Evaluation of Two Method to Measure Xylanase Activity in Relation with Rheological Effects of Xylanases on White Flour

Many methods are available to measure the xylanase activity of enzymatic preparation. Most of method uses different substrates from those that will be hydrolyzed in various applications in food industry. This fact will lead to a deformed image about how evaluated xylanases will really act in a specific application. The aim of this study is to evaluate the activity of seven xylanase preparation with two method, a colorimetric method with birch insoluble xylan as substrate and a viscometric method with wheat soluble arabinoxylan as substrate.

pH control of the production of recombinant glucose oxidase in Aspergillus nidulans.

Aims:  Recombinant Aspergillus nidulans sVAL040, capable of synthesizing and secreting glucose oxidase derived from Aspergillus niger was used to study the influence of pH and carbon source on enzyme production. Methods and Results:  Glucose oxidase gene (goxC) was expressed under transcriptional regulation by using the promoter of A. nidulans xlnB gene (encoding an acidic xylanase). A maximum specific glucose oxidase activity of approx. 10 U mg−1 protein and a maximum volumetric productivity of 29·9 U l−1 h−1 were obtained at pH 5·5, after 80 h of growth by using xylose as inducer. Xylan-degrading enzymes are crucial for the deconstruction of hemicellulosic biomass, making the hydrolysis products available for various industrial applications such as the production of biofuel. To determine the substrate specificities of these enzymes, we prepared a collection of complex xylan oligosaccharides by automated glycan assembly. Rheological Evaluation of Some Commercial Xylanases

The arabinoxylans play and the xylanases play an important role breadmaking. For a better selection of xylanases, it is necessary to understand how they work in breadmaking. At the same activity, the xylanases have different effect on dough rheology. The xylanases activity measured through birch xylan hydrolysis it is not correlated with extensographic effects. Weak but better correlations were observed when the xylanase activity was measured as the ability to decrease the viscosity of soluble wheat arabinoxylans solution.

Toxicological Assessment of Recombinant Xylanase X22 in Wine

Toxicological evaluation of xylanase X(22) from Aspergillus nidulans expressed in a wine yeast strain was carried out. The safety of the X(22) intake was assessed by digestibility, bioinformatic, and mouse short-term repeated dosing studies, although X(22) shows resistance to proteolytic degradation in the gastrointestinal system, is a minority protein component (<0.5 10(-)(6) %) of the produced wine, and shows no significant amino acid sequence homology to any known food allergens. We characterized the changes in cell-wall hemicellulosic polysaccharides and the hemicellulose-degrading enzymes associated with apple and tomato fruits infected by Penicillium expansum. Our results showed a reduction in the molecular mass of hemicelluloses, with this reduction being particularly notable in the xyloglucan associated with P. expansum infection. The activation of fungal beta-glucanases was also highlighted. Fruit xyloglucan endotransglucosylase/hydrolase (XTH)-specific activity decreased drastically during the infection process in both apple and tomato fruits. Xyloglucan-based hydrogel films for wound dressing: Structure-property relationships

Thin xyloglucan-based hydrogel films have been synthetized and characterized in the prospect of producing wound dressings. Polyvinyl alcohol (PVA) and glycerol (Gro) were added to have an optimal combination of softness, conformability and resilience. Temporal control of xyloglucan self-assembly into layered structures by radiation-induced degradation

Partially degalactosylated xyloglucan from tamarind seeds (Deg-XG) is a very appealing biopolymer for the production of in situ gelling systems at physiological temperature. In this work, we observe that the morphology of hydrogels evolves towards high degrees of structural organization with time, yielding to dense stacks of thin membranes within 24 h of incubation at 37 °C. Modified arabinoxylan-based films. Part B. Grafting of omega-3 fatty acids by oxygen plasma and electron beam irradiation.

Arabinoxylans (AXs) are byproducts of the cereal milling industry. To obtain high-value products, AXs have been used as a film-forming agent. Hence, AX-based films are poor water vapor barriers. The objectives of this study were to graft omega-3 (omega3) fatty acids onto AX polymeric chains by using two new technologies: cold plasma and electron beam (EB) irradiation. Results show that the surface hydrophobicity of the modified films is higher than that of a waxy coating or a low-density polyethylene (LDPE) film.
