spectrum of antiviral activity. But virazole affects the metabolism not only of virus infected cells. Virazole strongly inhibits the cell proliferation of mouse lymphoma cells (L 5178y), which were not infected with DNA- or RNA-viruses. Starting with 3 × 103 cells/ml and an incubation period of 72 hr, the drug reduces the cell proliferation to 50% (= ED50 concentration) in a concentration of 4,7 μM.
Structure and dynamics of RNA guanine quadruplexes in SARS-CoV-2 genome. Original strategies against emerging viruses

Guanine quadruplexes (G4) structures in viral genome have a key role in modulating viruses’ biological activity. While several DNA G4 structures have been experimentally resolved, RNA G4s are definitely less explored. We report the first calculated G4 structure of the RG-1 RNA sequence of SARS-CoV-2 genome, obtained by using a multiscale approach combining quantum and classical molecular modelling and corroborated by the excellent agreement between the corresponding calculated and experimental circular dichroism spectra. We prove the stability of RG-1 G4 arrangement as well as its interaction with G4 ligands potentially inhibiting viral protein translation.

Abstract:The aim of this investigation is to analyze sexual risk behaviors and attitudes for HIV/AIDS infection of MSM. The ENSI was administrated to 202 men, 168 homosexuals and 24 bisexuals, from 14 to 57 years. The results show a high confidence in condoms as a method of HIV prevention (75.7% very effective and 28.1% effective); however the systematic use of a condom is moderate in anal intercourse (54.64%) and in vaginal intercourse (49.05%) and very low in oral sex (6.22%). There is a low perceived probability (20.28%) and moderate fear (41.25%) become infected with HIV. 20.3% report having drunk alcohol in the last relationship and 5.4% say other drugs.
